Describing the hidden species diversity of Chaetozone (Annelida, Cirratulidae) in the Norwegian Sea using morphological and molecular diagnostics.

ABSTRACT: Using molecular markers and species delimitation analyses, a high diversity of bi-tentaculate Cirratulidae was discovered from the North-East Atlantic. Five new species are described: Chaetozone pseudosetosa sp. nov., Chaetozone quinta sp. nov., Chaetozone barentsensis sp. nov., Chaetozone monteverdii sp. nov., and Chaetozone chambersae sp. nov. Several morphogroups are also described, even though the presence of cryptic diversity prevented naming of individual species. For each species presented, a molecular diagnostic is given from the universal barcode COI and, when available, the D1-D2 domains of the 28S rRNA. This increases the number of species in Chaetozone in northern European waters from ten to at least 17 species, the exact number of species remaining uncertain as taxonomic issues with older names remain unresolved.

Timareteposteria, a new cirratulid species from Korea (Annelida, Polychaeta, Cirratulidae).

Project description:A new cirratulid species, Timareteposteria sp. n., is described from the intertidal habitats of the eastern coast of South Korea. The new species is closely related to Timareteluxuriosa (Moore, 1904) from southern California based on morphological similarity of the branchial and tentacular finalents and the noto- and neuropodial spines. However, T.posteria sp. n. differs from the latter based on the following characteristics: 1) evenly divided peristomium into three annulations; 2) 2-4 neuropodial spines originating in the posterior chaetigers alternated by a few capillaries; and 3) complete shift in branchial finalents located about one-third between the notopodium and the dorsal midline. The new species has a methyl green staining pattern (MGSP) distinct from other Timarete species. Detailed description and illustrations of the new species are provided with molecular information based on the partial sequences of the mitochondrial cytochrome c oxidase subunit I (COI) and 16S ribosomal RNA (16S). This study also includes a key and discussion of known Timarete species from East Asia.

Kirkegaardia Blake, 2016 (Annelida: Cirratulidae) from Southeastern Brazil with description of nine new species.

Project description:This is the first taxonomic study of cirratulid polychaetes of the genus Kirkegaardia Blake, 2016 from Brazil. Nine new species of the genus are described from the Southern Brazilian coast (50-3000 m deep). The genus Kirkegaardia is generally subdivided into three distinct groups of species (Kirkegaardia dorsobranchialis-heterochaeta, Kirkegaardia baptisteae-tesselata and Kirkegaardia luticastella) and several out-group species for which relationships remains to be defined. In this study, new species were included in the Kirkegaardia dorsobranchialis-heterochaeta and Kirkegaardia baptisteae-tesselata groups. Kirkegaardia dorsobranchialis-heterochaeta is characterized by thoracic parapodia elevated producing a channel between the notopodia, elongate pre-setigerous region that is either entirely smooth or modified with a dorsal ridge and/or rings, and noto- and neurosetae capillaries denticulated. As belonging to this group, K. blakei sp. nov., K. brisae sp. nov., K. goytaca sp. nov., K. jongo sp. nov. and K. papaveroi sp. nov. are described here. Kirkegaardia baptisteae-tesselata includes species that lack thoracic parapodia elevated and mid-dorsal thoracic groove, although a dorsal ridge is sometimes developed. In the pre-setigerous region dorsal ridges and rings are present or absent. Most species in this group have neurosetae denticulated, and notosetae capillaries of other types. This study adds K. helenae sp. nov., K. medusa sp. nov., K. nupem sp. nov. and K. zafirae sp. nov. to the latter species group. In addition, two new records are provided for K. hampsoni. A key to cirratulid polychaete species reported from Brazilian waters is provided.

Hidden diversity in the genus Tethya: comparing molecular and morphological techniques for species identification.

Project description:Correctly determining species' identity is critical for estimating biodiversity and effectively managing marine populations, but is difficult for species that have few morphological traits or are highly plastic. Sponges are considered a taxonomically difficult group because they lack multiple consistent diagnostic features, which coupled with their common phenotypic plasticity, makes the presence of species complexes likely, but difficult to detect. Here, we investigated the evolutionary relationship of Tethya spp. in central New Zealand using both molecular and morphological techniques to highlight the potential for cryptic speciation in sponges. Phylogenetic reconstructions based on two mitochondrial markers (rnl, COI-ext) and one nuclear marker (18S) revealed three genetic clades, with one clade representing Tethya bergquistae and two clades belonging to what was a priori thought to be a single species, Tethya burtoni. Eleven microsatellite markers were also used to further resolve the T. burtoni group, revealing a division consistent with the 18S and rnl data. Morphological analysis based on spicule characteristics allowed T. bergquistae to be distinguished from T. burtoni, but revealed no apparent differences between the T. burtoni clades. Here, we highlight hidden genetic diversity within T. burtoni, likely representing a group consisting of incipient species that have undergone speciation but have yet to express clear morphological differences. Our study supports the notion that cryptic speciation in sponges may go undetected and diversity underestimated when using only morphology-based taxonomy, which has broad scale implications for conservation and management of marine systems.

A new species of Fauveliopsidae (Annelida) from the North Sea.

