Toxicity of sediment oiled with diluted bitumens to freshwater and estuarine amphipods.

ABSTRACT: To address knowledge gaps and the lack of benchmarks on the toxicity of dilbit oiled sediments, weathered Cold Lake Blend (CLB) and Western Canadian Select (WCS) were assessed in 10-day sediment tests with the amphipods Hyalella azteca and Leptocheirus plumulosus. Lowest observed effect concentrations (LOECs) and 20% effect levels (EC20s) were determined for wet weight sediment concentrations of TPH and total PAHs normalized to 1% organic carbon. LOECs and EC20s for TPH ranged from 216 to 1165 mg/kg sediment in H. azteca, and from 64 to 75 mg/kg sediment in L. plumulosus. Dilbit LOECs and EC20s for total PAHs ranged from 2.9 to 11.8 mg/kg sediment in H. azteca, and from 0.75 to 0.87 mg/kg in L. plumulosus. Comparison of toxicity-based benchmarks derived from the current study to sediment concentrations from past spills indicate that dilbit spills in aquatic habitats may pose substantial risks to freshwater and estuarine benthic organisms.

Project description:The acceleration of global climate change draws increasing attention towards interactive effects of temperature and organic contaminants. Many studies reported a higher sensitivity of aquatic invertebrates towards contaminant exposure with increasing or fluctuating temperatures. The hypothesis of this study was that the higher sensitivity of invertebrates is associated with the changes of toxicokinetic processes that determine internal concentrations of contaminants and consequently toxic effects. Therefore, the influence of temperature on toxicokinetic processes and the underlying mechanisms were studied in two key amphipod species (Gammarus pulex and Hyalella azteca). Bioconcentration experiments were carried out at four different temperatures with a mixture of 12 exposure relevant polar organic contaminants. Tissue and medium samples were taken in regular intervals and analysed by online solid-phase extraction liquid chromatography high-resolution tandem mass spectrometry. Subsequently, toxicokinetic rates were modelled and analysed in dependence of the exposure temperature using the Arrhenius equation. An exponential relationship between toxicokinetic rates versus temperature was observed and could be well depicted by applying the Arrhenius equation. Due to a similar Arrhenius temperature of uptake and elimination rates, the bioconcentration factors of the contaminants were generally constant across the temperature range. Furthermore, the Arrhenius temperature of the toxicokinetic rates and respiration was mostly similar. However, in some cases (citalopram, cyprodinil), the bioconcentration factor appeared to be temperature dependent, which could potentially be explained by the influence of temperature on active uptake mechanisms or biotransformation. The observed temperature effects on toxicokinetics may be particularly relevant in non-equilibrated systems, such as exposure peaks in summer as exemplified by the exposure modelling of a field measured pesticide peak where the internal concentrations increased by up to fourfold along the temperature gradient. The results provide novel insights into the mechanisms of chemical uptake, biotransformation and elimination in different climate scenarios and can improve environmental risk assessment.

Project description:Compared to freshwater ecosystems, the health status of estuarine waters remains little studied despite their importance for many species. They also represent a zone of interest for Human settlements that make them the final sink of pollution in both the water column and sediment. Once in sediments, pollutants could represent a threat to benthic as well as pelagic estuarine species through resuspension events. In the Seine estuary, the copepod Eurytemora affinis has been previously presented as a relevant species to assess resuspended sediment contamination through the use of fitness-related effects at the individual level. The aim of the present study was to use E. affinis copepods to assess estuarine sediment-derived elutriates toxicity using both a molecular (i.e. transcriptomics) and a behavioral approach. Two sites along the Seine estuary were sampled. They were both under anthropic pressures from the industrial-port activities or wastewater treatment plants (i.e. Tancarville) or agricultural pressure from freshwater affluent (i.e. Fatouville). The analysis of sediments used to prepare elutriates reveals that both sites have close contamination profiles. The transcriptomic analysis reveals that exposure to both sites triggers the dysregulation of genes involved in biological function as defense response, immunity, ecdysone pathway or neurotoxicity. This analysis also reveals a higher count of dysregulated genes in the Fatouville site compared to the Tancarville despite their close contamination profile. These results emphasize the sensitivity of this molecular approach to assess environmental matrix toxicity with E. affinis. The analysis of the swimming behavior of E. affinis did not highlight significant effects after both sites elutriate exposure. However, our strategy to assess E. affinis swimming behavior (i.e the combination of the DanioVision observation chamber and the EthoVision analysis software) allows the discrimination of basal swimming behavior in this species. Thus, it represents a promising standardized tool to assess copepods swimming behavior in ecotoxicological studies.

Project description:As marine sediments are buried, microbial communities transition from sulfate-reduction to methane-production after sulfate is depleted. When this biogenic methane diffuses into the overlying sulfate-rich sediments, it forms a sulfate-methane transition zone (SMTZ) because sulfate reducers deplete hydrogen concentrations and make hydrogenotrophic methanogenesis exergonic in the reverse direction, a process called the anaerobic oxidation of methane (AOM). Microbial participation in these processes is often inferred from geochemistry, genes, and gene expression changes with sediment depth, using sedimentation rates to convert depth to time. Less is known about how natural sediments transition through these geochemical states transition in real-time. We examined 16S rRNA gene amplicon libraries and metatranscriptomes in microcosms of anoxic sediment from the White Oak River estuary, NC, with three destructively sampled replicates with methane added (586-day incubations) and three re-sampled un-amended replicates (895-day incubations). Sulfate dropped to a low value (∼0.3 mM) on similar days for both experiments (312 and 320 days, respectively), followed by a peak in hydrogen, intermittent increases in methane-cycling archaea starting on days 375 and 362 (mostly Methanolinea spp. and Methanosaeta spp., and Methanococcoides sp. ANME-3), and a methane peak 1 month later. However, methane δ13C values only show net methanogenesis 6 months after methane-cycling archaea increase and 4 months after the methane peak, when sulfate is consistently below 0.1 mM and hydrogen increases to a stable 0.61 ± 0.13 nM (days 553-586, n = 9). Sulfate-reducing bacteria (mostly Desulfatiglans spp. and Desulfosarcina sp. SEEP-SRB1) increase in relative abundance only during this period of net methane production, suggesting syntrophy with methanogens in the absence of sulfate. The transition from sulfate reduction to methane production in marine sediments occurs through a prolonged period of methane-cycling by methanogens at low sulfate concentrations, and steady growth of sulfate reducers along with methanogens after sulfate is depleted.
