Light- and magnetically actuated FePt microswimmers.

ABSTRACT: Externally controlled microswimmers offer prospects for transport in biological research and medical applications. This requires biocompatibility of the swimmers and the possibility to tailor their propulsion mechanisms to the respective low Reynolds number environment. Here, we incorporate low amounts of the biocompatible alloy of iron and platinum (FePt) in its [Formula: see text] phase in microstructures by a versatile one-step physical vapor deposition process. We show that the hard magnetic properties of [Formula: see text] FePt are beneficial for the propulsion of helical micropropellers with rotating magnetic fields. Finally, we find that the FePt coatings are catalytically active and also make for Janus microswimmers that can be light-actuated and magnetically guided.

Automatic Manipulation of Magnetically Actuated Helical Microswimmers in Static Environments.

Project description:Electromagnetically actuated microswimmers have been widely used in various biomedical applications due to their minor invasive traits and their easy access to confined environments. In order to guide the microswimmers autonomously towards a target, an obstacle-free path must be computed using path planning algorithms, meanwhile a motion controller must be formulated. However, automatic manipulations of magnetically actuated microswimmers are underdeveloped and still are challenging topics. In this paper, we develop an automatic manipulation system for magnetically actuated helical microswimmers in static environments, which mainly consists of a mapper, a path planner, and a motion controller. First, the mapper processes the captured image by morphological transformations and then labels the free space and the obstacle space. Second, the path planner explores the obstacle-free space to find a feasible path from the start to the goal by a global planning algorithm. Last, the motion controller guides the helical microswimmers along the desired path by a closed-loop algorithm. Experiments are conducted to verify the effectiveness of the proposed automatic manipulation. Furthermore, our proposed approach presents the first step towards applications of microswimmers for targeted medical treatments, such as micromanipulation, targeted therapy, and targeted drug delivery.

Propulsion of magnetically actuated achiral planar microswimmers in Newtonian and non-Newtonian fluids.

Project description:Magnetic achiral planar microswimmers can be massively fabricated at low cost and are envisioned to be useful for in vivo biomedical applications. To understand locomotion in representative in vivo environments, we investigated the swimming performance of achiral planar microswimmers in methylcellulose solutions. We observed that these microswimmers displayed very similar swimming characteristics in methylcellulose solutions as in water. Furthermore, this study indicated that the range of precession angles increased as the concentration of MC solution increased. Last, it was demonstrated that achiral planar microswimmers with similar precession angles exhibited nearly the same dimensionless speeds in different concentrations of the methylcellulose solutions. Upon understanding swimmer kinematics, more effective control over the achiral planar microswimmers can be achieved to perform multiple biomedical tasks in in vivo environments.

Magnetically Powered Biodegradable Microswimmers.

Project description:The propulsive efficiency and biodegradability of wireless microrobots play a significant role in facilitating promising biomedical applications. Mimicking biological matters is a promising way to improve the performance of microrobots. Among diverse locomotion strategies, undulatory propulsion shows remarkable efficiency and agility. This work proposes a novel magnetically powered and hydrogel-based biodegradable microswimmer. The microswimmer is fabricated integrally by 3D laser lithography based on two-photon polymerization from a biodegradable material and has a total length of 200 μm and a diameter of 8 μm. The designed microswimmer incorporates a novel design utilizing four rigid segments, each of which is connected to the succeeding segment by spring to achieve undulation, improving structural integrity as well as simplifying the fabrication process. Under an external oscillating magnetic field, the microswimmer with multiple rigid segments connected by flexible spring can achieve undulatory locomotion and move forward along with the directions guided by the external magnetic field in the low Reynolds number (Re) regime. In addition, experiments demonstrated that the microswimmer can be degraded successfully, which allows it to be safely applied in real-time in vivo environments. This design has great potential in future in vivo applications such as precision medicine, drug delivery, and diagnosis.

Magnetically-actuated hydrogel-based achiral planar microswimmers for SERS detection: In situ coprecipitation for continuous loading of iron oxide nanoparticles.

