Adaptive optics enhanced sensitivity in Fabry-Perot based photoacoustic tomography.

ABSTRACT: All-optical ultrasound detection bears a number of unique advantages for photoacoustic tomography, including the ability for high resolution sampling of the acoustic field and its compatibility with a wide variety of other optical modalities. However, optical schemes based on miniaturized cavities are sensitive to optical aberrations as well as manufacturing-induced cavity imperfections which degrade sensor sensitivity and deteriorate photoacoustic image quality. Here we present an experimental method based on adaptive optics that is capable of enhancing the overall sensitivity of Fabry-Pérot based photoacoustic sensors. We experimentally observe clear improvements in photoacoustic signal detection as well as overall image quality after photoacoustic tomography reconstructions when applied to mammalian tissues in vivo.

Project description:Transformation optics is a powerful tool to design various novel devices, such as invisibility cloak. Fantastic effects from this technique are usually accompanied with singular mappings, resulting in challenging implementations and narrow bands of working frequencies. Here in this article, Fabry-Pérot resonances in materials of extreme anisotropy are used to design various transformation optical devices that are not only easy to realize but also work well for a set of resonant frequencies (multiple frequencies). As an example, a prototype of a cylindrical concentrator is fabricated for microwaves.

Adaptive optics photoacoustic microscopy.

Project description:We have developed an adaptive optics photoacoustic microscope (AO-PAM) for high-resolution imaging of biological tissues, especially the retina. To demonstrate the feasibility of AO-PAM we first designed the AO system to correct the wavefront errors of the illuminating light of PAM. The aberrations of the optical system delivering the illuminating light to the sample in PAM was corrected with a close-loop AO system consisting of a 141-element MEMS-based deformable mirror (DM) and a Shack-Hartmann (SH) wavefront sensor operating at 15 Hz. The photoacoustic signal induced by the illuminating laser beam was detected by a custom-built needle ultrasonic transducer. When the wavefront errors were corrected by the AO system, the lateral resolution of PAM was measured to be better than 2.5 µm using a low NA objective lens. We tested the system on imaging ex vivo ocular samples, e.g., the ciliary body and retinal pigment epithelium (RPE) of a pig eye. The AO-PAM images showed significant quality improvement. For the first time we were able to resolve single RPE cells with PAM.

Photoacoustic Mouse Brain Imaging Using an Optical Fabry-Perot Interferometric Ultrasound Sensor.

Project description:Photoacoustic (PA, or optoacoustic, OA) mesoscopy is a powerful tool for mouse cerebral imaging, which offers high resolution three-dimensional (3D) images with optical absorption contrast inside the optically turbid brain. The image quality of a PA mesoscope relies on the ultrasonic transducer which detects the PA signals. An all-optical ultrasound sensor based on a Fabry-Pérot (FP) polymer cavity has the following advantages: broadband frequency response, wide angular coverage and small footprint. Here, we present 3D PA mesoscope for mouse brain imaging using such an optical sensor. A heating laser was used to stabilize the sensor's cavity length during the imaging process. To acquire data for a 3D angiogram of the mouse brain, the sensor was mounted on a translation stage and raster scanned. 3D images of the mouse brain vasculature were reconstructed which showed cerebrovascular structure up to a depth of 8 mm with high quality. Imaging segmentation and dual wavelength imaging were performed to demonstrate the potential of the system in preclinical brain research.

A total-internal-reflection-based Fabry-Perot resonator for ultra-sensitive wideband ultrasound and photoacoustic applications.

Project description:In photoacoustic and ultrasound imaging, optical transducers offer a unique potential to provide higher responsivity, wider bandwidths, and greatly reduced electrical and acoustic impedance mismatch when compared with piezoelectric transducers. In this paper, we propose a total-internal-reflection-based Fabry-Pérot resonator composed of a 12-nm-thick gold layer and a dielectric resonant cavity. The resonator uses the same Kretschmann configuration as surface plasmon resonators (SPR). The resonators were analyzed both theoretically and experimentally. The experimental results were compared with those for an SPR for benchmarking. The 1.9-μm-thick-PMMA- and 3.4-μm-thick-PDMS-based resonators demonstrated responsivities of 3.6- and 30-fold improvements compared with the SPR, respectively. The measured bandwidths for the PMMA, PDMS devices are 110 MHz and 75 MHz, respectively. Single-shot sensitivity of 160 Pa is obtained for the PDMS device. The results indicate that, with the proposed resonator in imaging applications, sensitivity and the signal-to-noise ratio can be improved significantly without compromising the bandwidth.

