Positioning Accuracy in Holographic Optical Traps.

ABSTRACT: Spatial light modulators (SLMs) have been widely used to achieve dynamic control of optical traps. Often, holographic optical tweezers have been presumed to provide nanometer or sub-nanometer positioning accuracy. It is known that some features concerning the digitalized structure of SLMs cause a loss in steering efficiency of the optical trap, but their effect on trap positioning accuracy has been scarcely analyzed. On the one hand, the SLM look-up-table, which we found to depend on laser power, produces positioning deviations when the trap is moved at the micron scale. On the other hand, phase quantization, which makes linear phase gratings become phase staircase profiles, leads to unexpected local errors in the steering angle. We have tracked optically trapped microspheres with sub-nanometer accuracy to study the effects on trap positioning, which can be as high as 2 nm in certain cases. We have also implemented a correction strategy that enabled the reduction of errors down to 0.3 nm.

Positioning Accuracy in Otosurgery Measured with Optical Tracking.

Project description:ObjectivesTo assess positioning accuracy in otosurgery and to test the impact of the two-handed instrument holding technique and the instrument support technique on surgical precision. To test an otologic training model with optical tracking.Study designIn total, 14 ENT surgeons in the same department with different levels of surgical experience performed static and dynamic tasks with otologic microinstruments under simulated otosurgical conditions.MethodsTip motion of the microinstrument was registered in three dimensions by optical tracking during 10 different tasks simulating surgical steps such as prosthesis crimping and dissection of the middle ear using formalin-fixed temporal bone. Instrument marker trajectories were compared within groups of experienced and less experienced surgeons performing uncompensated or compensated exercises.ResultsExperienced surgeons have significantly better positioning accuracy than novice ear surgeons in terms of mean displacement values of marker trajectories. The instrument support and the two-handed instrument holding techniques significantly reduce surgeons' tremor. The laboratory set-up presented in this study provides precise feedback for otosurgeons about their surgical skills and proved to be a useful device for otosurgical training.ConclusionsSimple tremor compensation techniques may offer trainees the potential to improve their positioning accuracy to the level of more experienced surgeons. Training in an experimental otologic environment with optical tracking may aid acquisition of technical skills in middle ear surgery and potentially shorten the learning curve. Thus, simulated exercises of surgical steps should be integrated into the training of otosurgeons.

A vector holographic optical trap.

Project description:The invention of optical tweezers almost forty years ago has triggered applications spanning multiple disciplines and has also found its way into commercial products. A major breakthrough came with the invention of holographic optical tweezers (HOTs), allowing simultaneous manipulation of many particles, traditionally done with arrays of scalar beams. Here we demonstrate a vector HOT with arrays of digitally controlled Higher-Order Poincaré Sphere (HOPS) beams. We employ a simple set-up using a spatial light modulator and show that each beam in the array can be manipulated independently and set to an arbitrary HOPS state, including replicating traditional scalar beam HOTs. We demonstrate trapping and tweezing with customized arrays of HOPS beams comprising scalar orbital angular momentum and cylindrical vector beams, including radially and azimuthally polarized beams simultaneously in the same trap. Our approach is general enough to be easily extended to arbitrary vector beams, could be implemented with fast refresh rates and will be of interest to the structured light and optical manipulation communities alike.

Constructing 3D microtubule networks using holographic optical trapping.

Project description:Developing abilities to assemble nanoscale structures is a major scientific and engineering challenge. We report a technique which allows precise positioning and manipulation of individual rigid filaments, enabling construction of custom-designed 3D filament networks. This approach uses holographic optical trapping (HOT) for nano-positioning and microtubules (MTs) as network building blocks. MTs are desirable engineering components due to their high aspect ratio, rigidity, and their ability to serve as substrate for directed nano-transport, reflecting their roles in the eukaryotic cytoskeleton. The 3D architecture of MT cytoskeleton is a significant component of its function, however experimental tools to study the roles of this geometric complexity in a controlled environment have been lacking. We demonstrate the broad capabilities of our system by building a self-supporting 3D MT-based nanostructure and by conducting a MT-based transport experiment on a dynamically adjustable 3D MT intersection. Our methodology not only will advance studies of cytoskeletal networks (and associated processes such as MT-based transport) but will also likely find use in engineering nanostructures and devices.