Project description:Ultraviolet lithography is a very promising technology used for the batch fabrication of biomedical microswimmers. However, creating microswimmers that can swim at low Reynolds number using biocompatible materials while retaining strong magnetic properties and excellent biomedical functionality is a great challenge. Most of the previously reported biomedical microswimmers possess either strong magnetic properties by using non-biocompatible nickel coating or good biocompatibility by using iron oxide particle-embedded hydrogel with weak magnetism, but not both. Alternatively, iron oxide nanoparticles can be coated on the surface of microswimmers to improve magnetic properties; however, this method limited the usability of the microswimmers' surfaces. To address these shortcomings, this work utilized an in situ synthesis technique to generate high magnetic content inside hydrogel-based achiral planar microswimmers while leaving their surfaces free to be functionalized for SERS detection. The hydrogel matrices of the magnetically actuated hydrogel-based microswimmers were first prepared by ultraviolet lithography. Then, the high concentration of iron oxide was achieved through multiple continuous in situ coprecipitation cycles. Finally, the SERS detection capability of magnetically actuated hydrogel-based microswimmers was enabled by uniformly growing silver nanoparticles on the surface of the microswimmers. In the motion control tests, the microswimmers showed a high swimming efficiency, high step-out frequency, and consistent synchronized motion. Furthermore, the magnetically actuated hydrogel-based microswimmers were able to improve the detection efficiency of analytes under magnetic guidance.

Magnetically Actuated Tunable Soft Electronics.

Project description:Variable electronics are vital in tunable filters, transmitters, and receivers, among other applications. In addition, the ability to remotely tune soft capacitors, resistors, and inductors is important for applications in which the device is not accessible. In this paper, a uniform method of remotely tuning the characteristic properties of soft electronic units (i.e. inductance, capacitance, and resistance) is presented. In this method, magnetically actuated ferrofluid mixed with iron powder is dragged in a soft fluidic channel made of polydimethylsiloxane (PDMS) to tune the electrical properties of the component. The effects of position and quantity of the ferrofluid and iron powder are studied over a range of frequencies, and the changes in inductance, capacitance, resistance, quality factor, and self-resonance frequency are reported accordingly. The position plays a bigger role in changing inductance, capacitance, and resistance. With the proposed design, the inductance can be changed by 20.9% from 3.31 μH for planar inductors and 23% from 0.44 μH for axial inductors. In addition, the capacitance of capacitors and impedance of resistors can be changed by 12.7% from 2.854 pF and 185.3% from 0.353 kΩ, respectively. Furthermore, the changes in the inductance, capacitance, and resistance follow "quasi-linear profiles" with the input during position and quantity effect experiments.

Magnetically-actuated microposts stimulate axon growth.

Project description:New strategies to promote neuronal regeneration should aim to increase the speed of axonal elongation. Biochemical signaling is a key factor in axon growth, but recent discoveries have shown that mechanical force, through a process referred to as stretch growth, can significantly influence the elongation rate. Here, we develop a method to apply forces to primary hippocampal neurons from mice using magnetic microposts that actuate in response to an external magnetic field. Neurons are cultured onto these microposts and subjected to an average displacement of 0.2 μm at a frequency of 5 Hz. We find that the mechanical stimulation promotes an increase in the length of the axons compared to control conditions. In addition, there is an increase in the density of microtubules and in the amount of cisternae of the endoplasmic reticulum, providing evidence that stretch growth is accompanied by a mass addition to the neurite. Together, these results indicate that magnetically-actuated microposts can accelerate the rate of axon growth, paving the way for future applications in neuronal regeneration. VIDEO ABSTRACT.

Magnetically Actuated Fiber-Based Soft Robots.

