A practical method for regiocontrolled one-carbon ring contraction.

ABSTRACT: A practical and efficient method for the perfluorobutanesulfonyl azide-mediated one-carbon ring contraction of cyclic enoxysilanes is described. High-yielding procedures for the elaboration of the resulting N-acyl sulfonamide products are reported.

Project description:The 6,6-quinolone scaffolds on which viridicatin-type fungal alkaloids are built are frequently found in metabolites that display useful biological activities. Here we report in vitro and computational analyses leading to the discovery of a hemocyanin-like protein AsqI from the Aspergillus nidulans aspoquinolone biosynthetic pathway that forms viridicatins via a conversion of the cyclopenin-type 6,7-bicyclic system into the viridicatin-type 6,6-bicyclic core through elimination of carbon dioxide and methylamine through methyl isocyanate.

Photomediated ring contraction of saturated heterocycles.

Project description:Saturated heterocycles are found in numerous therapeutics and bioactive natural products and are abundant in many medicinal and agrochemical compound libraries. To access new chemical space and function, many methods for functionalization on the periphery of these structures have been developed. Comparatively fewer methods are known for restructuring their core framework. Herein, we describe a visible light-mediated ring contraction of α-acylated saturated heterocycles. This unconventional transformation is orthogonal to traditional ring contractions, challenging the paradigm for diversification of heterocycles including piperidine, morpholine, thiane, tetrahydropyran, and tetrahydroisoquinoline derivatives. The success of this Norrish type II variant rests on reactivity differences between photoreactive ketone groups in specific chemical environments. This strategy was applied to late-stage remodeling of pharmaceutical derivatives, peptides, and sugars.

A photo-Favorskii ring contraction reaction: the effect of ring size.

Project description:The effect of ring size on the photo-Favorskii induced ring-contraction reaction of the hydroxybenzocycloalkanonyl acetate and mesylate esters (7a-d, 8a-c) has provided new insight into the mechanism of the rearrangement. By monotonically decreasing the ring size in these cyclic derivatives, the increasing ring strain imposed on the formation of the elusive bicyclic spirocyclopropanone 20 results in a divergence away from rearrangement and toward solvolysis. Cycloalkanones of seven or eight carbons undergo a highly efficient photo-Favorskii rearrangement with ring contraction paralleling the photochemistry of p-hydroxyphenacyl esters. In contrast, the five-carbon ring does not rearrange but is diverted to the photosolvolysis channel avoiding the increased strain energy that would accompany the formation of the spirobicyclic ketone, the "Favorskii intermediate 20". The six-carbon analogue demonstrates the bifurcation in reaction channels, yielding a solvent-sensitive mixture of both. Employing a combination of time-resolved absorption measurements, quantum yield determinations, isotopic labeling, and solvent variation studies coupled with theoretical treatment, a more comprehensive mechanistic description of the rearrangement has emerged.

Pyrrolidine synthesis via ring contraction of pyridines.

Project description:A ring contraction of easily available cyclic compounds to smaller cycles that are valuable but difficult to synthetically access is one of important skeletal editing strategies. Pyrrolidine synthesis via a ring contraction of pyridines, which are abundant, cheap, and readily available bulk chemicals in chemical industry, is highly promising to accelerate drug discovery and development research due to the great demand of pyrrolidine skeletons in medicinal molecules. Herein we report a photo-promoted ring contraction of pyridines with silylborane to afford pyrrolidine derivatives bearing a 2-azabicyclo[3.1.0]hex-3-ene skeleton. The reaction demonstrates broad substrate scope with high functional group compatibility, realizing facile access to 6-silyl-2-azabicyclo[3.1.0]hex-3-ene derivatives that work as powerful synthons for the synthesis of functionalized pyrrolidines and nitrogen-containing compounds. The reaction mechanism is clarified to proceed via 2-silyl-1,2-dihydropyridine and vinylazomethine ylide as intermediates, which are connected via photochemical or thermal silyl migration.