Photodetector based on Vernier-Enhanced Fabry-Perot Interferometers with a Photo-Thermal Coating.

Project description:We present a new type of fiber-coupled photodetector with a thermal-based optical sensor head, which enables it to operate even in the presence of strong electro-magnetic interference and in electrically sensitive environments. The optical sensor head consists of three cascaded Fabry-Perot interferometers. The end-face surface is coated with copper-oxide micro-particles embedded in hydrogel, which is a new photo-thermal coating that can be readily coated on many different surfaces. Under irradiation, photons are absorbed by the photo-thermal coating, and are converted into heat, changing the optical path length of the probing light and induces a resonant wavelength shift. For white-light irradiation, the photodetector exhibits a power sensitivity of 760 pm/mW, a power detection limit of 16.4 μW (i.e. specific detectivity of 2.2 × 105 cm.√Hz/W), and an optical damage threshold of ~100 mW or ~800 mW/cm2. The response and recovery times are 3.0 s (~90% of change within 100 ms) and 16.0 s respectively.

Simply Fabricated Inexpensive Dual-Polymer-Coated Fabry-Perot Interferometer-Based Temperature Sensors with High Sensitivity.

Project description:We designed simply fabricated, highly sensitive, and cost-effective dual-polymer-coated Fabry-Perot interferometer (DFPI)-based temperature sensors by employing thermosensitive polymers and non-thermosensitive polymers, as well as different two successive dip-coating techniques (stepwise dip coating and polymer mixture coating). Seven sensors were fabricated using different polymer combinations for performance optimization. The experiments demonstrated that the stepwise dip-coated dual thermosensitive polymer sensors exhibited the highest sensitivity (2142.5 pm °C-1 for poly(methyl methacrylate)-polycarbonate (PMMA_PC) and 785.5 pm °C-1 for poly(methyl methacrylate)- polystyrene (PMMA_PS)). Conversely, the polymer-mixture-coated sensors yielded low sensitivities (339.5 pm °C-1 for the poly(methyl methacrylate)-polycarbonate mixture (PMMA_PC mixture) and 233.5 pm °C-1 for the poly(methyl methacrylate)-polystyrene mixture (PMMA_PS mixture). Thus, the coating method, polymer selection, and thin air-bubble-free coating are crucial for high-sensitivity DFPI-based sensors. Furthermore, the DFPI-based sensors yielded stable readouts, based on three measurements. Our comprehensive results confirm the effectiveness, reproducibility, stability, fast response, feasibility, and accuracy of temperature measurements using the proposed sensors. The excellent performance and simplicity of our proposed sensors are promising for biomedical, biochemical, and physical applications.

Adaptive optics optical coherence tomography in glaucoma.

Project description:Since the introduction of commercial optical coherence tomography (OCT) systems, the ophthalmic imaging modality has rapidly expanded and it has since changed the paradigm of visualization of the retina and revolutionized the management and diagnosis of neuro-retinal diseases, including glaucoma. OCT remains a dynamic and evolving imaging modality, growing from time-domain OCT to the improved spectral-domain OCT, adapting novel image analysis and processing methods, and onto the newer swept-source OCT and the implementation of adaptive optics (AO) into OCT. The incorporation of AO into ophthalmic imaging modalities has enhanced OCT by improving image resolution and quality, particularly in the posterior segment of the eye. Although OCT previously captured in-vivo cross-sectional images with unparalleled high resolution in the axial direction, monochromatic aberrations of the eye limit transverse or lateral resolution to about 15-20 μm and reduce overall image quality. In pairing AO technology with OCT, it is now possible to obtain diffraction-limited resolution images of the optic nerve head and retina in three-dimensions, increasing resolution down to a theoretical 3 μm3. It is now possible to visualize discrete structures within the posterior eye, such as photoreceptors, retinal nerve fiber layer bundles, the lamina cribrosa, and other structures relevant to glaucoma. Despite its limitations and barriers to widespread commercialization, the expanding role of AO in OCT is propelling this technology into clinical trials and onto becoming an invaluable modality in the clinician's arsenal.