Project description:Broad adoption of magnetic soft robotics is hampered by the sophisticated field paradigms for their manipulation and the complexities in controlling multiple devices. Furthermore, high-throughput fabrication of such devices across spatial scales remains challenging. Here, advances in fiber-based actuators and magnetic elastomer composites are leveraged to create 3D magnetic soft robots controlled by unidirectional fields. Thermally drawn elastomeric fibers are instrumented with a magnetic composite synthesized to withstand strains exceeding 600%. A combination of strain and magnetization engineering in these fibers enables programming of 3D robots capable of crawling or walking in magnetic fields orthogonal to the plane of motion. Magnetic robots act as cargo carriers, and multiple robots can be controlled simultaneously and in opposing directions using a single stationary electromagnet. The scalable approach to fabrication and control of magnetic soft robots invites their future applications in constrained environments where complex fields cannot be readily deployed.

Cardiac tissue engineering in magnetically actuated scaffolds.

Project description:Cardiac tissue engineering offers new possibilities for the functional and structural restoration of damaged or lost heart tissue by applying cardiac patches created in vitro. Engineering such functional cardiac patches is a complex mission, involving material design on the nano- and microscale as well as the application of biological cues and stimulation patterns to promote cell survival and organization into a functional cardiac tissue. Herein, we present a novel strategy for creating a functional cardiac patch by combining the use of a macroporous alginate scaffold impregnated with magnetically responsive nanoparticles (MNPs) and the application of external magnetic stimulation. Neonatal rat cardiac cells seeded within the magnetically responsive scaffolds and stimulated by an alternating magnetic field of 5 Hz developed into matured myocardial tissue characterized by anisotropically organized striated cardiac fibers, which preserved its features for longer times than non-stimulated constructs. A greater activation of AKT phosphorylation in cardiac cell constructs after applying a short-term (20 min) external magnetic field indicated the efficacy of magnetic stimulation to actuate at a distance and provided a possible mechanism for its action. Our results point to a synergistic effect of magnetic field stimulation together with nanoparticulate features of the scaffold surface as providing the regenerating environment for cardiac cells driving their organization into functionally mature tissue.

Magnetically Actuated Biodegradable Nanorobots for Active Immunotherapy.

Project description:An efficient and cost-effective therapeutic vaccine is highly desirable for the prevention and treatment of cancer, which helps to strengthen the immune system and activate the T cell immune response. However, initiating such an adaptive immune response efficiently remains challenging, especially the deficient antigen presentation by dendritic cells (DCs) in the immunosuppressive tumor microenvironment. Herein, an efficient and dynamic antigen delivery system based on the magnetically actuated OVA-CaCO3 -SPIO robots (OCS-robots) is rationally designed for active immunotherapy. Taking advantage of the unique dynamic features, the developed OCS-robots achieve controllable motion capability under the rotating magnetic field. Specifically, with the active motion, the acid-responsiveness of OCS-robots is beneficial for the tumor acidity attenuating and lysosome escape as well as the subsequent antigen cross-presentation of DCs. Furthermore, the dynamic OCS-robots boost the crosstalk between the DCs and antigens, which displays prominent tumor immunotherapy effect on melanoma through cytotoxic T lymphocytes (CTLs). Such a strategy of dynamic vaccine delivery system enables the active activation of immune system based on the magnetically actuated OCS-robots, which presents a plausible paradigm for incredibly efficient cancer immunotherapy by designing multifunctional and novel robot platforms in the future.

Microfluidic viscometry using magnetically actuated micropost arrays.

Project description:Here we describe development of a microfluidic viscometer based on arrays of magnetically actuated micro-posts. Quantitative viscosities over a range of three orders of magnitude were determined for samples of less than 20 μL. This represents the first demonstration of quantitative viscometry using driven flexible micropost arrays. Critical to the success of our system is a comprehensive analytical model that includes the mechanical and magnetic properties of the actuating posts, the optical readout, and fluid-structure interactions. We found that alterations of the actuator beat shape as parameterized by the dimensionless "sperm number" must be taken into account to determine the fluid properties from the measured actuator dynamics. Beyond our particular system, the model described here can provide dynamics predictions for a broad class of flexible microactuator designs. We also show how the model can guide the design of new arrays that expand the accessible range of measurements.
