Antimicrobial and antifungal activity of soil actinomycetes isolated from coal mine sites.

ABSTRACT: In the current study, twenty-eight soil samples were collected from coalmine sites of Telangana, India. The isolates were purified and identified based on their culture characterization on oatmeal agar, glycerol asparagine agar, yeast extract-malt extract agar, inorganic salt starch agar, and starch casein agar medium. Further, the supernatant of all the isolates were tested for antimicrobial and antifungal activities. The biochemical and microscopic studies of isolated strains results indicates the potential isolate strains belongs to Streptomyces genus. Among all the strains the biological activity of BHPL-KSKU5 showed higher anti-bacterial and anti-funagal activity. The molecular characterization of BHPL-KSKU5 16s rDNA gene sequence and phylogenetic tree showed that is mostly related to the Streptomysis felleus (S. felleus) strain. This isolate was submitted to gene bank NCBI with accession number MH553077. In addition, physiological studies such as utilization of carbon, nitrogen, amino acid sources of potential isolated were studied. Further, optimization, purification and characterization of the novel compound producing strain may be helpful for discovering the new therapeutic microbial agent.

Project description:Antibiotic resistance is one of the most important global healthcare challenges and is responsible for the mortality of millions of people worldwide every year. It is a crisis attributed to misuse of antibiotics and a lack of new drug development. Actinomycetes constitute a group of Gram-positive bacteria known for their distinctive high guanine-cytosine (G+C) content in their genomic DNA. These microorganisms are widely recognized for their capability to generate a wide range of secondary metabolites with diverse biological activities. These versatile microorganisms are ubiquitous in diverse ecosystems, including soil, freshwater, marine sediments, and within the bodies of insects. A recent study has demonstrated that social insects, such as ants, host a diverse array of these bacteria. In this study, we involved the isolation and characterization of a total of 72 actinomycete strains obtained from 18 distinct ant species collected from various regions across Thailand. Utilizing 16S rRNA gene analysis, these isolated actinomycetes were classified into four distinct genera: Amycolatopsis (2 isolates), Micromonospora (1 isolate), Nocardia (8 isolates), and Streptomyces (61 isolates). Among the Streptomyces strains, 23 isolates exhibited antimicrobial activity against a panel of Gram-positive bacteria, including Bacillus subtilis ATCC 6633, Staphylococcus epidermidis ATCC 12228, Staphylococcus aureus ATCC 25923, Kocuria rhizophila ATCC 9341, and Methicillin-resistant Staphylococcus aureus (MRSA) DMST 20646. Additionally, two isolates displayed antifungal activity against Candida albicans TISTR 5554. Based on 16S rRNA gene sequence similarity studies, these two isolates, ODS25 and ODS28, were demonstrated to be closely related to Streptomyces lusitanus NBRC 13464T (98.07%) and Streptomyces haliclonae DSM 41970T (97.28%), respectively. The level of 16S rRNA gene sequence similarity below 98.65% cutoff indicates its potential as a novel actinomycete species. These findings underscore the potential of actinomycetes sourced from ants as a valuable reservoir of novel antimicrobials.

Project description:This study assessed the water suitability for various purposes, evaluated heavy metal contamination and soil fertility, and investigated seasonal variations in water and soil parameters near the Barapukuria coal mine in Bangladesh. A total of nine sampling locations were selected, resulting in 18 samples (12 water and 6 soil) collected during the summer and winter seasons. The water samples were analyzed at the Environmental Engineering Laboratory, while the soil samples were analyzed at the Soil Resource Development Institute (SRDI). The suitability of drinking water was evaluated using the Water Quality Index (WQI), and heavy metal contamination was assessed using the geo-accumulation index. The findings indicate that surface water is generally suitable for irrigation due to several positive attributes, with slight to moderate seasonal variations. However, treatment is required for high turbidity and chemical oxygen demand (COD). On the other hand, wastewater quality exhibits elevated levels of turbidity, total suspended solids, and chemical oxygen demand, although most parameters are within acceptable limits. The assessment of drinking water quality using the WQI reveals that both hand and deep tubewells are generally unfit for consumption. The WQI values peak at 441.84 in summer and 202.79 in winter for the hand tubewell, and 129.66 in summer and 95.88 in winter for the deep tubewell. This finding emphasizes the need for effective treatment and continuous monitoring. The soil analysis indicates that most heavy metals are not present in significant amounts, except for cadmium, which shows uncontaminated to moderately contaminated conditions during the summer. Moreover, the soil treated with coal water exhibits significantly higher levels of organic matter, micronutrients, and macronutrients compared to the normal field soil. Further analysis of the coal water treated soil shows a shift from a slightly acidic pH in summer (6.30) to a neutral pH in winter (7.10). The organic matter content remains consistently high, and nutrient analysis shows high to very high levels of Zn, Cu, K, Ca, Mg, Mn, and Fe, while phosphorus remains low. Seasonal analysis reveals increased levels of zinc, potassium, and phosphorus in winter, whereas other nutrients generally decrease. The primary novel contribution of this study is the detailed seasonal analysis of water and soil quality near the coal mining area, capturing changes across summer and winter. Additionally, it compares coal water treated soil with normal field soil, revealing significant improvements in soil quality and fertility.