Nanoscopy of bacterial cells immobilized by holographic optical tweezers.

Project description:Imaging non-adherent cells by super-resolution far-field fluorescence microscopy is currently not possible because of their rapid movement while in suspension. Holographic optical tweezers (HOTs) enable the ability to freely control the number and position of optical traps, thus facilitating the unrestricted manipulation of cells in a volume around the focal plane. Here we show that immobilizing non-adherent cells by optical tweezers is sufficient to achieve optical resolution well below the diffraction limit using localization microscopy. Individual cells can be oriented arbitrarily but preferably either horizontally or vertically relative to the microscope's image plane, enabling access to sample sections that are impossible to achieve with conventional sample preparation and immobilization. This opens up new opportunities to super-resolve the nanoscale organization of chromosomal DNA in individual bacterial cells.

Precision assembly of complex cellular microenvironments using holographic optical tweezers.

Project description:The accurate study of cellular microenvironments is limited by the lack of technologies that can manipulate cells in 3D at a sufficiently small length scale. The ability to build and manipulate multicellular microscopic structures will facilitate a more detailed understanding of cellular function in fields such as developmental and stem cell biology. We present a holographic optical tweezers based technology to accurately generate bespoke cellular micro-architectures. Using embryonic stem cells, 3D structures of varying geometries were created and stabilized using hydrogels and cell-cell adhesion methods. Control of chemical microenvironments was achieved by the temporal release of specific factors from polymer microparticles positioned within these constructs. Complex co-culture micro-environmental analogues were also generated to reproduce structures found within adult stem cell niches. The application of holographic optical tweezers-based micromanipulation will enable novel insights into biological microenvironments by allowing researchers to form complex architectures with sub-micron precision of cells, matrices and molecules.

Holographic optical field recovery using a regularized untrained deep decoder network.

Project description:Image reconstruction using minimal measured information has been a long-standing open problem in many computational imaging approaches, in particular in-line holography. Many solutions are devised based on compressive sensing (CS) techniques with handcrafted image priors or supervised deep neural networks (DNN). However, the limited performance of CS methods due to lack of information about the image priors and the requirement of an enormous amount of per-sample-type training resources for DNNs has posed new challenges over the primary problem. In this study, we propose a single-shot lensless in-line holographic reconstruction method using an untrained deep neural network which is incorporated with a physical image formation algorithm. We demonstrate that by modifying a deep decoder network with simple regularizers, a Gabor hologram can be inversely reconstructed via a minimization process that is constrained by a deep image prior. The outcoming model allows to accurately recover the phase and amplitude images without any training dataset, excess measurements, or specific assumptions about the object's or the measurement's characteristics.

Holographic sol-gel monoliths: optical properties and application for humidity sensing.

Project description:Sol-gel monoliths based on SiO2, TiO2 and ZrO2 with holographic colourful diffraction on their surfaces were obtained via a sol-gel synthesis and soft lithography combined method. The production was carried out without any additional equipment at near room temperature and atmospheric pressure. The accurately replicated wavy structure with nanoscale size of material particles yields holographic effect and its visibility strongly depends on refractive index (RI) of materials. Addition of multi-walled carbon nanotubes (MWCNTs) in systems increases their RI and lends absorbing properties due to extremely high light absorption constant. Further prospective and intriguing applications based on the most successful samples, MWCNTs-doped titania, were investigated as reversible optical humidity sensor. Owing to such property as reversible resuspension of TiO2 nanoparticles while interacting with water, it was proved that holographic xerogels can repeatedly act as humidity sensors. Materials which can be applied as humidity sensors in dependence on holographic response were discovered for the first time.

Fokker-Planck analysis of optical near-field traps.

