Movie-watching outperforms rest for functional connectivity-based prediction of behavior.

ABSTRACT: A major goal of human neuroscience is to relate differences in brain function to differences in behavior across people. Recent work has established that whole-brain functional connectivity patterns are relatively stable within individuals and unique across individuals, and that features of these patterns predict various traits. However, while functional connectivity is most often measured at rest, certain tasks may enhance individual signals and improve sensitivity to behavior differences. Here, we show that compared to the resting state, functional connectivity measured during naturalistic viewing-i.e., movie watching-yields more accurate predictions of trait-like phenotypes in the domains of both cognition and emotion. Traits could be predicted using less than three minutes of data from single video clips, and clips with highly social content gave the most accurate predictions. Results suggest that naturalistic stimuli amplify individual differences in behaviorally relevant brain networks.

Brain connectivity at rest predicts individual differences in normative activity during movie watching.

Project description:When exposed to the same sensory event, some individuals are bound to have less typical experiences than others. Previous research has investigated this phenomenon by showing that the typicality of one's sensory experience is associated with the typicality of their stimulus-evoked brain activity (as measured by intersubject correlation, or ISC). Individual differences in ISC have recently been attributed to variability in focal neural processing. However, the extent to which these differences reflect purely intra-regional variability versus variation in the brain's baseline ability to transmit information between regions has yet to be established. Here, we show that an individual's degree and spatial distribution of ISC are closely related to their brain's functional organization at rest. Using resting state and movie watching fMRI data from the Human Connectome Project, we reveal that resting state functional connectivity (RSFC) profiles can be used to predict cortex-wide ISC. Similar region-level analyses demonstrate that the levels of ISC exhibited by brain regions during movie watching are associated with their connectivity to other regions at rest, and that the nature of these connectivity-activity relationships varies as a function of regional roles in sensory information processing. Finally, we show that an individual's unique spatial distribution of ISC, independent of its magnitude, is also related to their RSFC profile. These findings contextualize reports of localized individual differences in ISC as potentially reflecting larger, network-level alterations in resting brain function and detail how the brain's ability to process complex sensory information is linked to its baseline functional organization.

Distinct modes of functional connectivity induced by movie-watching.

Project description:A fundamental question in systems neuroscience is how endogenous neuronal activity self-organizes during particular brain states. Recent neuroimaging studies have demonstrated systematic relationships between resting-state and task-induced functional connectivity (FC). In particular, continuous task studies, such as movie watching, speak to alterations in coupling among cortical regions and enhanced fluctuations in FC compared to the resting-state. This suggests that FC may reflect systematic and large-scale reorganization of functionally integrated responses while subjects are watching movies. In this study, we characterized fluctuations in FC during resting-state and movie-watching conditions. We found that the FC patterns induced systematically by movie-watching can be explained with a single principal component. These condition-specific FC fluctuations overlapped with inter-subject synchronization patterns in occipital and temporal brain regions. However, unlike inter-subject synchronization, condition-specific FC patterns were characterized by increased correlations within frontal brain regions and reduced correlations between frontal-parietal brain regions. We investigated these condition-specific functional variations as a shorter time scale, using time-resolved FC. The time-resolved FC showed condition-specificity over time; notably when subjects watched both the same and different movies. To explain self-organisation of global FC through the alterations in local dynamics, we used a large-scale computational model. We found that condition-specific reorganization of FC could be explained by local changes that engendered changes in FC among higher-order association regions, mainly in frontal and parietal cortices.

Anxiety and amygdala connectivity during movie-watching.

Project description:Rodent and human studies have implicated an amygdala-prefrontal circuit during threat processing. One possibility is that while amygdala activity underlies core features of anxiety (e.g. detection of salient information), prefrontal cortices (i.e. dorsomedial prefrontal/anterior cingulate cortex) entrain its responsiveness. To date, this has been established in tightly controlled paradigms (predominantly using static face perception tasks) but has not been extended to more naturalistic settings. Consequently, using 'movie fMRI'-in which participants watch ecologically-rich movie stimuli rather than constrained cognitive tasks-we sought to test whether individual differences in anxiety correlate with the degree of face-dependent amygdala-prefrontal coupling in two independent samples. Analyses suggested increased face-dependent superior parietal activation and decreased speech-dependent auditory cortex activation as a function of anxiety. However, we failed to find evidence for anxiety-dependent connectivity, neither in our stimulus-dependent or -independent analyses. Our findings suggest that work using experimentally constrained tasks may not replicate in more ecologically valid settings and, moreover, highlight the importance of testing the generalizability of neuroimaging findings outside of the original context.

