Trace Element and Mineral Levels in Serum, Hair, and Urine of Obese Women in Relation to Body Composition, Blood Pressure, Lipid Profile, and Insulin Resistance.

ABSTRACT: The objective of this study was to evaluate serum, hair, and urinary trace element and mineral content in normal-weight and obese women in relation to metabolic risk factors. A total of 80 women aged 30-70 y.o. were enrolled in the obese group (n = 40) and normal-weight group (n = 40). Serum, hair, and urinary trace element and mineral levels were assessed using inductively coupled plasma spectrometry. Body fat percentage was evaluated using bioimpedance. Obese subjects were characterized by significantly higher body fat percentage, blood pressure, serum triglyceride concentration, and insulin resistance. Serum Ca, Fe, Mg, Se, V, Zn levels, hair Fe, Mg, V content, and urinary Se and V concentrations were found to be lower in obese subjects as compared to lean controls. In turn, serum Cu and urinary Fe levels in obese women were characterized by a significant increase. In multiple regression models serum Cu, Se, and Zn levels were significantly associated with BMI even after adjustment for blood biochemistry, body composition, and blood pressure. Serum trace element and mineral levels also significantly contributed to group discrimination. These findings allow to propose that obesity-associated disturbances in trace element and mineral status may at least partially contribute to metabolic risk in obese subjects.

Project description:Honey is an extensively utilized sweetener containing sugars and water, together with small quantities of vitamins, minerals, fatty acids, amino acids and proteins. Naturally produced by honeybees (Apis mellifera) from floral nectar, honey is increasingly sold as a health food product due to its nutritious features. Certain honeys are retailed as premium, trendy products. Honeybees are regarded as environmental monitors, but few reports examine the impact of environment on Australian honey trace elements and minerals. In higher density urban and industrial environments, heavy metals can be common, while minerals and trace elements can have ubiquitous presence in both agricultural and urban areas. Honey hives are traditionally placed in rural and forested areas, but increasingly the trend is to keep hives in more urban areas. This study aimed to determine the levels of 26 minerals and trace elements and assess elemental differences between honeys from various regional Queensland and Australian sources. Honey samples (n = 212) were acquired from markets, shops and supermarkets in Queensland while urban honeys were purchased online. The honey samples were classified into four groups according to their regional sources: urban, rural, peri-urban and blend honey. Elemental analyses of honey were performed using ICP-MS and ICP-OES after microwave and hot block digestion. Considerable variations of essential trace elements (Co, Cu, Cr, Fe, Mn, Mo and Zn) and mineral levels (Ca, K, Mg, Na and P) were found in honeys surveyed. There were significant differences (p < 0.05) between urban and rural honey samples for B, Na, P, Mn, K, Ca and Cu. Significant differences (p < 0.05) were also found between blend and urban honey samples for K, Cu, P, Mn, Sr, Ni, B and Na. Peri-urban versus urban honeys showed significant differences in P, K and Mn. For rural and peri-urban honeys, the only significant difference (p < 0.05) was for Na. Toxic heavy metals were detected at relatively low levels in honey products. The study revealed that the Queensland/Australian honey studied is a good source of K and Zn and would constitute a good nutritional source of these elements.

Project description:Two experiments were conducted to determine: 1) the impact of strained rumen fluid (SRF) alone or SRF with particle-associated microorganisms (PAO) included and dilution on in vitro dry matter digestibility (DMD) and 2) the impact of trace mineral (TM) source on in vitro fermentation characteristics and TM solubility under simulated abomasal and intestinal conditions. In experiment 1, 3 cannulated steers were adapted to a diet formulated to meet the nutrient requirements for lactating dairy cows. Strained RF was obtained by straining rumen content through 2 layers of cheesecloth. Half of the remaining digesta was washed with McDougall's buffer and filtered through 2 layers of cheesecloth to obtain PAO. Both SRF and PAO were filtered again through 8 layers of cheesecloth. Strained RF was mixed with either McDougall's buffer (SRF) or PAO (SRF+ + PAO) at a ratio of 1:2 or 1:4 and incubated at 39 °C for 12 h using the ground basal diet as the substrate. Digestibility of DM was greater in digestion tubes containing SRF and SRF + PAO at a 1:2 ratio. In experiment 2, 8 steers fitted with a ruminal cannula were blocked by body weight and assigned to 1 of the 2 treatment groups. Treatments consisted of 10 mg Cu, 40 mg Mn, and 60 mg Zn/kg DM from either: 1) sulfate (STM) or 2) hydroxychloride (HTM) sources. Steers were housed in individual pens and fed the same diet as described in experiment 1. Dietary TM treatments were mixed with dried distiller grains and mixed in the diet, by hand, immediately after basal diet delivery. Dietary treatments were fed for 14 d. On day 15, SRF + PAO was collected from each steer (STM-RF and HTM-RF) and used in a series of in vitro crossover experiments. In vitro substrates (S) used were the ground diets consumed by the animals on each treatment (STM-S and HTM-S). Incubations containing HTM-S had greater (P < 0.01) total volatile fatty acid (VFA) concentration and propionic acid molar proportions, but lesser (P < 0.01) acetic acid molar proportions than STM-S. Rumen fluid from steers supplemented with HTM had a greater (P < 0.03) total VFA than STM-RF at 24 h post incubation. After 12 h post incubation, the molar proportion of propionic acid in HTM-RF was lesser (P = 0.04) than in STM-RF. After simulated abomasal digestion, soluble Mn concentration in HTM-S was greater (P < 0.01) than in STM-S. These data indicate that the source of TM can influence in vitro rumen fermentation characteristics and Mn solubility under simulated abomasal conditions.
