Evidence for deficits in behavioural and physiological responses in aged mice relevant to the psychiatric symptom of apathy.

ABSTRACT: Apathy is widely reported in patients with neurological disorders or post viral infection but is also seen in otherwise-healthy aged individuals. This study investigated whether aged male mice express behavioural and physiological changes relevant to an apathy phenotype. Using measures of motivation to work for reward, we found deficits in the progressive ratio task related to rate of responding. In an effort-related decision-making task, aged mice were less willing to exert effort for high value reward. Aged mice exhibited reduced reward sensitivity but also lower measures of anxiety in the novelty supressed feeding test and an attenuated response to restraint stress with lower corticosterone and reduced paraventricular nucleus c-fos activation. This profile of affective changes did not align with those observed in models of depression but suggested emotional blunting. In a test of cognition (novel object recognition), aged mice showed no impairments, but activity was lower in a measure of exploration in a novel environment. Together, these data suggest aged mice show changes across the domains of motivated behaviour, reward sensitivity and emotional reactivity and may be a suitable model for the pre-clinical study of the psychiatric symptom of apathy.

Project description:Automated monitoring of behaviour can offer a wealth of information in circumstances where observing behaviour is difficult or time consuming. However, this often requires attaching monitoring devices to the animal which can alter behaviour, potentially invalidating any data collected. Birds often show increased preening and energy expenditure when wearing devices and, especially in laying hens, there is a risk that individuals wearing devices will attract aggression from conspecifics. We studied the behavioural and physiological response of 20 laying hens to backpacks containing monitoring devices fastened with elastic loops around the wing base. We hypothesised that backpacks would lead to a stress-induced decrease in peripheral temperature, increased preening, more aggression from conspecifics, and reduced bodyweights. This was evaluated by thermography of the eye and comb (when isolated after fitting backpacks), direct observations of behaviour (when isolated, when placed back into the group, and on later days), and weighing (before and after each 7-day experimental period). Each hen wore a backpack during one of the two experimental periods only and was used as her own control. Contrary to our hypothesis, eye temperature was higher when hens wore a backpack (No backpack: 30.2 °C (IQR: 29.0-30.6) vs. Backpack: 30.9 °C (IQR: 30.0-32.0), P < 0.001). Eye temperature of hens wearing a backpack was strongly correlated to the time spent preening (rs = 0.8, P < 0.001), suggesting that the higher temperatures may have been due to preening itself, or to a low head position or decreased heat dissipation when preening under the wings. Aggressive behaviour was very rare and no effect of the backpacks was found. In line with our hypothesis, backpacks increased preening on the day of fitting, both when isolated (No backpack: 0% (IQR: 0-1) vs. Backpack: 22% (IQR: 1-43), P < 0.01) and when back in the group (No backpack: 0% (IQR: 0-27) vs. Backpack: 43% (IQR: 5-77), P < 0.001). However, no effect on preening was observed 2-7 days afterwards. Other behavioural changes suggested that on the day of fitting hens prioritized attempts to (re)move the backpack and were less attentive to their surroundings. However, only equipment pecking (i.e., pecking the backpack or leg rings) was still affected 2-7 days after fitting (No backpack: 0 pecks/hen/minute (IQR: 0-0), vs. Backpack: 0 (IQR: 0-0.07), P < 0.05). We found no effect of our backpacks on bodyweight. In conclusion, our backpacks seem suitable to attach monitoring equipment to hens with only a very minor effect on their behaviour after a short acclimation period (≤2 days).

Project description:The regulations for minimal space and direction of travel for land transport in horses vary worldwide and there is currently no definitive guidance to promote equine health and welfare. This study evaluated the effects of bay size and direction of travel (forwards/backwards) in horses by comparing the behavioural, physiological, laboratory and gastroscopy parameters between transported and confined horses. A total of twenty-six mares took part in the study; 12 horses were confined for 12 h, and all mares underwent 12 hours' transportation, travelling in single (n = 18) or wide bays (n = 8), and forward (n = 10) or rear (n = 16) facing. Behaviour was recorded during confinement/transportation and analysed using a behaviour sampling ethogram. Clinical examination, blood samples and gastroscopy were conducted before and after confinement/transportation. The frequency of behaviours relating to stress and balance increased during transport, and horses transported in a rear-facing position and in a wider bay size showed fewer balance-related behaviours. Balance behaviours, particularly loss of balance, were positively associated with the severity of gastric ulceration after transportation and elevated muscle enzymes, while increased stress behaviours correlated with decreased gastrointestinal sounds. Heart rate and rectal temperature after transportation were positively associated with balance and stress behaviours, and with squamous gastric ulcer scores. Transportation was associated with expected increases in cortisol and muscle enzymes, but positioning and space allowance had minimal effects on these analytes. Findings suggest that transportation in a rear-facing position and in wider bays might reduce the impact of transport on horse health and welfare, and monitoring behaviour in transit and physiological measurements after transportation should be recommended. Behavioural and physiological parameters were more sensitive than haematological, biochemical or endocrine analytes to identify horses suffering from transport stress.