Spatially resolved imaging of human macular capillaries using adaptive optics-enhanced optical coherence tomography angiography.

Project description:Documenting the organization of the retinal capillaries is of importance to understand the visual consequences of vascular diseases which may differentially affect the microvascular layers. Here we detailed the spatial organization of the macular capillaries in ten healthy human subjects using a prototypic adaptive optics-enhanced optical coherence tomography angiography (AO-OCTA) system. Within the central 6° × 6°, the radial peripapillary capillaries and the superficial, intermediate and deep vascular plexuses (SVP, IVP and DVP, respectively) were consistently resolved. In 8 out of the 10 eyes, the capillary segments composing the perifoveal arcade (PFA) were perfused only by the SVP, while drainage of the PFA showed more variability, comprising a case in which the PFA was drained by the DVP. Around the center, a distinct central avascular zone could be documented for each layer in 7 of the 10 cases; in three eyes, the IVP and SVP merged tangentially around the center. In all eyes, the foveal avascular zone was larger in the DVP than in the SVP and IVP. In one eye with incomplete separation of the inner foveal layers, there was continuity of both the SVP and the IVP; a central avascular zone was only present in the DVP. The diversity of perfusion and drainage patterns supported a connectivity scheme combining parallel and serial organizations, the latter being the most commonly observed in perifoveal vessels. Our results thus help to further characterize the diversity of organization patterns of the macular capillaries and to robustly analyze the IVP, which will help to characterize early stages of microvascular diseases.

Versatile tissue lasers based on high-Q Fabry-Perot microcavities.

Project description:Biolasers are an emerging technology for next generation biochemical detection and clinical applications. Progress has recently been made to achieve lasing from biomolecules and single living cells. Tissues, which consist of cells embedded in an extracellular matrix, mimic more closely the actual complex biological environment in a living body and therefore are of more practical significance. Here, we developed a highly versatile tissue laser platform, in which tissues stained with fluorophores are sandwiched in a high-Q Fabry-Pérot microcavity. Distinct lasing emissions from muscle and adipose tissues stained respectively with fluorescein isothiocyanate (FITC) and boron-dipyrromethene (BODIPY), and hybrid muscle/adipose tissue with dual staining were achieved with a threshold of only ∼10 μJ mm-2. Additionally, we investigated how the tissue structure/geometry, tissue thickness, and staining dye concentration affect the tissue laser. Lasing emission from FITC conjugates (FITC-phalloidin) that specifically target F-actin in muscle tissues was also realized. It is further found that, despite the large fluorescence spectral overlap between FITC and BODIPY in tissues, their lasing emissions could be clearly distinguished and controlled due to their narrow lasing bands and different lasing thresholds, thus enabling highly multiplexed detection. Our tissue laser platform can be broadly applicable to various types of tissues/diseases. It provides a new tool for a wide range of biological and biomedical applications, such as diagnostics/screening of tissues and identification/monitoring of biological transformations in tissue engineering.

Guide-star-based computational adaptive optics for broadband interferometric tomography.

Project description:We present a method for the numerical correction of optical aberrations based on indirect sensing of the scattered wavefront from point-like scatterers ("guide stars") within a three-dimensional broadband interferometric tomogram. This method enables the correction of high-order monochromatic and chromatic aberrations utilizing guide stars that are revealed after numerical compensation of defocus and low-order aberrations of the optical system. Guide-star-based aberration correction in a silicone phantom with sparse sub-resolution-sized scatterers demonstrates improvement of resolution and signal-to-noise ratio over a large isotome. Results in highly scattering muscle tissue showed improved resolution of fine structure over an extended volume. Guide-star-based computational adaptive optics expands upon the use of image metrics for numerically optimizing the aberration correction in broadband interferometric tomography, and is analogous to phase-conjugation and time-reversal methods for focusing in turbid media.
