Protein nanofibrils for next generation sustainable water purification.

ABSTRACT: Water scarcity is rapidly spreading across the planet, threatening the population across the five continents and calling for global sustainable solutions. Water reclamation is the most ecological approach for supplying clean drinking water. However, current water purification technologies are seldom sustainable, due to high-energy consumption and negative environmental footprint. Here, we review the cutting-edge technologies based on protein nanofibrils as water purification agents and we highlight the benefits of this green, efficient and affordable solution to alleviate the global water crisis. We discuss the different protein nanofibrils agents available and analyze them in terms of performance, range of applicability and sustainability. We underline the unique opportunity of designing protein nanofibrils for efficient water purification starting from food waste, as well as cattle, agricultural or dairy industry byproducts, allowing simultaneous environmental, economic and social benefits and we present a case analysis, including a detailed life cycle assessment, to establish their sustainable footprint against other common natural-based adsorbents, anticipating a bright future for this water purification approach.

Project description:Self-standing nanocomposite films were prepared by three-dimensional UV-induced radical copolymerization of methacrylated alginate (MALG) with acrylic acid (AA) and reinforced with graphene oxide (GO) to improve both mechanical strength and dye adsorption capacity in wastewater decontamination operations. Dynamic mechanical-thermal analysis revealed variations in storage modulus: the higher the GO content, the higher the storage modulus (E') values. Also, the higher the temperature (associated with a lower and lower water content of films), the larger values of E' for the films of the same composition (E'(25 °C) = 676.6-1538.7 MPa; E'(100 °C) = 886.9-2066.6 MPa), providing insights into the compatibility between GO and the MALG/AA matrix, as well as, assessing the improvement in the nanocomposite's final mechanical properties. These crosslinked films in a dry state exhibited rapid water uptake and relatively short drying times (ca. 30 min at room temperature for the MALG/AA/GO composites) resulting from the swelling-drying studies and water contact angle measurements. The efficacy of methylene blue removal from water assessed via UV-VIS spectrometry revealed excellent results, expressed as an adsorption yield of 70-80% and 85-98% after 30 h and 258 h, respectively, of immersion time of films into an MB aqueous solution of 12.5 mg/L (as the contaminated water model). The reusability of the same films was evaluated by consecutive extraction processes of MB from the composite membranes when the content of desorbed dye was also spectrophotometrically monitored and conducted in acidic conditions (HCl aqueous solutions of pH 2). Overall, the introduction of GO in the developed self-standing MALG/AA nanocomposite films exhibited enhanced mechanical properties and increased efficiency for dye removal applications. Their great reutilization potential was highlighted by low drying times and a good ability to release the dye initially adsorbed. Thus, the prepared films could be suitable materials for sustainable and effective water treatment technologies.

Project description:Crop production is a serious challenge to provide food for the 10 billion individuals forecasted to live across the globe in 2050. The scientists' emphasize establishing an equilibrium among diversity and quality of crops by enhancing yield to fulfill the increasing demand for food supply sustainably. The exploitation of genetic resources using genomics and metabolomics strategies can help generate resilient plants against stressors in the future. The innovation of the next-generation sequencing (NGS) strategies laid the foundation to unveil various plants' genetic potential and help us to understand the domestication process to unmask the genetic potential among wild-type plants to utilize for crop improvement. Nowadays, NGS is generating massive genomic resources using wild-type and domesticated plants grown under normal and harsh environments to explore the stress regulatory factors and determine the key metabolites. Improved food nutritional value is also the key to eradicating malnutrition problems around the globe, which could be attained by employing the knowledge gained through NGS and metabolomics to achieve suitability in crop yield. Advanced technologies can further enhance our understanding in defining the strategy to obtain a specific phenotype of a crop. Integration among bioinformatic tools and molecular techniques, such as marker-assisted, QTLs mapping, creation of reference genome, de novo genome assembly, pan- and/or super-pan-genomes, etc., will boost breeding programs. The current article provides sequential progress in NGS technologies, a broad application of NGS, enhancement of genetic manipulation resources, and understanding the crop response to stress by producing plant metabolites. The NGS and metabolomics utilization in generating stress-tolerant plants/crops without deteriorating a natural ecosystem is considered a sustainable way to improve agriculture production. This highlighted knowledge also provides useful research that explores the suitable resources for agriculture sustainability.

Project description:All currently available Streptococcus pneumoniae (Spn) vaccines have limitations due to their capsular serotype composition. Both the 23-valent Spn polysaccharide vaccine (PPV) and 7, 10, or 13-valent Spn conjugate vaccines (PCV-7, 10, -13) are serotype-based vaccines and therefore they elicit only serotype-specific immunity. Emergence of replacement Spn strains expressing other serotypes has consistently occurred following introduction of capsular serotype based Spn vaccines. Furthermore, capsular polysaccharide vaccines are less effective in protection against non-bacteremic pneumonia and acute otitis media (AOM) than against invasive pneumococcal disease (IPD). These shortcomings of capsular polysaccharide-based Spn vaccines have created high interest in development of non-serotype specific protein-based vaccines that could be effective in preventing both IPD and non-IPD infections. This review discusses the progress to date on development of Spn protein vaccine candidates that are highly conserved by all Spn strains, are highly conserved, exhibit maximal antigenicity and minimal reactogenicity to replace or complement the current capsule-based vaccines. Key to development of a protein based Spn vaccine is an understanding of Spn pathogenesis. Based on pathogenesis, a protein-based Spn vaccine should include one or more ingredients that reduce NP colonization below a pathogenic inoculum. Elimination of all Spn colonization may not be achievable or even advisable. The level of expression of a target protein antigen during pathogenesis is another key to the success of protein based vaccines.. As with virtually all currently licensed vaccines, production of a serum antibody response in response to protein based vaccines is anticipated to provide protection from Spn infections. A significant advantage that protein vaccine formulations can offer over capsule based vaccination is their potential benefits associated with natural priming and boosting to all strains of Spn. One of the most universal and comprehensive approaches of identifying novel vaccine candidates is the investigation of human sera from different disease stages of natural infections. Antigens that are robustly reactive in preliminary human serum screening constitute a pathogen-specific antigenome. This strategy has identified a number of Spn protein vaccine candidates that are moving forward in human clinical trials.
