The first complete mitochondrial genome of Euroleon coreanus (Okamoto, 1926) (Neuroptera: Myrmeleontidae) and its phylogeny.

ABSTRACT: The first complete mitochondrial genome of Euroleon coreanus (Okamoto, 1926) was 15,797 bp in length, and contained 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and the control region. Compared to the classic insect mitochondrial genome, E. coreanus showed a gene rearrangement of ND2-C-W-Y-COX1. The overall AT content of the mitochondrial genome was 75.5%. The monophyly of Ascalaphidae, Myrmeleontidae, Nemopteridae, Nymphidae, and Psychopsidae was supported in both BI and ML trees. And E. coreanus was a sister clade to the clade of genus Myrmeleon.

Project description:The complete mitochondrial genome (mitogenome) of Bullanga florida (Neuroptera: Myrmeleontidae) was determined. The entire sequence is 15,937 bp in length which contains 13 protein-coding genes (PCGs), 22 tRNAs, two rRNAs and one non-coding AT-rich region. The overall A + T content of mitogenome is 73.3%. The gene order and arrangement are similar to other Neuroptera mitogenomes. Thirteen PCGs start with standard ATN initiation codons and stop with termination codons TAA or T. All tRNA genes have a typical clover-leaf secondary structure except for tRNASer (AGN), whose dihydrouridine (DHU) arm do not form a stable stem-loop structure. Two rRNA genes (rrnL and rrnS) are 1325 bp and 776 bp in size, respectively. The phylogenetic analysis is based on the amino acid sequences of 13 PCGs indicates that B. florida is a sister group to Epacanthaclisis banks, and Myrmeleontiformia is the monophyletic group.

Project description:In the holometabolous insect order Neuroptera (lacewings), the cosmopolitan Myrmeleontidae (antlions) are the most species-rich family, while the closely related Nymphidae (split-footed lacewings) are a small endemic family from the Australian-Malesian region. Both families belong to the suborder Myrmeleontiformia, within which controversial hypotheses on the interfamilial phylogenetic relationships exist. Herein, we describe the complete mitochondrial (mt) genomes of an antlion (Myrmeleon immanis Walker, 1853) and a split-footed lacewing (Nymphes myrmeleonoides Leach, 1814), representing the first mt genomes for both families. These mt genomes are relatively small (respectively composed of 15,799 and 15,713 bp) compared to other lacewing mt genomes, and comprise 37 genes (13 protein coding genes, 22 tRNA genes and two rRNA genes). The arrangement of these two mt genomes is the same as in most derived Neuroptera mt genomes previously sequenced, specifically with a translocation of trnC. The start codons of all PCGs are started by ATN, with an exception of cox1, which is ACG in the M. immanis mt genome and TCG in N. myrmeleonoides. All tRNA genes have a typical clover-leaf structure of mitochondrial tRNA, with the exception of trnS1(AGN). The secondary structures of rrnL and rrnS are similar with those proposed insects and the domain I contains nine helices rather than eight helices, which is common within Neuroptera. A phylogenetic analysis based on the mt genomic data for all Neuropterida sequenced thus far, supports the monophyly of Myrmeleontiformia and the sister relationship between Ascalaphidae and Myrmeleontidae.

Project description:The complete mitochondrial genome (mitogenome) of Conwentzia sinica Yang, 1974 was sequenced and analyzed. The sequenced mitogenome is 15,153 bp in size, including 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes, and one control region. Thirteen PCGs use ATN as the start codon. Most of PCGs terminate with TAA codons but ND5, ND4 with a single T residue, and ND3 terminates with TAG. The lrRNA gene is 1290 bp. The measured length of srRNA gene is 782 bp. Twenty-two tRNA genes possess the typical clover-leaf secondary structure except for tRNASer(AGN) . The phylogenetic result supports the monophyly of the family Coniopterygidae and a closer relationship between Conwentzia and Coniopteryx.

The complete mitochondrial genome is a typical double-stranded circular molecule of 16,343 bp (GenBank accession number: MT268963) comprising of 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and a control region. The gene order is identical to that of the putative ancestral arrangement of insects and other lacewings. All protein-coding genes initiate with ATN, except COI use CGA as start codons and terminate with TAG or TAA, expect ND5 and ND4 use TA- or a single T-- residue as the stop codon. All tRNAs, ranging from 63 to 72 bp, can be folded into typical clover-leaf secondary structure except for tRNASer(AGN) , in which the dihydrouridine (DHU) arm did not form a stable stem-loop structure. The control region is 1433 bp long with an A + T content of 91.4%. In the sampled families of Neuroptera, each family showed a monophyletic cluster and Polystoechotidae + Rapismatidae, Hemerobiidae + (Chrysopidae + (Polystoechotidae + Rapismatidae)), are recovered in phylogenetic analyses with high supports.

The gene order is identical to that of the putative ancestral arrangement of insects and other lacewings. 13 protein-coding genes (PCGs) possessed common triplet initiation codons ATN except ND1 possessed TTG and mostly terminated with TAN codons except for ND5 and ND4 with a single T residue adjacent to a downstream tRNA gene. All of the 22 tRNAs, ranging from 63 to 72 bp, can be folded into classic clover-leaf secondary structure except for tRNASer(AGN) , in which the dihydrouridine (DHU) arm did not form a stable stem-loop structure. The control region is 1416 bp long with an A + T content of 90.3%. In the sampled families of Neuroptera, each family showed a monophyletic cluster and Polystoechotidae + Rapismatidae, Osmylidae + the remaining families, Hemerobiidae + (Chrysopidae + (Polystoechotidae + Rapismatidae)) are recovered in phylogenetic analyses with high supports.

Complete mitochondrial genome of Asiagomphus coreanus (Odonata: Gomphidae), which is endemic to South Korea.

Project description:Asiagomphus coreanus (Doi & Okumura, 1937) belongs to the family Gomphidae in the order Odonata, and has been listed as an endemic species in South Korea. Here, we assembled its complete mitochondrial genome (mitogenome) which is 15,649 base pairs (bp) in length. The A. coreanus mitogeneome consists of a typical set of genes [13 protein-coding genes (PCGs), 2 ribosomal RNA (rRNA) genes, and 22 transfer RNA (tRNA) genes] and one major non-coding A + T-rich region which is 846 bp long. The gene arrangement of the species was identical to that of commonly found in the majority of the insects. Phylogenetic analyses using the concatenated sequences of 13 PCGs and two rRNA genes of the representative odonate mitogenomes by Bayesian inference method revealed that A. coreanus belongs to the Gomphidae family with a strong nodal support (Bayesian posterior probabilities = 1). Unlike previous phylogenetic analyses (with regards to suborder relationships) the suborder Anisozygoptera-which was represented by a single species, Epiophlebia superstes-was placed as the sister to Zygoptera.

Project description:The complete mitochondrial (mt) genome of Hemerobius simulans Walker (Neuroptera, Hemerobiidae) is reported in this work. The whole mt genome is 17,985 bp long and contains 37canonical genes and an A + T-rich region, which is the same with insect ancestral mt genome arrangement. All 13 PCGs used the typical ATN as initiation codons. The control region of H. simulans mt genome is 1,416 bp long and the base composition is 90.0% of A + T. The phylogenetic analysis revealed that Hemerobiidae was monophyletic and was the sister group to Chrysopidae.

However, dedicated samplings in the Mediterranean region carried out in the last years suggest that the state of knowledge of the representatives of this family occurring in outhern Europe is far from ideal.