Project description:A new species of the genus Laubieriopsis Petersen, 2000 is described based on 28 specimens collected in the north-east part of the North Sea. It is characterized by fixed number of chaetigers (22), paired genital papillae, bidentate neurochaeta of chaetigers 1-4, the absence of acicular chaetae on chaetigers 5-21 and, on the last chaetiger, one acicular and three capillary chaetae enlarged and directed backward. The present study brings the number of known species of Laubieriopsis to five and the number of Northeast Atlantic species of this genus to two.

Hidden diversity in Antarctica: Molecular and morphological evidence of two different species within one of the most conspicuous ascidian species.

Project description:The Southern Ocean is one of the most isolated marine ecosystems, characterized by high levels of endemism, diversity, and biomass. Ascidians are among the dominant groups in Antarctic benthic assemblages; thus, recording the evolutionary patterns of this group is crucial to improve our current understanding of the assembly of this polar ocean. We studied the genetic variation within Cnemidocarpa verrucosa sensu lato, one of the most widely distributed abundant and studied ascidian species in Antarctica. Using a mitochondrial and a nuclear gene (COI and 18S), the phylogeography of fifteen populations distributed along the West Antarctic Peninsula and Burdwood Bank/MPA Namuncurá (South American shelf) was characterized, where the distribution of the genetic distance suggested the existence of, at least, two species within nominal C. verrucosa. When reevaluating morphological traits to distinguish between genetically defined species, the presence of a basal disk in one of the genotypes could be a diagnostic morphological trait to differentiate the species. These results are surprising due to the large research that has been carried out with the conspicuous C. verrucosa with no differentiation between species. Furthermore, it provides important tools to distinguish species in the field and laboratory. But also, these results give new insights into patterns of differentiation between closely related species that are distributed in sympatry, where the permeability of species boundaries still needs to be well understood.

The multitentaculate Cirratulidae of the genera Cirriformia and Timarete (Annelida: Polychaeta) from shallow waters of Brazil.

Project description:A large number multitentaculate cirratulids have been described worldwide but most are only known through the original descriptions. Type material, voucher and recently collected specimens from Brazil were revisited in order to reveal their true identity and confirm the records of widely distributed species in this region. Six species are described, three of which are new, Cirriformia capixabensis sp. nov., Cirriformia chicoi sp. nov. and Timarete ceciliae sp. nov. COI and 16S sequences were obtained and used for inter-specific comparisons. Timarete caribous is reported from several localities along the Brazilian coast and a new synonym, Cirratulus melanacanthus, is proposed. The species Timarete oculata, originally described from Brazil and lumped into the Timarete filigera species complex, is herein revalidated and redescribed. The occurrence of the species Timarete filigera and Cirriformia tentaculata is not confirmed from the Brazilian coast. Descriptions, illustrations and a key to genera and species are provided.

A new species of Amphictene (Annelida, Pectinariidae) from the northern South China Sea.

Project description:Pectinariids are a family of polychaetes commonly found in shallow coastal waters around the world, but their diversity is poorly known along the coasts of Asia. Here we describe Amphictene alata sp. n. (Pectinariidae), based on 15 specimens collected from the coastal waters of Guangdong in the northern South China Sea. This new species can be distinguished from all other 13 described species and one described subspecies of Amphictene by having a pair of dorsolateral lobes on segment 3, a pair of large lateral lobes on segment 21, and more scaphal hooks (26 to 37 pairs).

Hidden diversity in sardines: genetic and morphological evidence for cryptic species in the goldstripe sardinella, Sardinella gibbosa (Bleeker, 1849).

Project description:Cryptic species continue to be uncovered in many fish taxa, posing challenges for fisheries conservation and management. In Sardinella gibbosa, previous investigations revealed subtle intra-species variations, resulting in numerous synonyms and a controversial taxonomy for this sardine. Here, we tested for cryptic diversity within S. gibbosa using genetic data from two mitochondrial and one nuclear gene regions of 248 individuals of S. gibbosa, collected from eight locations across the Philippine archipelago. Deep genetic divergence and subsequent clustering was consistent across both mitochondrial and nuclear markers. Clade distribution is geographically limited: Clade 1 is widely distributed in the central Philippines, while Clade 2 is limited to the northernmost sampling site. In addition, morphometric analyses revealed a unique head shape that characterized each genetic clade. Hence, both genetic and morphological evidence strongly suggests a hidden diversity within this common and commercially-important sardine.

Morphological and molecular evidence uncovers hidden species diversity in the leatherleaf slug genus Valiguna (Systellommatophora, Veronicellidae) from Thailand.

Project description:The poorly studied leatherleaf slug genus Valiguna in Thailand was carefully investigated. Members of this genus are phenotypically similar, making their identification very challenging. This study clarifies the taxonomic status of all Valiguna species in Thailand by combining morphological and anatomical studies with DNA barcoding. Monophyly of all Valiguna species was confirmed by analysis of the mitochondrial COI data and that all Valiguna species have the acropleurocaulis type of penis. Currently, three Valiguna species are recognised: V.siamensis, V.semicerina Mitchueachart & Panha, sp. nov., and V.crispa Mitchueachart & Panha, sp. nov. that are new to science. For distinct characteristics, V.siamensis is characterised by having a cylindrical penis and honeycomb-like glans, V.semicerina sp. nov. has a lanceolate penis with half honeycomb-like glans, and V.crispa sp. nov. has a cylindrical penis with wavy-like glans. In addition, more detailed descriptions of the radula and genitalia of all three species and their distribution are also carefully presented, enhancing the understanding of this leatherleaf slug genus in Thailand.
