The complete chloroplast genome sequence of Potentilla glabra Lodd.

ABSTRACT: Potentilla glabra Lodd is an important traditional Chinese medicine, which is widely distributed in many areas of China. It has a variety of pharmacological activities and has been drunk as a tea for a long time. Therefore, it has great economic, research, and social value. However, no plastid genome has been reported to date. Here, we report species of this genus complete chloroplast genome of Potentilla glabra Lodd. The chloroplast genome of Potentilla glabra Lodd is found to be 152,900 bp in length with 37.24% GC contents. The cp genome sequences contained 130 genes, including 84 mRNA genes, 37 tRNA, eight rRNA genes, and one pseudogenes, respectively. A phylogenetic tree reconstructed by 42 chloroplast genomes reveals that Potentilla glabra Lodd is most related with Potentilla parvifolia. The complete chloroplast genome of these plants will be benefit for studies on the general characteristics and evolution of the Potentilla family genome.

The whole chloroplast genome was 157, 902 base pairs in length with 129 genes, including 84 protein-coding genes, 37 tRNAs, and 8 rRNAs. The maximum likelihood phylogenetic analysis revealed that the species of P. bifurca was isolated first among the genus Potentilla. This result will be helpful for the conservation and phylogeny programs of the genus Potentilla.

Characterization of the complete chloroplast genome sequence of Sarcandra glabra (Chloranthales).

Project description:Sarcandra glabra is a perennial evergreen subshrub, with high ornamental and medicinal value. Using the Illumina high-throughput sequencing data, its chloroplast genome is well assembled and characterized. The complete chloroplast genome is 158,872 bp in length with a typical quadripartite structure: a pair of inverted repeats (IRs) of 26,122 bp for each, an 88,182 bp large single-copy (LSC) region and an 18,445 bp small single-copy (SSC) region. It was composed of 128 genes and they were identified 84 coding genes, 8 rRNA genes, 36 tRNA genes. Phylogenetic analysis confirmed that the position of S. glabra lay within the order Chloranthales instead of Piperales simply according to classical morphological taxonomy.

The complete chloroplast genome sequence of is 155,841 bp in length, contains 128 complete genes, including 84 protein-coding genes (84 PCGs), 8 ribosomal RNA genes (4 rRNAs), and 37 tRNA genes (37 tRNAs). The overall GC content of cp DNA is 37.1%, the corresponding values of the LSC, SSC, and IR regions are 35.0%, 31.0%, and 42.8%. Phylogenetic tree shows that P. bifurca was identified as the basal clade of Potentilla.

The complete chloroplast genome of Waldheimia glabra.

Project description:Waldheimia glabra (Decne.) Rgl. 1879 (family Asteraceae) is a perennial herb with high economic and medicinal values. In this study, we sequenced the complete chloroplast (cp) genome of W. glabra by high-throughput Illumina sequencing. The size of the W. glabra cp genome is 151,499 bp, with overall GC content of 37.3%. It contains a large single copy and a small single copy region of 83,078 bp and 18,457 bp, respectively, separated by a pair of inverted repeats regions of 24,982 bp. We also discovered 131 genes, including 86 protein-coding genes, 37 transfer RNA genes, and 8 ribosomal RNA genes in the genome. The maximum-likelihood phylogenetic tree demonstrated that W. glabra is closely related to Leucanthemella linearis.

The complete chloroplast genome of Bougainvillea glabra.

Project description:Bougainvillea glabra is one of the most popular ornamental and landscaping plants planted in tropical and subtropical regions. The brightly colored bracts, long florescence and strong stress resistance make B. glabra perfect ornamental horticulture plant. Bougainvillea plants have been frequently hybridized, resulting in more than 400 varieties. To investigate the chloroplast genome will help us to understand the biological diversity and stress resistance of Bougainvillea plants better. Here, we report the complete chloroplast genome of B. glabra, which is 154,542 bp in length, including a large single copy (LSC) region of 85,695 bp and a small single copy (SSC) region of 18,077 bp, separated by a pair of identical inverted repeat regions (IRs) of 25,385 bp each. A total of 128 genes were identified, including 83 protein-coding genes, 37 tRNA genes, and 8 rRNA genes. Phylogenetic analysis based on 12 chloroplast genomes showed that B. glabra, accompanied with its sister species B. spectabilis, formed a base clade in Nyctaginaceae which was close to Pisonia aculeata. This study will be helpful for better understanding of the genetic diversity and stress resistance of Bougainvillea plants.

The complete chloroplast genome of Potentilla Freyniana Bornm. (Rosaceae).

Project description:Potentilla freyniana Bornm. has radical leaves with petioles and trifoliolate distinguished from Potentilla fragarioides and Duchesnea indica. In this study, we presented first complete chloroplast genome of P. freyniana to understand its phylogenetic position. Its length is 156,381 bp long and has four subregions: 85,724 bp of large single copy (LSC) and 18,617 bp of small single copy (SSC) regions are separated by 26,020 bp of inverted repeat (IR) regions including 129 genes (84 protein-coding genes, 8 rRNAs, and 37 tRNAs). The overall GC content of the chloroplast genome is 36.9% and those in the LSC, SSC, and IR regions are 34.8%, 30.7%, and 42.7%, respectively. Phylogenetic trees show that phylogenetic position of P. freyniana disagrees with three phylogenetic studies, showing that more Potentilla chloroplast genomes are required for clarifying Potentilla phylogeny.

Characterization of the complete chloroplast genome sequence of Potentilla gageodoensis (rosaceae), endemic to the continental islands of Korea.

Project description:This study reports the complete chloroplast genome sequence of a continental island endemic, Potentilla gageodoensis. The total plastome size was 156,397 bp, comprising one large single-copy (LSC; 85,768 bp), one small single-copy (SSC; 18,589 bp), and two inverted repeat (IR) regions (IRa and IRb, each with 26,020 bp). The overall GC content was 36.92%, and the plastome contained 131 genes, comprising 84 protein-coding genes with two pseudogenes (infA and ycf1), 37 transfer RNA genes, and eight ribosomal RNA genes. Phylogenetic analysis performed using 27 representative Rosoideae plastomes suggests that the genus Potentilla is not monophyletic and that P. gageodoensis is sister to the clade containing four taxa of Potentilla (P. freyniana, P. freyniana var. chejuensis, P. stolonifera, and P. stolonifera var. quelpaertensis). The present study reveals the taxonomic distinction of P. gageodoensis from its congeneric species in Korea and the plastome sequence obtained from this study can be used to study phylogenetic relationships and taxonomic status.

Project description:Potentilla sischanensis Bunge ex Lehm. is a widespread perennial herb in north China. The plant has little yellow flowers, and the petioles are white-tomentose and sparsely villous. To determine the chloroplast genome, total genomic DNA was extracted from fresh leaves and sequenced. The complete chloroplast genome was assembled and annotated. The chloroplast genome of this plant is a circular form with a length of 156,240 bp, including a large single-copy region (LSC, 85,748 bp), a small single-copy region (SSC, 18,566 bp), and two inverted repeats (IRs, 25,963 bp). A total of 132 genes were predicted, comprising 87 encoded proteins, 8 rRNAs and 37 tRNAs. The evolutionary history indicates that P. sischanensis was grouped within Potentilla and formed a clade with Potentilla chinensis and Potentilla stolonifera with a 100% bootstrap support value. The complete cp genome can serve as a reference for future studies on molecular biology, evolution, population genetics, taxonomy and resource protection.
