The complete mitochondrial genome of Cerceris bucculata (Hymenoptera: Crabronidae).

ABSTRACT: The complete mitochondrial genome of the Cerceris bucculata (A. Costa, 1860) (Hymenoptera: Crabronidae) was obtained via next-generation sequencing. This mitochondrial genome is 16178 bp in length with 37 classical eukaryotic mitochondrial genes and an A + T-rich region. All the 13 PCGs begin with typical ATN codons. Among them, eleven PCG genes terminate with TAA, two with T--. All of the 22 tRNA genes, ranging from 58 to 72 bp with typical cloverleaf structure except for trnS1, whose dihydrouridine (DHU) arm forms a simple loop. Phylogenetic analysis highly supported Crabronidae shown as sister group of anthophila bees.

Wasp Size and Prey Load in Cerceris fumipennis (Hymenoptera, Crabronidae): Implications for Biosurveillance of Pest Buprestidae.

Project description:The relationship between predator and prey size was studied in the buprestid hunting wasp Cerceris fumipennis Say in eight widely distributed nesting aggregations in North Carolina, USA. Initial work indicated a significant linear relationship between wasp head width and wasp wet weight; thus, head width was used to estimate wasp body mass in subsequent studies. Prey loads of hunting females was studied by measuring the head width of the wasp, then identifying and weighing the prey item brought back to the nest. There was significant variation in wasp size among nesting aggregations; the average estimated wasp body mass in one site was double that in another. Prey weight varied with wasp weight, but larger wasps had a slight tendency to carry proportionally larger prey. Beetles captured by large wasps (≥120 mg) were significantly more variable in weight than those taken by small wasps (<80 mg). All but the smallest wasps could carry more than their own body weight. Prey loads ranged from 4.8⁻150.2% of wasp weight. Evidence suggests that small wasps bring back more of the economically important buprestid genus Agrilus and thus would be most efficient in biosurveillance for pest buprestids.

Temporal and spatial dynamics of the emerald ash borer invasion in Connecticut as shown by the native digging wasp Cerceris fumipennis (Hymenoptera: Crabronidae).

Project description:Detecting and monitoring populations of the invasive emerald ash borer (EAB) is crucial to successful management of the pest and evaluation of its ecological impacts. However, the beetle's cryptic habit makes accurate monitoring costly and time-consuming. Biosurveillance takes advantage of the foraging effort of a predatory wasp Cerceris fumipennis (Hymenoptera: Crabronidae). This native, solitary, ground-nesting hunting wasp hunts adult buprestid beetles to provision its brood cells. By intercepting the hunting wasps, we can learn which species of buprestids are in the surrounding forest. The resulting data provides information on the presence and relative abundance of invasive buprestids like EAB which can supplement other monitoring efforts. In this paper we share results of ten years of biosurveillance surveys of the EAB in Connecticut. Among 112 sites, we observed EAB populations; from first detection, through the population peak and then through to the population crash, matching patterns observed in other regions of the United States. We also observed the spread of the EAB relative abundance as it moved through the state following an invasion front starting in New Haven, Co. The average time from first detection to population crash was nine years. On average, populations peaked three years after first detection, and remained at peak levels for three to four years. Population decline was gradual and took another three to four years. Notably, no evidence of a second introduction to Connecticut was seen with proportional abundance increasing over time after expanding outward from the introduction point. These results corroborate other traditional monitoring efforts in the eastern U.S. and provide independent validation of predicted population dynamics in ash stands.

Project description:The complete mitochondrial genome of Bombus breviceps, which belongs to the subgenus of Alpigenobombus in Bombus genus, was sequenced. This circular mitogenome is 16,743 bp (83.5% AT) in length, with two rRNA genes, 22 tRNA genes, and 13 protein-coding genes. All protein-coding genes are initiated with the common invertebrate initiation codon except TTA for ND2, and terminated by the typical stop codon TAA or TAG. tRNA-Ser (TCT) lacks a dihydrouridine (DHU) arm while other tRNA genes form a cloverleaf structure. Phylogenetic analysis shows that genus Bombus has a closer relationship with genus Melipona than any other genera in Apidae family, and B. breviceps clusters closer to B. lapidarius than any other bumblebees with sequenced mitogenome.