Phospholenes from Phosphabenzenes by Selective Ring Contraction.

Project description:A 3-amino-functionalized phosphabenzene (phosphinine) has been synthesized and structurally characterized. The pyramidalized nitrogen atom of the dimethylamino substituent indicates only a weak interaction between the lone pair of the nitrogen atom and the aromatic phosphorus heterocycle, resulting in somewhat basic character. It turned out that the amino group can indeed be protonated by HCl. In contrast to pyridines, however, the phosphabenzene-ammonium salt undergoes a selective ring contraction to form a hydroxylphospholene oxide in the presence of additional water. Based on deuterium labeling experiments and quantum chemical calculations, a rational mechanism for this hitherto unknown conversion is proposed.

Formicamycin biosynthesis involves a unique reductive ring contraction.

Project description:Fasamycin natural products are biosynthetic precursors of the formicamycins. Both groups of compounds are polyketide natural products that exhibit potent antibacterial activity despite displaying different three-dimensional topologies. We show here that transformation of fasamycin into formicamycin metabolites requires two gene products and occurs via a novel two-step ring expansion-ring contraction pathway. Deletion of forX, encoding a flavin dependent monooxygenase, abolished formicamycin production and leads to accumulation of fasamycin E. Deletion of the adjacent gene forY, encoding a flavin dependent oxidoreductase, also abolished formicamycin biosynthesis and led to the accumulation of new lactone metabolites that represent Baeyer-Villiger oxidation products of the fasamycins. These results identify ForX as a Baeyer-Villiger monooxygenase capable of dearomatizing ring C of the fasamycins. Through in vivo cross feeding and biomimetic semi-synthesis experiments we showed that these lactone products represent biosynthetic intermediates that are reduced to formicamycins in a unique reductive ring contraction reaction catalyzed by ForY.

Asymmetric one-carbon ring expansion of diverse N-heterocycles via copper-catalyzed diyne cyclization.

Project description:One-carbon ring expansion reaction of N-heterocycles has gained particular attention in the past decade because this method allows for the conversion of readily available N-heterocycles into potentially useful complex ring-expanded N-heterocycles, which are inaccessible by traditional methods. However, the catalytic asymmetric variant of this reaction has been rarely reported to date. Herein, we disclose an enantioselective one-carbon ring expansion reaction through chiral copper-catalyzed diyne cyclization, leading to the practical, atom-economic and divergent assembly of an array of valuable chiral N-heterocycles bearing a quaternary stereocenter in generally good to excellent yields with excellent enantioselectivities (up to >99% ee). This protocol represents the first example of asymmetric one-carbon ring expansion reaction of N-heterocycles based on alkynes.

One-Pot Preparation of Imidazole-Ring-Modified Graphitic Carbon Nitride Nanozymes for Colorimetric Glucose Detection.

Project description:Nanozymes are highly desired to overcome the shortcomings of natural enzymes, such as low stability, high cost and difficult storage during biosensing applications. Herein, by imitating the structure of natural enzymes, we propose a one-pot annealing process to synthesis imidazole-ring-modified graphitic carbon nitride (g-C3N4-Im) with enhanced peroxidase-like activity. g-C3N4-Im shows enhanced peroxidase-like activity by 46.5 times compared to unmodified g-C3N4. Furthermore, imidazole rings of g-C3N4-Im make it possible to anchor Cu(II) active sites on it to produce g-C3N4-Im-Cu, which shows a further increase in peroxidase-like activity by three times. It should be noted that the as-prepared g-C3N4-Im-Cu could show obvious peroxidase-like activity over a broad range of pH values and at a low temperature (5 °C). The ultrahigh peroxidase-like activity is attributed to the electronic effect of imidazole rings and the active sites of Cu(II) for ·OH production. Based on the enhanced peroxidase-like activity, a H2O2 and glucose biosensor was developed with a high sensitivity (limit of detection, 10 nM) and selectivity. Therefore, the biosensor shows potential for applications in diabetic diagnoses in clinical practice.
