Effects of Supplementing Rumen-Protected Methionine and Lysine on Milk Performance and Oxidative Status of Dairy Ewes.

ABSTRACT: There is limited information on the impact of dietary supplementation with separate rumen-protected (RP) amino acids (AA), or with their combination, on ewes' oxidative status. Sixty ewes were divided into five groups; C: basal diet (control); M: basal diet + 6 g/ewe RP methionine; L: basal diet + 5 g/ewe RP lysine; LML: basal diet + 6 g methionine and 5 g lysine/ewe; and HML: basal diet + 12 g methionine + 5 g lysine/ewe. Milk's fat content increased in RP-AA fed ewes, while that of protein in M and L only. In blood plasma, the malondialdehyde (MDA) content was reduced in the M, LML, and HML compared to C-fed ewes. An increase in glutathione transferase activity in the blood plasma of the M and LML compared to the C and HML-fed ewes were found. In milk, lower values of the ferric reducing ability of plasma (FRAP) in the LML and HML-fed ewes and of 2,2'-Azino-bis 3-ethylbenzthiazoline-6-sulfonic acid (ABTS) in LML only, were found. Lysine increased milk's FRAP values and MDA content. Both L and HML diets increased milk's protein carbonyls content. Methionine improves the organism's oxidative status, without adversely affecting milk's oxidative stability. Lysine dietary inclusion affects negatively the oxidative stability of milk.

Project description:ObjectiveOur objective was to examine the relationships of supplemental rumen-protected lysine (RPL) or lysine + methionine (RPLM) on lactational performance, plasma amino acids (AA) concentration, and nitrogen use efficiency of lactating dairy cows by using a meta-analysis approach.MethodsA total of 56 articles comprising 77 experiments with either RPL or RPLM supplementation were selected and analyzed using a mixed model methodology by considering the treatments and other potential covariates as fixed effects and different experiments as random effects.ResultsIn early lactating cows, milk yield was linearly increased by RPL (β1 = 0.013; p<0.001) and RPLM (β1 = 0.014; p<0.028) but 3.5% fat-corrected milk (FCM) and energy-corrected milk (ECM) (kg/d) was increased by only RPL. RPL and RPLM did not affect dry matter intake (DMI) but positively increased (p<0.05) dairy efficiency (Milk yield/DMI and ECM/DMI). As a percentage, milk fat, protein, and lactose were unchanged by RPL or RPLM but the yield of all components was increased (p<0.05) by feeding RPL while only milk protein was increased by feeding RPLM. Plasma Lys concentration was linearly increased (p<0.05) with increasing supplemental RPL while plasma Met increased (p<0.05) by RPLM supplementation. The increase in plasma Lys had a strong linear relationship (R2 = 0.693 in the RPL dataset and R2 = 0.769 in the RPLM dataset) on milk protein synthesis (g/d) during early lactation. Nitrogen metabolism parameters were not affected by feeding RPL or RPLM, either top-dress or when supplemented to deficient diets. Lactation performance did not differ between AA-deficient or AA-adequate diets in response to RPL or RPLM supplementation.ConclusionRPL or RPLM showed a positive linear relationship on the lactational performance of dairy cows whereas greater improvement effects were observed during early lactation. Supplementing RPL or RPLM is recommended on deficient-AA diet but not on adequate-AA diet.