This circular mitogenome is 17,966 bp (86.7% of AT) in length, which contains 22 tRNA genes, 13 protein-coding genes and two rRNA genes. All protein-coding genes are initiated with the typical ATN codon and terminated by TAA codon except an incomplete stop codon T for ND4 gene. tRNA-Ser (AGN) lacks a dihydrouridine (DHU) arm while other tRNA genes form a typical cloverleaf structure. The complete mitochondrial genome of B. consobrinus will be useful to understand the inter-species relationship and genetic diversity of bumblebees.

The complete mitochondrial genome of Leptopilina syphax (Hymenoptera: Figitidae).

Project description:Leptopilina syphax (Hymenoptera: Figitidae) is a newly recorded species of parasitic wasp, and it attacks the larval stage of Drosophilidae, mainly the Drosophila species. Few works have been done in the basic study of L. syphax, including the data of mitochondrial genome. In this study, the complete mitochondrial genome of L. syphax (GeneBank accession number: MT649407) was sequenced using Illumina HiSeq X Ten system. The mitochondrial genome is 15,882bp long and comprises 13 protein-coding genes, 2 ribosomal RNA genes and 22 transfer RNA genes. Meanwhile, 26 genes are in majority strand, and the remaining 11 genes are in minority strand. The overall base composition is 41.7% for A, 6.0% for G, 13.6% for C, and 38.7% for T, with an A + T content of 80.4%, respectively. We also performed a phylogenetic analysis with other known mitochondrial genomes of some parasitic wasps. The results show that L. syphax is closely related to L. boulardi, which is another Drosophila parasitoid.

Project description:Trichopria drosophilae (Hymenoptera: Diapriidae) is an important pupal endoparasitoid of Drosophila species, which has been found to be an ideal biocontrol agent to D. suzukii. In this study, the complete mitochondrial genome of T. drosophilae (GeneBank accession number: MN966974) was sequenced using Illumina HiSeq X Ten system. The mitochondrial genome is 16,375 bp long and comprises 13 protein-coding genes, 22 transfer RNA genes and 2 ribosomal RNA genes. Among them, 24 genes are in majority strand, while the others are in minority strand. The nucleotide composition of A, G, C, T is 44.9%, 6.4%, 5.6%, 43.2% respectively. We also performed a phylogenetic analysis with other known mitochondrial genomes within four families that have been shown to parasitize drosophilid species. The result shows that T. drosophilae is closely related to Ismarus sp.

Complete mitochondrial genome sequence of Tapinoma melanocephalum (Hymenoptera: Formicidae).

Project description:Tapinoma melanocephalum is a ubiquitous invasive species and widely distributed in subtropical and tropical regions around the world. Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of T. melanocephalum. This mitogenome was 15,499 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA unit genes (rRNAs). Compared to other Formicidae species, gene order of T. melanocephalum was not conserved and one tRNA cluster trnW-trnC-trnY converted to trnW-trnY-trnC. The whole mitogenome exhibited heavy AT nucleotide bias (79.5%). All PCGs started with the standard ATN codons. Except for cox1 and nad5 end with the incomplete codon T-, all PCGs terminated with the stop codon TAA. Phylogenetic analysis showed that T. melanocephalum got together with three same subfamily Dolichoderinae species and one Dorylinae species, indicating the close relationship of Dolichoderinae and Dorylinae.

The complete mitochondrial genome of Aphidius colemani (Hymenoptera: Braconidae: Aphidiinae).

Project description:The genome-level features are crucial genetic resources for species identification and phylogenetic analysis. Here, the complete mitochondrial genome of Aphidius colemani Viereck 1912 (Hymenoptera: Braconidae: Aphidiinae) was sequenced, determined and analyzed. The circular genome is 16,372 bp in length with an overall base composition of 38.9% for A, 46.2% for T, 6.7% for C, and 8.2% for G. The mitochondrial genome of A. colemani contained 13 protein-coding genes that initiated by the ATN codon, 22 transfer RNA genes, two ribosomal RNA genes (rRNAs), and a control region (CR). It shared the same gene arrangement patterns that occurred in two tRNA clusters of trnI-trnQ-trnM and trnW-trnC-trnY with Aphidius gifuensis. Phylogenetic analyses using Bayesian inference and Maximum-likelihood methods supported that the two species of Aphidiinae formed a clade and sister to other subfamilies of Braconidae.