Project description:Sound sensitive dogs have exaggerated responses to sound stimuli that can negatively impact the welfare of the dog. Behavioural reactions combined with the response to sound involve a marked autonomic imbalance towards sympathetic predominance and release of cortisol. The purpose of the present study was to evaluate, in the laboratory, the cardiac autonomic modulation using heart rate variability (HRV) analysis, serum cortisol levels and behavioural parameters in response to sounds of fireworks in dogs with a history of sensitivity to fireworks. Based on these data, and combining qualitative measures and categorical measures, we propose one short and one full index of sound sensitivity in dogs. Six privately owned dogs with no history and another twelve dogs with a history of sound sensitivity to fireworks were used. The sound stimulus consisted of a standardised recording of fireworks (180-seconds long) with a peak intensity of 103-104 dB. The cardiac intervals were recorded using a frequency meter (Polar® RS800CX model) to evaluate the HRV, and the acquired data were processed using CardioSeries 2.4.1 software. Twenty-one behavioural parameters were analysed quantitatively by time, frequency or categorically by scores and were grouped in behavioural categories of arousal, fear, relaxation and "other". Sound sensitive dogs had exacerbated autonomic responses to the sound stimulus in the laboratory compared to non-sensitive dogs, with higher LF/HF ratios suggesting autonomic imbalance towards sympathetic predominance, but the cortisol levels were similar between the sensitive and non-sensitive dogs. Sound sensitive dogs showed pronounced responses for the parameters: alert and attention, search sound, startle, trembling, hiding, run away and less intense responses for the parameters rest and wink/sleep. Furthermore, the behavioural categories of arousal, fear, relaxation (lack of) and LF/HF were correlated to the caregiver's perception of the sound sensitivity of the dogs. Not only the short index for sound sensitivity (behavioural categories arousal, fear and relaxation, and LF/HF ratio) but also the full index for sound sensitivity (all behavioural categories, LF/HF and cortisol levels) was highly correlated to sound fear response at home. These indexes can contribute to the development of strategies to treat sound sensitive dogs.

Project description:BackgroundA diagnosis of dementia in middle-aged and elder people is often complicated by physical frailty and comorbid neuropsychiatric symptoms (NPSs). Previous studies have identified NPSs as a risk factor for dementia. The aim of this study was to figure out to what extent individual NPS and certain demographic factors increased the risk of dementia in middle-aged and senior psychiatric inpatients.MethodsOne hundred twenty-seven middle-aged and senior patients admitted to psychiatric wards for late-onset (age ≥ 50 years) psychiatric symptoms were included and categorized into dementia or non-demented psychiatric disorders (NDPD). The patients' demographic information and medical records were collected during the first hospitalization and subjected to statistical analyses.Results41.73% of the registered psychiatric inpatients were diagnosed as dementia in which Alzheimer's disease (AD) was the dominant subtype. The NDPD group consisted of nine individual diagnoses, except for schizophrenia. The frequencies of dementia inpatients increased with first episode age while that of NDPD inpatients decreased with first episode age. In the enrolled inpatients, most of dementia patients were males while females accounted for a higher proportion of NDPD patients. 58.49% of enrolled dementia inpatients presented cognitive deficit (CD) as the initial symptom while the remaining 41.51% showed NPS as initial symptom. Of the 12 NPSs, agitation and apathy greatly and significantly increased risk of dementia in psychiatric inpatients with late-onset psychiatric symptoms.ConclusionsThese results added evidence that the demented patients admitted to psychiatric ward are more likely to be male, older first episode age, and have characteristic NPS including aberrant motor behavior (AMB), hallucinations, agitation, irritability and apathy. Further, this study emphasized the importance of agitation and apathy of NPSs functioning as risk factors of dementia in these inpatients.