Project description:Glutathione (GSH), widely present in plant and animal cells and crucial for combating oxidative stress and inflammation, has not been evaluated in dairy cows. This study aims to evaluate the effects of rumen-protected glutathione (RPGSH) supplementation on lactation, nutrient metabolism, oxidative stress, inflammation, and health in transition dairy cows. Forty Holstein dairy cows (2.65 ± 0.78 of parity, 2.81 ± 0.24 of body condition score, 9207.56 ± 1139.18 kg of previous 305-day milk yield, 657.53 ± 55.52 kg of body weight, mean ± SD) were selected from a large cohort of 3215 cows on day 21 before expected calving (day -21 ± 3 d). Cows were randomly stratified into four dietary treatment groups (n = 10 per group): control (basal diet + 0 g/d RPGSH); T1 (basal diet + 1.5 g/d RPGSH); T2 (basal diet + 2 g/d RPGSH); and T3 (basal diet + 3 g/d RPGSH). Supplementation commenced approximately 21 days (±3) prepartum and continued through 21 days postpartum. Blood samples were collected at -21 ± 3, -14 ± 3, -7 ± 3, 0, 7, 14, and 21 d for analysis of serum metabolic parameters related to oxidative stress and inflammation. Milk composition was analyzed from samples collected on days 3, 7, 14, and 21 postpartum. Compared with the control group, supplementation with 2 g/d of RPGSH reduced somatic cell count (p < 0.05) and the incidence of postpartum diseases in dairy cows. No differences were observed among the groups in milk yield, milk fat, protein, lactose, total solids, dry matter intake, or energy-corrected milk. However, fat-corrected milk and feed efficiency were higher in the T2 group compared to the control (p < 0.05). Calcium and phosphorus levels did not differ among the groups. Compared to the control group, cows supplemented with 2 g/d RPGSH had lower β-hydroxybutyrate levels and higher glucose levels on days 14 and 21 postpartum (p < 0.05). From days 14 to 21 postpartum, RPGSH supplementation increased blood GSH, serum catalase, and total antioxidant capacity while reducing malondialdehyde, reactive oxygen species, haptoglobin, cortisol, C-reactive protein, and interleukin-6 levels compared with the control group (p < 0.05). The supplementation of 2 g/d RPGSH showed relatively better effects. RPGSH supplementation at 2 g/d improved lactation performance, nutrient metabolism, oxidative stress, and inflammation status in dairy cows, playing a crucial role in maintaining their health. To our knowledge, this is the first report on the effects of supplementing RPGSH additive in Holstein cows.

Project description:The aim of this study was to investigate the effect of low-protein diets supplemented with rumen-protected lysine (RPLys) and methionine (RPMet) on growth performance, rumen fermentation, blood biochemical parameters, nitrogen metabolism, and gene expression related to N metabolism in the liver of Holstein bulls. Thirty-six healthy and disease-free Holstein bulls with a similar body weight (BW) (424 ± 15 kg, 13 months old) were selected. According to their BW, they were randomly divided into three groups with 12 bulls in each group in a completely randomized design. The control group (D1) was fed with a high-protein basal diet (CP13%), while bulls in two low-protein groups were supplied a diet with 11% crude protein and RPLys 34 g/d·head + RPMet 2 g/d·head (low protein with low RPAA, T2) or RPLys 55 g/d·head + RPMet 9 g/d·head (low protein with high RPAA, T3). At the end of the experiment, the feces and urine of dairy bulls were collected for three consecutive days. Blood and rumen fluid were collected before morning feeding, and liver samples were collected after slaughtering. The results showed that the average daily gain (ADG) of bulls in the T3 group was higher than those in D1 (p < 0.05). Compared with D1, a significantly higher nitrogen utilization rate (p < 0.05) and serum IGF-1 content (p < 0.05) were observed in both T2 and T3 groups; however, blood urea nitrogen (BUN) content was significantly lower in the T2 and T3 groups (p < 0.05). The content of acetic acid in the rumen of the T3 group was significantly higher than that of the D1 group. No significant differences were observed among the different groups (p > 0.05) in relation to the alpha diversity. Compared with D1, the relative abundance of Christensenellaceae_R-7_group in T3 was higher (p < 0.05), while that of Prevotellaceae _YAB2003_group and Succinivibrio were lower (p < 0.05). Compared with D1 and T2 group, the T3 group showed an expression of messenger ribonucleic acid (mRNA) that is associated with (CPS-1, ASS1, OTC, ARG) and (N-AGS, S6K1, eIF4B, mTORC1) in liver; moreover, the T3 group was significantly enhanced (p < 0.05). Overall, our results indicated that low dietary protein (11%) levels added with RPAA (RPLys 55 g/d +RPMet 9 g/d) can benefit the growth performance of Holstein bulls by reducing nitrogen excretion and enhancing nitrogen efficiency in the liver.