Thalamo-cortical inter-subject functional correlation during movie watching across the adult lifespan.

Project description:An increasing number of studies have shown that the functional interactions between the thalamus and cerebral cortices play an important role in cognitive function and are influenced by age. Previous studies have revealed age-related changes in the thalamo-cortical system within individuals, while neglecting differences between individuals. Here, we characterized inter-subject functional correlation (ISFC) between the thalamus and several cortical brain networks in 500 healthy participants aged 18-87 years old from the Cambridge Centre for Aging and Neuroscience (Cam-CAN) cohort using movie-watching state fMRI data. General linear models (GLM) were performed to assess age-related changes in ISFC of thalamo-cortical networks and the relationship between ISFC and fluid intelligence. We found significant age-related decreases in ISFC between the posterior thalamus (e.g., ventral posterior nucleus and pulvinar) and the attentional network, sensorimotor network, and visual network (FDR correction with p < 0.05). Meanwhile, the ISFC between the thalamus (mainly the mediodorsal nucleus and ventral thalamic nuclei) and higher-order cortical networks, including the default mode network, salience network and control network, showed complex changes with age. Furthermore, the altered ISFC of thalamo-cortical networks was positively correlated with decreased fluid intelligence (FDR correction with p < 0.05). Overall, our results provide further evidence that alterations in the functional integrity of the thalamo-cortical system might play an important role in cognitive decline during aging.

Brain dynamics in ASD during movie-watching show idiosyncratic functional integration and segregation.

Project description:To refine our understanding of autism spectrum disorders (ASD), studies of the brain in dynamic, multimodal and ecological experimental settings are required. One way to achieve this is to compare the neural responses of ASD and typically developing (TD) individuals when viewing a naturalistic movie, but the temporal complexity of the stimulus hampers this task, and the presence of intrinsic functional connectivity (FC) may overshadow movie-driven fluctuations. Here, we detected inter-subject functional correlation (ISFC) transients to disentangle movie-induced functional changes from underlying resting-state activity while probing FC dynamically. When considering the number of significant ISFC excursions triggered by the movie across the brain, connections between remote functional modules were more heterogeneously engaged in the ASD population. Dynamically tracking the temporal profiles of those ISFC changes and tying them to specific movie subparts, this idiosyncrasy in ASD responses was then shown to involve functional integration and segregation mechanisms such as response inhibition, background suppression, or multisensory integration, while low-level visual processing was spared. Through the application of a new framework for the study of dynamic experimental paradigms, our results reveal a temporally localized idiosyncrasy in ASD responses, specific to short-lived episodes of long-range functional interplays.

Anxiety Shapes Amygdala-Prefrontal Dynamics During Movie Watching.

Project description:BackgroundA well-characterized amygdala-dorsomedial prefrontal circuit is thought to be crucial for threat vigilance during anxiety. However, engagement of this circuitry within relatively naturalistic paradigms remains unresolved.MethodsUsing an open functional magnetic resonance imaging dataset (Cambridge Centre for Ageing Neuroscience; n = 630), we sought to investigate whether anxiety correlates with dynamic connectivity between the amygdala and dorsomedial prefrontal cortex during movie watching.ResultsUsing an intersubject representational similarity approach, we saw no effect of anxiety when comparing pairwise similarities of dynamic connectivity across the entire movie. However, preregistered analyses demonstrated a relationship between anxiety, amygdala-prefrontal dynamics, and anxiogenic features of the movie (canonical suspense ratings). Our results indicated that amygdala-prefrontal circuitry was modulated by suspense in low-anxiety individuals but was less sensitive to suspense in high-anxiety individuals. We suggest that this could also be related to slowed habituation or amplified anticipation. Moreover, a measure of threat-relevant attentional bias (accuracy/reaction time to fearful faces) demonstrated an association with connectivity and suspense.ConclusionsOverall, this study demonstrated the presence of anxiety-relevant differences in connectivity during movie watching, varying with anxiogenic features of the movie. Mechanistically, exactly how and when these differences arise remains an opportunity for future research.

Cross-movie prediction of individualized functional topography.

