Triterpenoids from Vitellaria paradoxa Stem Barks Reduce Nitrite Levels in LPS-Stimulated Macrophages.

ABSTRACT: Vitellaria paradoxa C. F. Gaertn is widely used in African traditional medicine as an anti-inflammatory remedy to treat rheumatism, gastric problems, diarrhea, and dysentery. The phytochemical investigation of the ethyl acetate extract of V. paradoxa stem bark collected in Burkina Faso led to the isolation of eight known and two triterpenes undescribed to date (7 and 10), in the free alcohol form or as acetyl and cinnamyl ester derivatives. The stereostructures of the new compounds were elucidated using HR-ESIMS and 1D and 2D NMR data. The isolated compounds were evaluated in vitro for their inhibitory effect on nitrite levels on murine macrophages J774 stimulated with the lipopolysaccharide (LPS). Among all the compounds tested, lupeol cinnamate (3) and betulinic acid (5) showed a beneficial effect in reducing nitrite levels produced after LPS stimulation.

Project description:A madecassoside-rich fraction obtained from the industrial purification of Centella asiatica leaves afforded a new triterpene glycoside, named isomadecassoside (4), characterized by an ursane-type skeleton and migration of the double bond at Δ20(21) in ring E. The structure of isomadecassoside was established by means of HR-ESIMS and detailed analysis of 1D and 2D NMR spectra, which allowed a complete NMR assignment. Studies on isolated J774A.1 macrophages stimulated by LPS revealed that isomadecassoside (4) inhibited nitrite production at non-cytotoxic concentrations, thus indicating an anti-inflammatory effect similar to that of madecassoside.

Influence of Shea tree (Vitellaria paradoxa) on maize and soybean production.

Project description:Vitellaria paradoxa provides many benefits to farmers within the Shea belt. However, increased threats to it necessitate its conservation, and one common approach is the practice of agroforestry. A number of studies have shown that Shea tree has influence on crop production, and yet, some of these studies were done using single season experiments or bioassays using mature Shea tree components. In this study, the seasonal influence of young and mature Shea trees on Maize and Soybean yields was investigated using field experiments in Otuke district of northern Uganda, where, Shea tree parklands are dominant and Maize and Soybean are used for food security and income. Our results show that there are differential responses of maize and soybean yield to rainy seasons and physiological variations of Vitellaria paradoxa with age. We find yield reduction for maize more pronounced than yield reduction for soybeans under different Shea age (Mature and Young) for two rainy seasons. We attribute the variance to the differential maize and soybean responses to Vitellaria paradoxa shading and its differential allelopathic inhibition of these crops. We recommend that Soybeans should be preferred to maize when planting under Shea canopy.

Past climate changes explain the phylogeography of Vitellaria paradoxa over Africa.

Project description:The evolution of the savanna biome has been deeply marked by repeated contraction/expansion phases due to climate perturbations during the Quaternary period. In this study, we investigated the impact of the last glacial maximum (LGM) on the present genetic pattern of Vitellaria paradoxa (shea tree), a major African savanna tree. A range-wide sampling of the species enabled us to sample 374 individuals from 71 populations distributed throughout sub-Sahelian Africa. Trees were genotyped using 3 chloroplasts and 12 nuclear microsatellites, and were sequenced for 2 polymorphic chloroplast intergenic spacers. Analyses of genetic diversity and structure were based on frequency-based and Bayesian methods. Potential distributions of V. paradoxa at present, during the LGM and the last interglacial period, were examined using DIVA-GIS ecological niche modelling (ENM). Haplotypic and allelic richness varied significantly across the range according to chloroplast and nuclear microsatellites, which pointed to higher diversity in West Africa. A high but contrasted level of differentiation was revealed among populations with a clear phylogeographic signal, with both nuclear (F(ST) = 0.21; R(ST) = 0.28; R(ST) > R(ST) (permuted)) and chloroplast simple sequence repeats (SSRs) (G(ST) = 0.81; N(ST) = 0.90; N(ST) > N(ST) (permuted)). We identified a strong geographically related structure separating western and eastern populations, and a substructure in the eastern part of the area consistent with subspecies distinction. Using ENM, we deduced that perturbations during the LGM fragmented the potential eastern distribution of shea tree, but not its distribution in West Africa. Our main results suggest that climate variations are the major factor explaining the genetic pattern of V. paradoxa.

In vitro trypanocidal activity of extracts and compounds isolated from Vitellaria paradoxa.

