Genome Sequences of Microviruses Identified in a Sample from a Sewage Treatment Oxidation Pond.

ABSTRACT: Oxidation ponds are often used in the treatment of sewage as an aeration step prior to discharge. We identified 99 microvirus genomes from a sample from a sewage oxidation pond. This diverse group of microviruses expands our knowledge of bacteriophages associated with sewage oxidation pond ecosystems.

Characterization of the chloroplast genome of <i>Chlorolobion braunii</i> ITBB-AG6 isolated from a treatment pond of sanitary sewage.

Project description:Sphaeropleales is an order of fast-growing microalgae with high oil content and high efficiency in sewage treatment, in which photosynthesis plays a critical role. We isolated a strain of Sphaeropleales, Chlorolobion braunii ITBB-AG6 from an azolla community in a sewage pond, and sequenced its chloroplast genome. The complete genome has a length of 154 kb with a GC content of 31.7%. A total of 89 genes were annotated, including 56 protein-coding genes, 30 tRNA genes, and three rRNA genes. Out of the protein coding genes, 64.3% are involved in photosynthesis, 28.6% are involved in protein synthesis, and 7.1% are involved in ATP synthesis. Transfer RNA genes for 20 amino acids were identified, in which tRNA genes for methionine, leucine, and arginine are tripled, whereas tRNA genes for glutamic acid, glycine, serine, and threonine are doubled. Terminal inverted repeats of 27.9 kb containing 10 genes related to photosynthesis and chloroplast division are present in the genome, suggesting that photosynthesis was strengthened in the evolutionary history. Phylogenetic analysis indicates that C. braunii ITBB-AG6 falls in the family Selenastraceae and is most closely related to Monoraphidium neglectum.

Synergistic effects of bacterial consortium and thermal energy on treatment of sewage by waste stabilisation pond.

Project description:The low rates of biodegradation of organic pollutants in wastewater have been attributed to the daily fluctuation of temperatures, which affects microbial metabolism and activities in reactors. This work aimed to develop a method to degrade sewage pollutants using a synergistic effect of bacterial consortium and thermal energy, while a grey concrete pond served as the control. The results demonstrated that the temperature profile of ICCP showed that all through the experiment, the temperature was above 25 °C, which is a suitable temperature for mesophilic bacterial growth. A properly-stabilised effluent was achieved by the ICCP with a low biodegradation index between 0.11 and 0.14. The values of BOD (95%) and COD (74%) removal efficiencies were obtained at a 10-day retention time in ICCP, which is in accordance with standard of the United State Environmental protection Agency. Moreover, a comparison between a control and ICCP revealed that the latter emits heat energy 30% higher than the first. The temperature of 30 °C (dark) and 30.8 °C (light) produced a BOD removal > 90%. Therefore, this method could be considered to bridge the gap in daily fluctuation of temperature for enhanced biodegradation.•Designing of a thermal coated concrete pond to investigate their thermal performance during the dark and light condition•Bioremediation test for selection of mixed bacteria strain of high degradation potential used as inoculum•A detention time of 10 days under natural sunlight used for investigation for concentration balance of organic pollutant.

Complete Annotated Genome Sequences of Four Klebsiella pneumoniae Phages Isolated from Sewage in Poland.

Project description:Four lytic phages, vB_KpnP_BIS33, vB_KpnP_IL33, and vB_KpnP_PRA33 of the Podoviridae family and vB_KpnM_BIS47 of the Myoviridae family, which act against animal-pathogenic Klebsiella pneumoniae strains, were isolated from sewage plants in Poland. They possess double-stranded DNA genomes of 41,697 bp, 41,335 bp, 40,605 bp, and 147,443 bp, respectively.

Hepatitis E Virus Genotype 4 Sequences Detected in Sewage from Treatment Plants of China.

Project description:The aim of this study was to investigate the occurrence of hepatitis E virus (HEV) in sewage samples in Shen Zhen, China. Sewage samples were collected from 152 sewage plants including livestock sewage, domestic sewage and treated sewage from May to July of 2015. Two of 152 samples were HEV positive (1.32%) from the livestock sewage plants. Partial ORF2 fragments of HEV were sequenced and a phylogenetic tree was constructed using MEGA5.1. Blast and phylogenetic analyses showed that both of these two sequences belonged to HEV Genotype 4. To the best of our knowledge, this is the first study on the molecular characterization of HEV in wastewater in China and the first time to detect Genotype 4 in the sewage. Results from this study indicate that the possibilities of sporadic infections of HEV should be emphasized because virus still has the possibility to be circulating in the sewage in China.