Project description:BackgroundDietary protein level and amino acid (AA) balance are crucial determinants of animal health and productivity. Supplementing rumen-protected AAs in low-protein diets was considered as an efficient strategy to improve the growth performance of ruminants. The colon serves as a crucial conduit for nutrient metabolism during rumen-protected methionine (RPMet) and rumen-protected lysine (RPLys) supplementation, however, it has been challenging to clarify which specific microbiota and their metabolites play a pivotal role in this process. Here, we applied metagenomic and metabolomic approaches to compare the characteristic microbiome and metabolic strategies in the colon of lambs fed a control diet (CON), a low-protein diet (LP) or a LP diet supplemented with RPMet and RPLys (LR).ResultsThe LP treatment decreased the average daily weight gain (ADG) in lambs, while the LR treatment tended to elicit a remission in ADG. The butyrate molar concentration was greater (P < 0.05), while acetate molar concentration (P < 0.05) was lower for lambs fed the LP and LR diets compared to those fed the CON diet. Moreover, the LP treatment remarkably decreased total AA concentration (P < 0.05), while LR treatment showed an improvement in the concentrations of methionine, lysine, leucine, glutamate, and tryptophan. Metagenomic insights proved that the microbial metabolic potentials referring to biosynthesis of volatile fatty acids (VFAs) and AAs in the colon were remarkably altered by three dietary treatments. Metagenomic binning identified distinct microbial markers for the CON group (Alistipes spp., Phocaeicola spp., and Ruminococcus spp.), LP group (Fibrobacter spp., Prevotella spp., Ruminococcus spp., and Escherichia coli), and LR group (Akkermansia muciniphila and RUG099 spp.).ConclusionsOur findings suggest that RPMet and RPLys supplementation to the low-protein diet could enhance the microbial biosynthesis of butyrate and amino acids, enriche the beneficial bacteria in the colon, and thereby improve the growth performance of lambs.

Project description:To investigate the difference between rumen-protected niacin (RPN) and rumen-protected nicotinamide (RPM) in the transcriptome of genes relating to the lipid metabolism of the liver of periparturient dairy cows, 10 healthy Chinese Holstein cows were randomly divided into two groups and fed diets supplemented with 18.4 g/d RPN or 18.7 g/d RPM, respectively. The experiment lasted from 14 days before to 21 days after parturition. Liver biopsies were taken 21 days postpartum for transcriptomic sequencing. In addition, human LO2 cells were cultured in a medium containing 1.6 mmol/L of non-esterified fatty acids and 1 mmol/L niacin (NA) or 2 mmol/L nicotinamide (NAM) to verify the expression of the 10 genes selected from the transcriptomic analysis of the liver biopsies. The expression of a total of 9837 genes was detected in the liver biopsies, among which 1210 differentially expressed genes (DEGs) were identified, with 579 upregulated and 631 downregulated genes. These DEGs were associated mainly with lipid metabolism, oxidative stress, and some inflammatory pathways. Gene ontology (GO) enrichment analysis showed that 355 DEGs were enriched in 38 GO terms. The differences in the expression of these DEGs between RPN and RPM were predominantly related to the processes of steroid catabolism, steroid hydroxylase, monooxygenase activity, oxidoreductase activity, hemoglobin binding, and ferric iron binding, which are involved mainly in lipid anabolism and redox processes. The expressions of FADS2, SLC27A6, ARHGAP24, and THRSP in LO2 cells were significantly higher (p < 0.05) while the expressions of BCO2, MARS1, GARS1, S100A12, AGMO, and OSBPL11 were significantly lower (p < 0.05) on the NA treatment compared to the NAM treatment, indicating that NA played a role in liver metabolism by directly regulating fatty acid anabolism and transport, inflammatory factor expression, and oxidative stress; and NAM functioned more as a precursor of nicotinamide adenine dinucleotide (NAD, coenzyme I) and nicotinamide adenine dinucleotide phosphate (NADP, coenzyme II) to participate indirectly in biological processes such as ether lipid metabolism, cholesterol metabolism, energy metabolism, and other processes.