Project description:BackgroundVitellaria paradoxa is used in traditional medicine for the treatment of various diseases in tropical countries; however, nothing is known about its anti-trypanosomal activity. Human African trypanosomiasis is a neglected tropical disease of Sub-Saharan Africa's poorest rural regions, and the efficacy of its treatment remains a challenge. This study investigates the as-yet-unknown trypanocidal activity of this plant.MethodsV. paradoxa, commonly known as shea tree, was selected for study based on an ethnobotanical investigation. Ultrasonicated extracts from bark and seeds were successively treated with ethyl acetate and water. Column chromatography, NMR spectroscopy and mass spectrometry were used to identify isolated compounds. Purified trypanosomes (Trypanosoma brucei brucei) were incubated with serial dilutions of the extracts and isolated compounds at 37 °C in 5% CO2 for 24 h. Parasite viability was evaluated under a microscope.ResultsThe ethyl acetate extracts of the bark showed the higher in vitro trypanocidal activity against T. brucei brucei with median inhibitory concentration (IC50) of 3.25 µg/mL. However, the triterpene 1α,2β,3β,19α-tretrahydroxyurs-12-en-28-oic acid and the pentadecanoic acid isolated from the ethyl acetate extract of the seeds showed in vitro trypanocidal activity with IC50 of 11.30 and 70.1 µM, respectively.ConclusionThe results obtained contribute to the validation of the traditional medicinal use of V. paradoxa. Our results encourage further investigations of this plant, mainly with respect to its in vivo efficacy and toxicity.

Anti-Inflammatory Triterpenoids from the Caulophyllum robustum Maximin LPS-Stimulated RAW264.7 Cells.

Project description:Caulophyllum robustum Maxim is widely distributed in China and used as a traditional herbal medicine to induce childbirth, ease the pain of labor, rectify delayed or irregular menstruation, alleviate heavy bleeding and pain during menstruation, and treat external injuries and irregular menses. According to our detailed chemical investigation, three new triterpene derivatives (1⁻3), together with seven known compounds, were isolated from the root and rhizome of C. robustum Maxim. Their structures were elucidated by 1D- and 2D-NMR spectroscopic analysis and physio-chemical methods. They were identified as (1) 23-hydroxy-3,19-dioxo-olean-12-en-28-oic-acid; (2) 23-hydroxy-3,11-dioxo-olean-12-en-28-oic acid; and (3) 16α,23-dihydroxy-3-oxo-olean-12-en-28-oic acid. Compounds (1⁻10) inhibited the LPS-activated NO production in RAW264.7 cells. Furthermore, the anti-inflammatory characteristics of these compounds were confirmed on the basis of decreases in iNOS and NF-κB protein expression in RAW264.7 cells.

Glucose controls morphodynamics of LPS-stimulated macrophages.

Project description:Macrophages constantly undergo morphological changes when quiescently surveying the tissue milieu for signs of microbial infection or damage, or after activation when they are phagocytosing cellular debris or foreign material. These morphofunctional alterations require active actin cytoskeleton remodeling and metabolic adaptation. Here we analyzed RAW 264.7 and Maf-DKO macrophages as models to study whether there is a specific association between aspects of carbohydrate metabolism and actin-based processes in LPS-stimulated macrophages. We demonstrate that the capacity to undergo LPS-induced cell shape changes and to phagocytose complement-opsonized zymosan (COZ) particles does not depend on oxidative phosphorylation activity but is fueled by glycolysis. Different macrophage activities like spreading, formation of cell protrusions, as well as phagocytosis of COZ, were thereby strongly reliant on the presence of low levels of extracellular glucose. Since global ATP production was not affected by rewiring of glucose catabolism and inhibition of glycolysis by 2-deoxy-D-glucose and glucose deprivation had differential effects, our observations suggest a non-metabolic role for glucose in actin cytoskeletal remodeling in macrophages, e.g. via posttranslational modification of receptors or signaling molecules, or other effects on the machinery that drives actin cytoskeletal changes. Our findings impute a decisive role for the nutrient state of the tissue microenvironment in macrophage morphodynamics.

Genome-wide association study of fat content and fatty acid composition of shea tree (Vitellaria paradoxa C.F. Gaertn subsp. paradoxa).

Project description:BackgroundFat content (FC) and fatty acids (FA) are the most important traits in shea tree breeding, controlled by several genes with relatively small effects. Therefore, determining the genes involved in the biosynthesis of such traits is crucial for improving oil quantity and quality and for the domestication process of the species. To identify the quantitative trait nucleotides (QTNs) controlling FC and FA, we conducted a multi-locus genome-wide association study (GWAS) using six multi-locus GWAS methods for FC and FA in 122 superior shea trees (SSTs). SSTs were genotyped using DArTseq, resulting in 7,559 non-redundant single nucleotide polymorphism markers.ResultsFat content varied from 36 to 58% with a mean of 50%. Fatty acid composition was 51.26 ± 4.21, 38.76 ± 4.67, 6.45 ± 0.76 and 3.53 ± 0.52% for oleic, stearic, linoleic and palmitic acids, respectively. A very high negative correlation coefficient (-0.98) was found between stearic and oleic acids. A total of 47 significant QTNs associated with fat-related traits were detected by the GWAS methods. Among these QTNs, 25 were identified as common QTNs based on their detection by multiple GWAS methods. Using the superior allele information of the 4 common QTNs associated with fat content in 17 high-fat and 21 low-fat SSTs, we found a higher percentage of superior alleles in SSTs with high FC (47.1%) than in SSTs with low FC (14.3%). Pathway analysis of the common QTNs identified 24 potential candidate genes likely involved in the biosynthesis of FC and FA composition in shea tree seeds.ConclusionsThese findings will contribute to the discovery of the polygenic networks controlling FC in shea tree, improve our understanding of the genetic basis and regulation of FC, and be useful for molecular breeding of high-fat shea tree cultivars.
