The Taxonomic History of Ochlerotatus Lynch Arribalzaga, 1891 (Diptera: Culicidae).

ABSTRACT: A review of all taxonomic actions within the subgenus Ochlerotatus Lynch Arribálzaga, 1891 (Diptera: Culicidae) sensu Reinert et al. (2008) is provided. In particular, the complex historical taxonomic treatment of the type species of this group is dissected and explained in detail. Additionally, current challenges with the definition of the subgenus and its constituents are discussed, as are the requisite steps for a successful revision of the taxon. Going forward, we conclude that a taxonomic revision of the species should include a neotype designation for Ochlerotatus scapularis (Rondani, 1848) from topotypical material. Additionally, we provide a review of the characters and taxa that need to be re-evaluated and well-described in order to stabilize the taxonomy of the subgenus. This effort represents a key step towards a stable nomenclature of the Tribe Aedini.

Potential Distribution of Aedes (Ochlerotatus) scapularis (Diptera: Culicidae): A Vector Mosquito New to the Florida Peninsula.

Project description:Aedes scapularis is a neotropical mosquito known to transmit pathogens of medical and veterinary importance. Its recent establishment in southeastern Florida has potential public health implications. We used an ecological niche modeling approach to predict the abiotic environmental suitability for Ae. scapularis across much of the Americas and Caribbean Islands. Georeferenced occurrence data obtained from the Global Biodiversity Inventory Facility and recent collection records of Ae. scapularis from southern Florida served as input for model calibration. Environmental layers included bioclimatic variables provided in 2000 to 2010 average Modern Era Retrospective-analysis for Research and Applications climatic (MERRAclim) data. Models were run in the software program Maxent. Isothermality values often found in costal environments, had the greatest contribution to model performance. Model projections suggested that there are areas predicted to be suitable for Ae. Scapularis across portions of the Amazon Basin, the Yucatán Peninsula, the Florida Peninsula, and multiple Caribbean Islands. Additionally, model predictions suggested connectivity of highly suitable or relatively suitable environments spanning the United States Gulf Coast, which may facilitate the geographic expansion of this species. At least sixteen Florida counties were predicted to be highly suitable for Ae. scapularis, suggesting that vigilance is needed by vector control and public health agencies to recognize the further spread of this vector.

Description of six new large species of Argentinomyia Lynch-Arribalzaga, 1891 and redescription of Talahua fervida (Fluke, 1945) (Diptera, Syrphidae, Syrphinae).

Project description:The morphological similarities between five new large Argentinomyia species and Talahua fervida Fluke are characterized and presented. Six new species of Argentinomyia (10-12 mm long) are described: Argentinomyia andina Montoya & Wolff, sp. nov. (Colombia), Argentinomyia choachi Montoya, sp. nov. (Colombia), Argentinomyia quimbaya Montoya & Wolff, sp. nov. (Colombia), Argentinomyia huitepecensis Montoya, sp. nov. (México), Argentinomyia puntarena Montoya, sp. nov. (Costa Rica), and Argentinomyia talamanca Thompson, sp. nov. (Costa Rica). The genus Talahua Fluke is re-diagnosed and, Talahua fervida redescribed. A taxonomic key and a comparison of diagnostic characters are presented. Photographs of head, abdominal and wing maculae patterns, as well as illustrations of male genitalia are provided for species identification.

The salivary transcriptome of Anopheles gambiae (Diptera: Culicidae) larvae: A microarrray-based study

Project description:In spite of the many recent developments in the field of vector sialomics, the salivary glands of larvalmosquitoes have been largely unexplored. We used whole-transcriptome microarray analysis to create a gene-expression profile of the salivary gland tissue of fourth-instar Anopheles gambiae larvae, and compare it to the gene-expression profile of a matching group of whole larvae. We identified a total of 221 probes with expression values that were (a) significantly enriched in the salivary glands, and (b)sufficiently annotated as to allow the prediction of the presence/absence of signal peptides in their corresponding gene products. Based on available annotation of the protein sequences associated with these probes, we propose that the main roles of larval salivary secretions include: (a) immune response, (b) mouthpart lubrication, (c) nutrient metabolism, and (d) xenobiotic detoxification. Other highlights of the study include the cloning of a transcript encoding a previously unknown salivary defensin (AgDef5), the confirmation of mucus secretion by the larval salivary glands, and the first report of salivary lipocalins in the Culicidae. Molecular operational taxonomic units of mosquitoes (Diptera: Culicidae) collected in high Andean mountain ecosystems of Antioquia, Colombia.

Project description:Accurate taxonomic identification of highland mosquito species may be complicated because of the lack of comprehensive regional morphological keys and taxonomic specialists, particularly for mosquitoes of medical or ecological importance. We applied a multi-locus approach to explore the diversity of genera/species collected, to define the Molecular Operational Taxonomic Units (MOTUs) and to perform phylogenetic clustering. Twenty MOTUs and three single sequences were revealed from 78 concatenated cox1 + ITS2 sequences, and the species name was allocated for five of these. This study provides molecular taxonomic information of culicid fauna present in high Andean mountain ecosystems in Antioquia, Colombia. However, future morphological and integrative taxonomic studies should be conducted to achieve the specific identity of all detected MOTUs.

Anopheles darlingi (Diptera: Culicidae) in Panama.