Project description:Participant-specific, functionally defined brain areas are usually mapped with functional localizers and estimated by making contrasts between responses to single categories of input. Naturalistic stimuli engage multiple brain systems in parallel, provide more ecologically plausible estimates of real-world statistics, and are friendly to special populations. The current study shows that cortical functional topographies in individual participants can be estimated with high fidelity from naturalistic stimuli. Importantly, we demonstrate that robust, individualized estimates can be obtained even when participants watched different movies, were scanned with different parameters/scanners, and were sampled from different institutes across the world. Our results create a foundation for future studies that allow researchers to estimate a broad range of functional topographies based on naturalistic movies and a normative database, making it possible to integrate high-level cognitive functions across datasets from laboratories worldwide.

Mapping patterns of thought onto brain activity during movie-watching.

Project description:Movie-watching is a central aspect of our lives and an important paradigm for understanding the brain mechanisms behind cognition as it occurs in daily life. Contemporary views of ongoing thought argue that the ability to make sense of events in the 'here and now' depend on the neural processing of incoming sensory information by auditory and visual cortex, which are kept in check by systems in association cortex. However, we currently lack an understanding of how patterns of ongoing thoughts map onto the different brain systems when we watch a film, partly because methods of sampling experience disrupt the dynamics of brain activity and the experience of movie-watching. Our study established a novel method for mapping thought patterns onto the brain activity that occurs at different moments of a film, which does not disrupt the time course of brain activity or the movie-watching experience. We found moments when experience sampling highlighted engagement with multi-sensory features of the film or highlighted thoughts with episodic features, regions of sensory cortex were more active and subsequent memory for events in the movie was better-on the other hand, periods of intrusive distraction emerged when activity in regions of association cortex within the frontoparietal system was reduced. These results highlight the critical role sensory systems play in the multi-modal experience of movie-watching and provide evidence for the role of association cortex in reducing distraction when we watch films.

The Relations Among Anxiety, Movie-Watching, and in-Scanner Motion.

Project description:Movie-watching fMRI has emerged as a theoretically viable platform for studying neurobiological substrates of affective states and emotional disorders such as pathological anxiety. However, using anxiety-inducing movie clips to probe relevant states impacted by psychopathology could risk exacerbating in-scanner movement, decreasing signal quality/quantity and thus statistical power. This could be especially problematic in target populations such as children who typically move more in the scanner. Consequently, we assessed: (1) the extent to which an anxiety-inducing movie clip altered in-scanner data quality (movement, censoring, and DVARS) in a pediatric sample with and without anxiety disorders (n = 78); and (2) investigated interactions between anxiety symptoms and movie-attenuated motion in a highly powered, transdiagnostic pediatric sample (n = 2058). Our results suggest anxiogenic movie-watching in fact reduces in-scanner movement compared to resting-state, increasing the quantity/quality of data. In one measure, pathological anxiety appeared to impact movie-attenuated motion, but the effect was small. Given potential boosts to data quality, future developmental neuroimaging studies of anxiety may benefit from the use of movie paradigms.

Temporal fluctuations in the brain's modular architecture during movie-watching.

Project description:Brain networks are flexible and reconfigure over time to support ongoing cognitive processes. However, tracking statistically meaningful reconfigurations across time has proven difficult. This has to do largely with issues related to sampling variability, making instantaneous estimation of network organization difficult, along with increased reliance on task-free (cognitively unconstrained) experimental paradigms, limiting the ability to interpret the origin of changes in network structure over time. Here, we address these challenges using time-varying network analysis in conjunction with a naturalistic viewing paradigm. Specifically, we developed a measure of inter-subject network similarity and used this measure as a coincidence filter to identify synchronous fluctuations in network organization across individuals. Applied to movie-watching data, we found that periods of high inter-subject similarity coincided with reductions in network modularity and increased connectivity between cognitive systems. In contrast, low inter-subject similarity was associated with increased system segregation and more rest-like architectures. We then used a data-driven approach to uncover clusters of functional connections that follow similar trajectories over time and are more strongly correlated during movie-watching than at rest. Finally, we show that synchronous fluctuations in network architecture over time can be linked to a subset of features in the movie. Our findings link dynamic fluctuations in network integration and segregation to patterns of inter-subject similarity, and suggest that moment-to-moment fluctuations in functional connectivity reflect shared cognitive processing across individuals.
