The complete mitochondrial genome of Scatoglyphus polytrematus (Acari: Acaridae).

ABSTRACT: We assembled and annotated the complete mitochondrial genome of Scatoglyphus polytrematus. It is the first complete mitochondrial genome sequence from the genus Scatoglyphus. The mitogenome was 13,966 bp in length and contains 37 genes (including 13 protein-coding genes, 22 transfer RNA (tRNA), and two ribosomal RNA (rRNA)), and one largest non-coding region. The gene arrangement of S. polytrematus is consistent with the pattern of possible common ancestor of astigmatid mites. In the present study, phylogenetic analysis shows that genus Scatoglyphus was clustered into one branch with other Acaridae species.

Project description:Dermacentor (Indocentor) auratus Supino, 1897 is a prominent ixodid vector of numerous pathogens of public health and veterinary importance. Using long-range PCR of two overlapping regions sequenced on an Illumina MiSeq machine, the complete mitochondrial genome of D. auratus is reported here. The resulting contigs were able to be assembled into a complete and circularised genome which had the general organisation of the mitochondrial genomes of the Metastriates. It had a total length of 14,766 bp and contained 37 genes, including 13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes, as well as 2 non-coding control regions and 3 tick-boxes. The phylogenetic analysis on the whole mitogenome confirmed the position of D. auratus within the Dermacentor clade.

The complete mitochondrial genome of Histiostoma blomquisti (Acari: Histiostomatidae).

Project description:The mite Histiostoma blomquisti is a microorganism feeder that uses the red imported fire ant (Solenopsis invicta) as a phoretic carrier for dispersal. We sequenced the H. blomquisti mitogenome using next-generation sequencing methods. The circular mitogenome of H. blomquisti is 15,892 bp and is composed of 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and 6 non-coding regions >100 bp. Most tRNAs are highly reduced, like those found in other Acariformes. Phylogenetic analysis based on the concatenated nucleotide sequence of the 13 protein-coding genes supports Histiostomatid mites forming the basal-most lineage in Astigmata.

The complete mitochondrial genome of Ixodes vespertilionis (Acari: Ixodidae).

Project description:Ixodes vespertilionis is a tick parasitizing on the bodies of bats. In our study, the complete mitogenome of I. vespertilionis was determined by using Illumina sequencing technology. The mitogenome was 14,559 bp in size and was predicted to encode 37 genes including 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and one control region. The gene order of the mitogenome is identical to Argasidae and non-Australasian Prostriata. The phylogenetic analysis by the Maximum-likelihood method reveals that I. vespertilionis is phylogenetically closest to Ixodes simplex. These data provide novel reference for further studies on the population genetics and phylogenetics of ticks.

Complete mitochondrial genome and phylogenetic analysis of Ixodes ovatus (Acari: Ixodidae).

Project description:Ixodes ovatus is referred to as an obligatory blood-sucking ectoparasite that is capable to infest both humans and animals. In the present study, the complete mitochondrial genome of I. ovatus was sequenced and analyzed using next-generation sequencing (NGS) technology. With a size of 14,520 bp, the entire mitogenome contains 37 genes including 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), 2 ribosomal RNAs (rRNAs), and 3 control regions (D-loops). Based on the 13 PCGs nucleotide sequences, the phylogenetic relationship of I. ovatus was analyzed using Maximum-likelihood. As suggested by the results of the obtained phylogenety, I. ovatus is most closely associated with Ixodes hexagonus. This study is expected to promote further studies on the evolution of Ixodidae.

Complete mitochondrial genome of Hygrobates turcicus Pesic, Esen & Dabert, 2017 (Acari, Hydrachnidia, Hygrobatoidea).

Project description:The aim of the study was sequencing of the mitogenome of Hygrobates turcicus Pešić, Esen & Dabert, 2017 to expand knowledge of the polymorphism and cryptic or pseudocryptic diversity within Hydrachnidia. The samples originated from Bulgaria, Vidima River near Debnewo, 42°56'41.4''N, 24°48'44.6''E, depth 0.4 m, stones on the bottom, water flow 0.71 m/s, temperature 10 °C, pH 8.53, oxygen 110%, conductivity 279 µS/cm, hardness 121 CaO mg/l; 11 males, 27 females, 2 deutonymphs 12.x.2019 leg. Zawal, Michoński & Bańkowska; one male and one female dissected and slides mounted. The study was carried out using the following methods: DNA extraction, sequencing, assembly and annotation, comparison with other populations of H. turcicus, and multigene phylogeny. As a result of the study, it was determined that the mitogenome is 15,006 bp long and encodes for 13 proteins, 2 rRNAs, and 22 tRNAs. The genome is colinear with those of H. longiporus and H. taniguchii, the difference in size originating from a non-coding region located between protein-coding genes ND4L and ND3. Five genes have alternative start-codon, and four display premature termination. The multigene phylogeny obtained using all mitochondrial protein-coding genes unambiguously associates H. turcicus with the cluster formed by H. longiporus and H. taniguchii.

