Secondary Effects of Hypochlorite Treatment on the Emerging Pollutant Candesartan: The Formation of Degradation Byproducts and Their Toxicological Profiles.

ABSTRACT: In recent years, many studies have reported the frequent detection of antihypertensive agents such as sartans (olmesartan, valsartan, irbesartan and candesartan) in the influents and effluents of wastewater treatment plants (WWTPs) and in the superficial waters of rivers and lakes in both Europe and North America. In this paper, the degradation pathway for candesartan (CAN) was investigated by simulating the chlorination process that is normally used to reduce microbial contamination in a WWTP. Twelve isolated degradation byproducts (DPs), four of which were isolated for the first time, were separated on a C-18 column by employing a gradient HPLC method, and their structures were identified by combining nuclear magnetic resonance and mass spectrometry and comparing the results with commercial standards. On the basis of these results, a mechanism of formation starting from the parent drug is proposed. The ecotoxicity of CAN and its DPs was studied by conducting a battery of ecotoxicity tests; bioassays were performed using Aliivibrio fischeri (bacterium), Daphnia magna (planktonic crustacean) and Raphidocelis subcapitata (alga). The ecotoxicity results shed new light on the increased toxicity of DPs compared with the parent compound.

Project description:BACKGROUND:The presence of a wide range of bioactive organic pollutants in wastewater and municipal water sources is raising concerns about their potential effects on humans. Not surprisingly, various approaches are being explored that can efficiently degrade these persistent organic pollutants. Use of peroxidases has recently been recognized as a novel remediation approach that may have potential advantages over conventional degradation techniques. However, testing the abilities of different peroxidases to degrade diverse emerging pollutants is tedious and cumbersome. RESULTS:In the present study, we present a rapid and robust approach to easily test the degradability of 21 different emerging pollutants by five different peroxidases (soybean peroxidase, chloroperoxidase, lactoperoxidase, manganese peroxidase, and horseradish peroxidase) using an LC-MSMS approach. Furthermore, this approach was also used to examine the role of a redox mediator in these enzymatic degradation assays. Our results show that some of the organic pollutants can be easily degraded by all five of the peroxidases tested, whereas others are only degraded by a specific peroxidase (or when a redox mediator was present) and there are some that are completely resistant to degradation by any of the peroxidases tested (even in the presence of a redox mediator). The degradation of furosemide and trimethoprim by soybean peroxidase and chloroperoxidase, respectively, was investigated in detail by examining the transformation products generated during their degradation. Some of the products generated during enzymatic breakdown of these pollutants have been previously reported by others, however, we report many new transformation products. CONCLUSIONS:LC-MSMS approaches, like the one described here, can be used to rapidly evaluate the potential of different peroxidases (and redox requirements) to be used as bioremediation agents. Our preliminary result shows peroxidases hold tremendous potential for being used in a final wastewater treatment step.

Project description:Alloys or smelted metal mixtures have served as cornerstones of human civilization. The advent of smelted copper and tin, i.e., bronze, in the 4th millennium B.C. in Mesopotamia has pioneered the preparation of other metal composites, such as brass (i.e., mixture of copper and zinc), since the bronze age. The contemporary use of these alloys has expanded beyond using their physical strength. The catalytic chemistry of micron-scale brass or copper-zinc alloy can be utilized to effectively degrade emerging contaminants (ECs) in water, which are presenting significant risks to human health and wildlife. Here, we examine the photocatalytic activity of a commercially available micro-copper-zinc alloy (KDF® 55, MicroCuZn), made with earth abundant metals, for oxidative removal of two ECs. The micron-scale brass is independently characterized for its morphology, which confirms that it has the β-brass phase and that its plasmonic response is around 475 nm. Estriol (E3), a well-known EC, is removed from water with ultraviolet (UV) radiation catalyzed by MicroCuZn and H2O2-MicroCuZn combinations. The synergy between H2O2, UV, and MicroCuZn enhances hydroxyl radical (˙OH) generation and exhibit a strong pseudo-first-order kinetic degradation of E3 with a decay constant of 1.853 × 10-3 min-1 (r 2 = 0.999). Generation of ˙OH is monitored with N,N-dimethyl-4-nitrosoaniline (pNDA) and terephthalic acid (TA), which are effective ˙OH scavengers. X-ray photoelectron spectroscopy analysis has confirmed ZnO/CuO-Cu2O film formation after UV irradiation. The second EC studied here is Δ9-tetrahydrocannabinol or THC, a psychotropic compound commonly consumed through recreational or medicinal use of marijuana. The exceptionally high solids-water partitioning propensity of THC makes adsorption the dominant removal mechanism, with photocatalysis potentially supporting the removal efficacy of this compound. These results indicate that MicroCuZn can be a promising oxidative catalyst especially for degradation of ECs, with possible reusability of this historically significant material with environmentally-friendly attributes.
