The Study of pH Effects on Phase Transition of Multi-Stimuli Responsive P(NiPAAm-co-AAc) Hydrogel Using 2D-COS.

ABSTRACT: The temperature and mechanism of phase transition of poly(N-isopropylacrylamide-co-acrylic acid) [P(NiPAAm-co-AAc)], which is one of the multi-stimuli responsive polymers, were investigated at various pHs using infrared (IR) spectroscopy, two-dimensional (2D) gradient mapping, and two-dimensional correlation spectroscopy (2D-COS). The determined phase transition temperature of P(NiPAAm-co-AAc) at pH 4, 3, and 2 based on 2D gradient mapping and principal component analysis (PCA) showed that it decreases with decreasing pH, because COOH group in AAc changes with variation of pH. The results of 2D-COS analysis indicated that the phase transition mechanism of P(NiPAAm-co-AAc) hydrogel at pH4 is different from that at pH2 due to the effect of COOH group of AAc.

Project description:This paper presents a green method for fabricating dual temperature- and pH-responsive electrospun fibrous mats from an aqueous-based blend poly(N-isopropylacrylamide-co-acrylic acid) (P(NIPAAm-co-AAc)) and regenerated silk fibroin (RSF) by employing electrospinning technique. P(NIPAAm-co-AAc) was synthesized by free radical solution polymerization and its low critical solution temperature (LCST) was in the physiological range (38.8 °C). The P(NIPAAm-co-AAc)/RSF fibers were prepared by electrospinning technology in the presence of the crosslinking agents (EDC·HCl and NHS) with water as solvent. After in situ crosslinking and water-annealing process, the water-stable composite fibrous mats were obtained. Scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) were used to analyze the crosslinking process. Temperature and pH dual stimuli-responsive swelling-shrinking behavior of the fibrous mats were observed when the temperature was below and above the LCST of the copolymer at different pHs. In addition, rhodamine B-loaded the fibrous mats also showed dual temperature and pH controlled release behavior, demonstrating the potential use of the fibrous mats for "smart" controlled drug delivery applications.

Dual Stimuli-Responsive P(NIPAAm-co-SPA) Copolymers: Synthesis and Response in Solution and in Films.

Project description:We present the synthesis and solution properties of dual stimuli-responsive poly(N-isopropylacrylamide-co-spiropyran acrylate) (P(NIPAAm-co-SPA)) copolymers of varying composition prepared via nitroxide-mediated copolymerization. The resulting copolymers feature molar masses from 40,000 to 100,000 g/mol according to static light scattering and an SPA content of up to 5.3%. The latter was determined by ¹H NMR spectroscopy and UV⁻Vis spectroscopy. These materials exhibit reversible response upon irradiation in polymeric films for a minimum of three cycles; their response in solution to both light and temperature was also investigated in an aqueous TRIS buffer (pH 8). Irradiation was carried out using LED setups with wavelengths of 365 and 590 nm. In aqueous solution, a custom-made setup using a fiber-coupled 200 W Hg(Xe) lamp with 340 and 540 nm filters was used and additional heating of the copolymer solutions during irradiation allowed to study influence of the presence of either the spiropyran or merocyanine form on the cloud point temperature. Hereby, it was found that increasing the SPA content leads to a more pronounced difference between both states and decreasing cloud points in general.

Porous silver-coated pNIPAM-co-AAc hydrogel nanocapsules.

Project description:This paper describes the preparation and characterization of a new type of core-shell nanoparticle in which the structure consists of a hydrogel core encapsulated within a porous silver shell. The thermo-responsive hydrogel cores were prepared by surfactant-free emulsion polymerization of a selected mixture of N-isopropylacrylamide (NIPAM) and acrylic acid (AAc). The hydrogel cores were then encased within either a porous or complete silver shell for which the localized surface plasmon resonance (LSPR) extends from visible to near-infrared (NIR) wavelengths (i.e., ?max varies from 550 to 1050 nm, depending on the porosity), allowing for reversible contraction and swelling of the hydrogel via photothermal heating of the surrounding silver shell. Given that NIR light can pass through tissue, and the silver shell is porous, this system can serve as a platform for the smart delivery of payloads stored within the hydrogel core. The morphology and composition of the composite nanoparticles were characterized by SEM, TEM, and FTIR, respectively. UV-vis spectroscopy was used to characterize the optical properties.

