Synthesis of Chitosan/Diatomite Composite as an Advanced Delivery System for Ibuprofen Drug; Equilibrium Studies and the Release Profile.

ABSTRACT: Chitosan/diatomite nanocomposite (CS/D) was synthesized as a low-cost and highly porous structure of enhanced physicochemical properties to be applied as advanced carriers for ibuprofen drug (IB). The loading properties of CS/D were studied in comparison to diatomite as a separated phase and achieved a loading capacity of 562.6 mg/g. The loading reactions of IB into CS/D show pseudo-second-order kinetic behavior and Langmuir isotherm properties. This demonstrates homogeneous loading processes in monolayer forms and controlled essentially by physical mechanisms. This was confirmed by the calculated Gaussian energy (7.7 kJ/mol (D) and 7.9 kJ/mol (CS/D)) in addition to the thermodynamic parameters. The thermodynamic behavior for the IB loading process is related to spontaneous, favorable, and exothermic reactions. The CS/D composite is of promising IB release profile that extended to about 200 h with a maximum release of 91.5% at the gastric fluid (pH 1.2) and 97.3% in the intestinal fluid (pH 7.4). The IB release rate from CS/D can be controlled based on the ratio of the integrated chitosan in the composite. The IB release reactions from CS/D follow the assumption of Korsmeyer-Peppas kinetics with determined values for the diffusion exponent reflects complex diffusion and erosion as the affected mechanisms during the IB release process.

Project description:Intratumoral (IT) injection of chemotherapeutics into needle-accessible solid tumors can directly localize the anticancer drug in the tumor site, thus increasing its local bioavailability and reducing its undesirable effects compared to systemic administration. In this study, graphene oxide (GO)-based chitosan/β-glycerophosphate (CS/GP) thermosensitive injectable composite hydrogels (CH) were prepared and optimized for the localized controlled delivery of doxorubicin (DOX). A quality-by-design (QbD) approach was used to study the individual and combined effects of several formulation variables to produce optimal DOX-loaded GO/CS/GP CH with predetermined characteristics, including gelation time, injectability, porosity, and swelling capacity. The surface morphology of the optimal formulation (DOX/opt CH), chemical interaction between its ingredients and in vitro release of DOX in comparison to GO-free CS/GP CH were investigated. Cell viability and cellular uptake after treatment with DOX/opt CH were studied on MCF 7, MDB-MB-231 and FaDu cell lines. The statistical analysis of the measured responses revealed significant effects of the concentration of GO, the concentration of CS, and the CS:GP ratio on the physicochemical characteristics of the prepared GO/CS/GP CH. The optimization process showed that DOX-loaded GO/CS/GP CH prepared using 0.1% GO and 1.7% CS at a CS: GO ratio of 3:1 (v/v) had the highest desirability value. DOX/opt CH showed a porous microstructure and chemical compatibility between its ingredients. The incorporation of GO resulted in an increase in the ability of the CH matrices to control DOX release in vitro. Finally, cellular characterization showed a time-dependent increase in cytotoxicity and cellular uptake of DOX after treatment with DOX/opt CH. The proposed DOX/opt CH might be considered a promising injectable platform to control the release and increase the local bioavailability of chemotherapeutics in the treatment of solid tumors.

Project description:The present study focused on investigation of graphene oxide/hydroxyapatite (GO/HAp) and amine modified graphene oxide/hydroxyapatite (GO-NH2/HAp) composites as potential drug carrier agents for 5-Fluorouracil (5-FU). Incorporation of 5-Fluorouracil drug was performed via adsorption through π-π interactions and electrostatic attractions. Modification of graphene oxide was performed for the production of amine modified graphene oxide/hydroxyapatite composite with the intention of enhancing adsorption performance. The X-Ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), Thermogravimetric Analysis (TGA) and zeta potential/particle size analysis were performed for particle characterization while Scanning Electron Microscopy (SEM) and Transmission Electron Microscopy (TEM) analysis were used to analyze detailed morphological properties. Experimental design studies were followed out in order to determine the effect of adsorption parameters including graphene oxide amount, pH and initial drug concentration on 5-Fluorouracil adsorption behavior. Adsorption isotherms of both composites with unmodified and modified GO were best fitted to Freundlich model with R2 values of 0.9616 and 0.9682 respectively. The maximum adsorption capacities (qm) were calculated as 47.3 mg/g and 18.4 for graphene oxide/hydroxyapatite and amine modified graphene oxide/hydroxyapatite composites respectively at pH 2.0. The highest adsorption percentage was obtained for amine modified graphene oxide/hydroxyapatite composite as 40.87 % at pH 2.0 condition. In vitro release kinetic studies revealed that compliance with Higuchi and Korsmeyer-Peppas kinetic models were observed for graphene oxide/hydroxyapatite, whereas zero order and Korsmeyer-Peppas kinetic models pointed out as the well-fitted model for amine modified graphene oxide/hydroxyapatite composite. The release period of 5-FU drug from all composites were continued up to 8-10 h in physiological conditions (pH 7.4, 37 °C) indicating an achieved controlled release. Based on the overall findings, graphene oxide/hydroxyapatite and amine modified graphene oxide/hydroxyapatite composites could be suggested as a potential drug delivery agent for 5-FU in clinical applications.