Project description:The motion of a nanoparticle in the vicinity of a near-field optical trap is modeled using the Fokker-Planck equation. A plasmonic C-shaped engraving on a gold film is considered as the optical trap. The time evolution of the position probability density of the nanoparticle is calculated to analyze the trapping dynamics. A spatially varying diffusion tensor is used in the formulation to take into account the hydrodynamic interactions. The steady-state position distribution obtained from the Fokker-Planck equation is compared with experimental results and found to be in good agreement. Computational cost of the proposed method is compared with the conventionally used Langevin equation based approach. The proposed method is found to be computationally efficient (requiring 35 times less computation time) and scalable to more complex lab-on-a-chip systems.

Prolonged treatment time deteriorates positioning accuracy for stereotactic radiosurgery.

Project description:IntroductionThe accuracy of radiation delivery is increasingly important as radiotherapy technology continues to develop. The goal of this study was to evaluate intrafractional motion during intracranial radiosurgery and the relationship between motion change and treatment time.Methods and materialsA total of 50 treatment records with 5988 images, all acquired during treatments with the CyberKnife Radiosurgery System, were retrospectively analyzed in this study. We measured translation and rotation motion including superior-inferior (SI), right-left (RL), anterior-posterior (AP), roll, tilt and yaw. All of the data was obtained during the first 45 minutes of treatment. The records were divided into 3 groups based on 15-min time intervals following the beginning of treatment: group A (0-15 min), group B (16-30 min) and group C (31-45 min). The mean deviations, systematic errors, random errors and margin for planning target volume (PTV) were calculated for each group.ResultsThe mean deviations were less than 0.1 mm in all three translation directions in the first 15 minutes. Greater motion occurred with longer treatment times, especially in the SI direction. For the 3D vector, a time-dependent change was observed, from 0.34 mm to 0.77 mm (p=0.01). There was no significant correlation between the treatment time and deviations in the AP, LR and rotation axes. Longer treatment times were associated with increases in systematic error, but not in random error. The estimated PTV margin for groups A, B and C were 0.86 / 1.14 / 1.31 mm, 0.75 / 1.12 / 1.20 mm, and 0.43 / 0.54 / 0.81 mm in the SI, RL, and AP directions, respectively.ConclusionsDuring intracranial radiosurgery, a consistent increase in the positioning deviation over time was observed, especially in the SI direction. If treatment time is greater than 15 minutes, we recommend increasing the PTV margins to ensure treatment precision.

Clinical Accuracy of Holographic Navigation Using Point-Based Registration on Augmented-Reality Glasses.

Project description:BackgroundAs current augmented-reality (AR) smart glasses are self-contained, powerful computers that project 3-dimensional holograms that can maintain their position in physical space, they could theoretically be used as a low-cost, stand-alone neuronavigation system.ObjectiveTo determine feasibility and accuracy of holographic neuronavigation (HN) using AR smart glasses.MethodsWe programmed a fully functioning neuronavigation system on commercially available smart glasses (HoloLens®, Microsoft, Redmond, Washington) and tested its accuracy and feasibility in the operating room. The fiducial registration error (FRE) was measured for both HN and conventional neuronavigation (CN) (Brainlab, Munich, Germany) by using point-based registration on a plastic head model. Subsequently, we measured HN and CN FRE on 3 patients.ResultsA stereoscopic view of the holograms was successfully achieved in all experiments. In plastic head measurements, the mean HN FRE was 7.2 ± 1.8 mm compared to the mean CN FRE of 1.9 ± 0.45 (mean difference: -5.3 mm; 95% confidence interval [CI]: -6.7 to -3.9). In the 3 patients, the mean HN FRE was 4.4 ± 2.5 mm compared to the mean CN FRE of 3.6 ± 0.5 (mean difference: -0.8 mm; 95% CI: -3.0 to 4.6).ConclusionOwing to the potential benefits and promising results, we believe that HN could eventually find application in operating rooms. However, several improvements will have to be made before the device can be used in clinical practice.