Synthesis and Characterization of Thermosensitive P(NIPAAm-co-AAc)-Grafted Silica Nanocomposites for Smart Architectural Coatings.

Project description:In this study, a new class of thermosensitive poly(N-isopropylacrylamide)-co-poly(acrylic acid) (P(NIPAAm-co-AAc))-grafted modified silica (m-silica) nanocomposites was prepared using a sol-gel technique. The addition of silica to P(NIPAAm-co-AAc) copolymer hydrogel has the potential to open up new applications in the development of thermosensitive building materials by leveraging the favorable thermal characteristics of P(NIPAAm-co-AAc). The silica was prepared using 3-aminopropyltriethoxysilane and 4,4'-azobis(4-cyanovaleric acid) to form the m-silica powder, which increased the adhesion between the organic and inorganic hybrid materials. The P(NIPAAm-co-AAc) copolymer hydrogel was mixed with the m-silica to form the P(NIPAAm-co-AAc)-grafted m-silica nanocomposites. Scanning electron microscopy, X-ray diffraction analysis, thermogravimetric analysis, Fourier-transform infrared spectroscopy, and thermosensitive measurement were conducted to evaluate the structure and water-holding capacity of the nanocomposites. The results indicated that the P(NIPAAm-co-AAc)-grafted m-silica nanocomposites could retain water for more than 300 min at temperatures higher than the lower critical solution temperature. The P(NIPAAm-co-AAc)-grafted m-silica nanocomposites exhibited favorable thermosensitive properties and may therefore be applied in smart architectural coatings.

A pH-sensitive, stimuli-responsive, superabsorbent, smart hydrogel from psyllium (Plantago ovata) for intelligent drug delivery.

Project description:Herein, we report a polysaccharide-based hydrogel isolated from psyllium husk (a well-known dietary fiber) and evaluated for its swelling properties in deionized water (DW) at different physiological pH values, i.e., 1.2, 6.8 and 7.4. Swelling of psyllium hydrogel (PSH) in DW under the influence of temperature and at different concentrations of NaCl and KCl solutions was also examined. A pH-dependent swelling pattern of PSH was observed following the order DW > pH 7.4 > pH 6.8 > pH 1.2. Stimuli-responsive swelling and deswelling (on-off switching) behavior of PSH was observed in DW and ethanol, DW and normal saline, at pH 7.4 and pH 1.2 environments, respectively. Similar swelling behavior and on-off switching attribute of PSH-containing tablets indicated the unaltered nature of PSH even after compression. Scanning electron micrographs of swollen and then freeze-dried PSH via transverse and longitudinal cross-sections revealed hollow channels with an average pore size of 6 ± 2 μm. Furthermore, PSH concentration-dependent sustained release of theophylline from tablet formulation was witnessed for >15 h following the non-Fickian diffusion mechanism. Subacute toxicity studies revealed the non-toxic nature of PSH. Therefore, dietary fiber-based material, i.e., PSH could be a valuable pharmaceutical excipient for intelligent and targeted drug delivery.

Antibacterial and Cytocompatible pH-Responsive Peptide Hydrogel.

Project description:A short peptide, FHHF-11, was designed to change stiffness as a function of pH due to changing degree of protonation of histidines. As pH changes in the physiologically relevant range, G' was measured at 0 Pa (pH 6) and 50,000 Pa (pH 8). This peptide-based hydrogel is antimicrobial and cytocompatible with skin cells (fibroblasts). It was demonstrated that the incorporation of unnatural AzAla tryptophan analog residue improves the antimicrobial properties of the hydrogel. The material developed can have a practical application and be a paradigm shift in the approach to wound treatment, and it will improve healing outcomes for millions of patients each year.

Design of stimuli-responsive transition metal dichalcogenides.

Project description:Stimuli-responsive systems are an emerging class of materials in fields as diverse as electronics, optoelectronics, cancer detection, drug delivery, or sensing. Especially focusing on nanomaterials, 2D transition metal dichalcogenides have recently attracted the scientific community's attention due to their remarkable intrinsic stimuli-responsive behaviour upon external stimuli such as pH, light, voltage, or certain pathogens. This significant response can be further enhanced by forming mixed-dimensional heterostructures and by molecular functionalization, capitalizing on chemistry to manipulate and boost their intrinsic stimuli-responsive properties. Furthermore, thanks to the endless possibilities of chemistry, a new class of smart materials based on the combination of stimuli-responsive molecular systems with transition metal dichalcogenides has recently been synthesized. In these materials, the physical properties of the 2D layers are reversibly modified by the switchable molecules, not only enhancing their stimuli-responsive behaviour but also providing memory to the hybrid. Therefore, this review explores the recent breakthroughs in the chemical design of smart transition metal dichalcogenides with built-in responsiveness.