Project description:We report Anopheles darlingi in Darien Province in eastern Panama. Polymerase chain reaction-restriction fragment length polymorphism profiles of the single copy nuclear white gene and sequence comparisons confirmed the presence of 66 specimens of the northern lineage of An. darlingi. The parsimony network depicted 5 CO1 haplotypes in 40 specimens of An. darlingi, which connected through 7-8 mutational steps with sequences from Central and South America. Furthermore, the presence of haplotypes in Biroquera, Darien Province identical to those previously published from northern Colombia suggests that Panamanian samples originated in Colombia. Results of neutrality tests (R(2) and Fu's F(S)) were not significant and the mismatch distribution was multimodal and did not fit the model of sudden population growth. These findings may indicate a long and stable presence of An. darlingi in eastern Panama.

Transmission Potential of Zika Virus by Aedes aegypti (Diptera: Culicidae) and Ae. mediovittatus (Diptera: Culicidae) Populations From Puerto Rico.

Project description:Recurrence of local transmission of Zika virus in Puerto Rico is a major public health risk to the United States, where mosquitoes Aedes aegypti (Linnaeus) and Aedes mediovittatus (Coquillett) are abundant. To determine the extent to which Ae. mediovittatus are capable of transmitting Zika virus and the influence of viremia, we evaluated infection and transmission in Ae. mediovittatus and Ae. aegypti from Puerto Rico using serial dilutions of infectious blood. Higher doses of infectious blood resulted in greater infection rates in both mosquitoes. Aedes aegypti females were up to twice as susceptible to infection than Ae. mediovittatus, indicating a more effective midgut infection barrier in the latter mosquito species. Aedes aegypti exhibited higher disseminated infection (40-95%) than Ae. mediovittatus (<5%), suggesting a substantial midgut escape barrier in Ae. mediovittatus. For Ae. aegypti, transmission rates were low over a range of doses of Zika virus ingested, suggesting substantial salivary gland barriers.

Land Use and Larval Habitat Increase Aedes albopictus (Diptera: Culicidae) and Culex quinquefasciatus (Diptera: Culicidae) Abundance in Lowland Hawaii.

Project description:Vector abundance plays a key role in transmission of mosquito-borne disease. In Hawaii, Aedes albopictus (Skuse) (Diptera: Culicidae), the Asian tiger mosquito, has been implicated in locally-transmitted dengue outbreaks, while Culex quinquefasciatus Say (Diptera: Culicidae), the southern house mosquito, is the primary vector of avian malaria, a wildlife disease that has contributed to declines and extinctions of native Hawaiian birds. Despite the importance of these introduced species to human and wildlife health, little is known about the local-scale drivers that shape mosquito abundance across lowland Hawaii, where forest, agricultural, and residential land uses are prevalent. We examined landscape, larval habitat, and climate drivers of Ae. albopictus and Cx. quinquefasciatus abundance in eight lowland wet forest fragments on the Big Island of Hawaii. We found that the abundance of both species increased with the proportion of surrounding developed land and the availability of larval habitat, which were themselves correlated. Our findings suggest that conversion of natural habitats to residential and agricultural land increases mosquito larval habitats, increasing the abundance of Ae. albopictus and Cx. quinquefasciatus and increasing disease risk to humans and wildlife in Hawaii. Our results further indicate that while source reduction of artificial larval habitats-particularly moderately-sized human-made habitats including abandoned cars and tires-could reduce mosquito abundance, eliminating larval habitat will be challenging because both species utilize both natural and human-made larval habitats in lowland Hawaii.

Microhabitat partitioning of Aedes simpsoni (Diptera: Culicidae).

Project description:Yellow fever virus is a reemerging infection responsible for widespread, sporadic outbreaks across Africa. Although Aedes aegypti (L.) is the most important vector globally, in East Africa, epidemics may be vectored by Aedes bromeliae (Theobald), a member of the Aedes simpsoni (Theobald) species complex. The Ae. simpsoni complex contains 10 subspecies, of which Ae. bromeliae alone has been incriminated as a vector of yellow fever virus. However, morphological markers cannot distinguish Ae. bromeliae from conspecifics, including the sympatric and non-anthropophilic Aedes lilii (Theobald). Here, we used three sequenced nuclear markers to examine the population structure of Ae. simpsoni complex mosquitoes collected from diverse habitats in Rabai, Kenya. Gene trees consistently show strong support for the existence of two clades in Rabai, with segregation by habitat. Domestic mosquitoes segregate separately from forest-collected mosquitoes, providing evidence of habitat partitioning on a small spatial scale (< 5 km). Although speculative, these likely represent what have been described as Ae. bromeliae and Ae. lilii, respectively. The observation of high levels of diversity within Rabai indicates that this species complex may exhibit significant genetic differentiation across East Africa. The genetic structure, ecology, and range of this important disease vector are surprisingly understudied and need to be further characterized.

Online database for mosquito (Diptera, Culicidae) occurrence records in French Guiana.

Project description:A database providing information on mosquito specimens (Arthropoda: Diptera: Culicidae) collected in French Guiana is presented. Field collections were initiated in 2013 under the auspices of the CEnter for the study of Biodiversity in Amazonia (CEBA: http://www.labexceba.fr/en/). This study is part of an ongoing process aiming to understand the distribution of mosquitoes, including vector species, across French Guiana. Occurrences are recorded after each collecting trip in a database managed by the laboratory Evolution et Diversité Biologique (EDB), Toulouse, France. The dataset is updated monthly and is available online. Voucher specimens and their associated DNA are stored at the laboratory Ecologie des Forêts de Guyane (Ecofog), Kourou, French Guiana.