Project description:Heavy metals contaminations in mining areas aroused wide concerns globally. Efficient evaluation of its pollution status is a basis for further soil reclamation. Visible and near-infrared reflectance (Vis-NIR) spectroscopy has been diffusely used for retrieving heavy metals concentrations. However, the reliability and feasibility of calibrated models were still doubtful. The present study estimated zinc (Zn) concentrations via the random forest (RF) and partial least squares regression (PLSR) using ground in-situ Zn concentrations as well as soil spectral reflectance at an Opencast Coal Mine of Ordos, China in February 2020. The coefficient of determination (R2), root mean square error (RMSE), mean absolute error (MAE), and the ratio of performance to deviation (RPD) were selected to assess the robustness of the methods in estimating Zn contents. Moreover, the characteristic bands were chosen by Pearson correlation analysis and Boruta Algorithm. Finally, the comparison between RF and PLSR combined with eight spectral reflectance transformation methods was conducted for four concentration groups to determine the optimal model. The results indicated that: (1) Zn contents represented a skewed distribution (coefficient of variation (CV) = 33%); (2) the spectral reflectance tended to decrease with the increase of Zn contents during 580-1850 nm based on Savitzky-Golay smoothing (SG); (3) the continuous wavelet transform (CWT) demonstrated higher effectiveness than other spectral reflectance transformation methods in enhancing spectral responses, the R2 between Zn contents and the soil spectral reflectance achieved the highest (R2 = 0.71) by using CWT; (4) the RF combined with CWT exhibited the best performance than other methods in the current study (R2 = 0.97, RPD = 3.39, RMSE = 1.05 mg kg-1, MAE = 0.79 mg kg-1). The current study supplied a scientific scheme and theoretical support for predicting heavy metals concentrations via the Vis-NIR spectral method in possible contaminated areas such as coal mines and metallic mineral deposit areas.

Project description:Actinomycetes are Gram-positive, facultative anaerobic fungus-like filamentous bacteria which remain on the top of the natural antibiotic producers. Due to the climatic and geographical diversity of Nepal, a wide range of microorganisms with potent source of antimicrobials are available. The objective of this study was to isolate, identify, and screen the potential antimicrobial-producing actinomycetes from soils covering different altitude range of Nepal. Forty-one isolates of actinomycetes were isolated from 11 soil samples collected from different locations in Nepal with altitude ranging from 1500 to 4380 meters. The isolates were identified on the basis of morphological study, different sugar utilization, protein utilization, and hydrolysis tests. They were also characterized on the basis of temperature and pH. Primary screening for antimicrobial activity was carried out against several test organisms: Staphylococcus aureus (ATCC 25923), Escherichia coli (ATCC 25922), Klebsiella pneumoniae (ATCC 700603), and Pseudomonas aeruginosa (ATCC 27853) by the perpendicular streaking method, and secondary screening was carried out by the agar well diffusion method using ethyl acetate for solvent extraction. 70.7% of the isolates were identified as Streptomyces spp., 19.5% as Nocardia spp., and 9.5% as Micromonospora spp. 43.34% of actinomycete isolates was found to be potent antimicrobial producers from the primary screening among which 46.34% were effective against Gram-positive and 12.19% against Gram-negative test organisms. Isolate C7 (Micromonospora spp.) showed the best broad-spectrum antimicrobial activity during secondary screening. A total of 11 different types of pigments were observed to be produced by different isolates, of which, the yellow pigment was the most prominent. The association between elevation, pH, and pigment with the antimicrobial production was found to be insignificant. This finding can be of importance for further investigation towards obtaining broad-spectrum antibiotics for therapeutic purpose.