Determination of the complete mitochondrial genome of Lardoglyphus konoi (Acari: Astigmata).

Project description:The mitochondrial genome of Lardoglyphus konoi was 14,269 bp long and consisted of 37 genes including 22 transfer RNAs, 13 protein-coding genes and 2 ribosomal RNA genes. All the 13 protein-coding genes had complete start/stop codons. Most inferred tRNAs were extremely truncated and the T- or D-arm was missing. The position of two rRNAs and the largest non-coding region were separated by tRNAs. The Bayesian phylogenetic tree indicated that L. konoi was closely related to Carpoglyphus lactis.

Project description:In this study, we determined the mitogenome sequence of Ardeacarus ardeae (Canestrini, 1878) in the family Pterolichidae (Acari, Sarcoptiformes), which is the first complete mitogenome sequence in feather mite. The mitogenome of A. ardeae is 14,069 bp in length and contains 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), 2 ribosomal RNAs (rRNAs), and a control region (CR). The phylogenetic tree shows that A. ardeae belong to the supercohort Desmonomatides within the order Sarcoptiformes.

The complete mitochondrial genome and phylogenetic analysis of Haemaphysalis longicornis Neumann (Acari: Ixodidae).

Project description:Haemaphysalis longicornis ticks are vectors or reservoirs of numerous infectious pathogens and cause a variety of human and animal diseases worldwide. However, there is limited knowledge on available genetic sequence. Herein, we extracted the complete mitochondrial genome (mitogenome) from enriched mitochondria of H. longicornis first time in ticks and gained its sequence with 14,718bp in length. The mitogenome consisted of 13 PCGs, 22 tRNA, 2 rRNA, and 2 noncoding regions. Also, the monophyletic phylogenetic position of H. longicornis is inferred based on 28 complete mitogenomes in total comprised of various species from Ixodida ticks in addition to the mitogenome of H. longicornis.

Characterization of the complete mitochondrial genome of the predatory mite Stratiolaelaps scimitus (Acari: Laelapidae).

Project description:In this study, we recovered the complete mitochondrial genome of Stratiolaelaps scimitus through Illumina sequencing data. The circularized mitogenome is 16,009 in length, which consists of 37 genes (13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes). The overall base composition is 35.4% for A, 40.1% for T, 16.8% for G, 7.7% for C, demonstrating an extreme bias of high AT content (75.5%). The whole mitogenome of S. scimitus and other Acari mitogenomes (10 species, in total) were used for phylogenetic analysis, and the result showed that the relationship of S. scimitus was close to Varroa destructor in the same Superfamily Dermanyssoidea.

Project description:BACKGROUND: Limited available sequence information has greatly impeded population genetics, phylogenetics and systematics studies in the subclass Acari (mites and ticks). Mitochondrial (mt) DNA is well known to provide genetic markers for investigations in these areas, but complete mt genomic data have been lacking for many Acari species. Herein, we present the complete mt genome of the scab mite Psoroptes cuniculi. METHODS: P. cuniculi was collected from a naturally infected New Zealand white rabbit from China and identified by morphological criteria. The complete mt genome of P. cuniculi was amplified by PCR and then sequenced. The relationships of this scab mite with selected members of the Acari were assessed by phylogenetic analysis of concatenated amino acid sequence datasets by Bayesian inference (BI), maximum likelihood (ML) and maximum parsimony (MP). RESULTS: This mt genome (14,247 bp) is circular and consists of 37 genes, including 13 genes for proteins, 22 genes for tRNA, 2 genes for rRNA. The gene arrangement in mt genome of P. cuniculi is the same as those of Dermatophagoides farinae (Pyroglyphidae) and Aleuroglyphus ovatus (Acaridae), but distinct from those of Steganacarus magnus (Steganacaridae) and Panonychus citri (Tetranychidae). Phylogenetic analyses using concatenated amino acid sequences of 12 protein-coding genes, with three different computational algorithms (BI, ML and MP), showed the division of subclass Acari into two superorders, supported the monophylies of the both superorders Parasitiformes and Acariformes; and the three orders Ixodida and Mesostigmata and Astigmata, but rejected the monophyly of the order Prostigmata. CONCLUSIONS: The mt genome of P. cuniculi represents the first mt genome of any member of the family Psoroptidae. Analysis of mt genome sequences in the present study has provided new insights into the phylogenetic relationships among several major lineages of Acari species.