Multicolored pH-tunable and activatable fluorescence nanoplatform responsive to physiologic pH stimuli.

Project description:Tunable, ultra-pH responsive fluorescent nanoparticles with multichromatic emissions are highly valuable in a variety of biological studies, such as endocytic trafficking, endosome/lysosome maturation, and pH regulation in subcellular organelles. Small differences (e.g., <1 pH unit) and yet finely regulated physiological pH inside different endocytic compartments present a huge challenge to the design of such a system. Herein, we report a general strategy to produce pH-tunable, highly activatable multicolored fluorescent nanoparticles using commonly available pH-insensitive dyes with emission wavelengths from green to near IR range. The primary driving force of fluorescence activation between the ON (unimer) and OFF (micelle) states is the pH-induced micellization. Among three possible photochemical mechanisms, homo Förster resonance energy transfer (homoFRET)-enhanced decay was found to be the most facile strategy to render ultra-pH response over the H-dimer and photoinduced electron transfer (PeT) mechanisms. Based on this insight, we selected several fluorophores with small Stoke shifts (<40 nm) and established a panel of multicolored nanoparticles with wide emission range (500-820 nm) and different pH transitions. Each nanoparticle maintained the sharp pH response (ON/OFF < 0.25 pH unit) with corresponding pH transition point at pH 5.2, 6.4, 6.9, and 7.2. Incubation of a mixture of multicolored nanoparticles with human H2009 lung cancer cells demonstrated sequential activation of the nanoparticles inside endocytic compartments directly correlating with their pH transitions. This multicolored, pH-tunable nanoplatform offers exciting opportunities for the study of many important cell physiological processes, such as pH regulation and endocytic trafficking of subcellular organelles.

A Smart pH-Responsive Three Components Luminescent Hydrogel.

Project description:In this study, we report a novel three-component luminescent hydrogel, which is composed of amino acid derivatives (N,N'-di valine-3,4,9,10-perylenetetracarboxylic acid, NVPD), riboflavin (RF), and melamine (MM). The three-component hydrogel is attributed to multiple hydrogen bonds and the strong π-π stacking interaction between these molecules. Based on the strong hydrogen bonding of the gelator, when the reversible process between the gel and the solution take places it changes the pH of the system from 6.1 to 10.6. In addition, green fluorescence could be the emissive of the hydrogel under 498 nm and the conversion process of the aggregation state repeated reversibly by altering the value of ambient pH. This pH-responsive luminescent gel may display potential for use in nano pH sensors.

Stimuli-responsive hydrogel based on natural polymers for breast cancer.

Project description:Aims: Breast cancer is the most common malignancy among women in both high- and low-resource settings. Conventional breast cancer therapies were inefficient and had low patient compliance. Stimuli-responsive hydrogels possessing similar physicochemical features as soft tissue facilitate diagnostic and therapeutic approaches for breast cancer subtypes. Scope: Polysaccharides and polypeptides are major natural polymers with unique biocompatibility, biodegradability, and feasible modification approaches utilized frequently for hydrogel fabrication. Alternating the natural polymer-based hydrogel properties in response to external stimuli such as pH, temperature, light, ultrasonic, enzyme, glucose, magnetic, redox, and electric have provided great potential for the evolution of novel drug delivery systems (DDSs) and various advanced technologies in medical applications. Stimuli-responsive hydrogels are triggered by specific cancer tissue features, promote target delivery techniques, and modify release therapeutic agents at localized sites. This narrative review presented innovation in preparing and characterizing the most common stimuli-responsive natural polymer-based hydrogels for diagnostic and therapeutic applications in the breast cancer area. Conclusion: Stimuli-responsive hydrogels display bioinspiration products as DDSs for breast cancer subtypes, protect the shape of breast tissue, provide modified drug release, enhance therapeutic efficacy, and minimize chemotherapy agents' side effects. The potential benefits of smart natural polymer-based hydrogels make them an exciting area of practice for breast cancer diagnosis and treatment.
