Coordinated gas release among the physostomous fish sprat (Sprattus sprattus).

ABSTRACT: Previous experimental studies suggest that the production of sound associated with expelling gas from an open swimbladder may play a role in communication. This would suggest non-random gas release. We used deployed echosounders to study patterns of gas release among a fjord population of sprat (Sprattus sprattus). The echosounder records concurrently revealed individual fish and their release of gas. The gas release primarily occurred at night, partly following recurrent temporal patterns, but also varying between nights. In testing for non-randomness, we formulated a data-driven simulation approach. Non-random gas release scaled with the length of the analyzed time intervals from 1 min to 6 h, and above 30 min the release events in more than 50% of the intervals were significantly connected.

Emergence and repeatability of leadership and coordinated motion in fish shoals.

Project description:Studies of self-organizing groups like schools of fish or flocks of birds have sought to uncover the behavioral rules individuals use (local-level interactions) to coordinate their motion (global-level patterns). However, empirical studies tend to focus on short-term or one-off observations where coordination has already been established or describe transitions between different coordinated states. As a result, we have a poor understanding of how behavioral rules develop and are maintained in groups. Here, we study the emergence and repeatability of coordinated motion in shoals of stickleback fish (Gasterosteus aculeatus). Shoals were introduced to a simple environment, where their spatio-temporal position was deduced via video analysis. Using directional correlation between fish velocities and wavelet analysis of fish positions, we demonstrate how shoals that are initially uncoordinated in their motion quickly transition to a coordinated state with defined individual leader-follower roles. The identities of leaders and followers were repeatable across two trials, and coordination was reached more quickly during the second trial and by groups of fish with higher activity levels (tested before trials). The rapid emergence of coordinated motion and repeatability of social roles in stickleback fish shoals may act to reduce uncertainty of social interactions in the wild, where individuals live in a system with high fission-fusion dynamics and non-random patterns of association.

Barometric-pumping controls fugitive gas emissions from a vadose zone natural gas release.

Project description:Subsurface natural gas release from leaking oil and gas wells is a major environmental concern. Gas migration can cause aquifer contamination, explosive conditions in soil gas, and greenhouse gas emissions. Gas migration is controlled by complex interacting processes, thus constraining the distribution and magnitude of "fugitive gas" emissions remains a challenge. We simulated wellbore leakage in the vadose zone through a controlled release experiment and demonstrate that fugitive gas emissions can be directly influenced by barometric pressure changes. Decreases in barometric-pressure led to surface gas breakthroughs (>20-fold increase in <24 hours), even in the presence of low-permeability surficial soils. Current monitoring strategies do not consider the effect of barometric pressure changes on gas migration and may not provide adequate estimates of fugitive gas emissions. Frequent or continuous monitoring is needed to accurately detect and quantify fugitive gas emissions at oil and gas sites with a deep water table.

Genetic analysis redraws the management boundaries for the European sprat.

Project description:Sustainable fisheries management requires detailed knowledge of population genetic structure. The European sprat is an important commercial fish distributed from Morocco to the Arctic circle, Baltic, Mediterranean, and Black seas. Prior to 2018, annual catch advice on sprat from the International Council for the Exploration of the Sea (ICES) was based on five putative stocks: (a) North Sea, (b) Kattegat-Skagerrak and Norwegian fjords, (c) Baltic Sea, (d) West of Scotland-southern Celtic Seas, and (e) English Channel. However, there were concerns that the sprat advice on stock size estimates management plan inadequately reflected the underlying biological units. Here, we used ddRAD sequencing to develop 91 SNPs that were thereafter used to genotype approximately 2,500 fish from 40 locations. Three highly distinct and relatively homogenous genetic groups were identified: (a) Norwegian fjords; (b) Northeast Atlantic including the North Sea, Kattegat-Skagerrak, Celtic Sea, and Bay of Biscay; and (c) Baltic Sea. Evidence of genetic admixture and possibly physical mixing was detected in samples collected from the transition zone between the North and Baltic seas, but not between any of the other groups. These results have already been implemented by ICES with the decision to merge the North Sea and the Kattegat-Skagerrak sprat to be assessed as a single unit, thus demonstrating that genetic data can be rapidly absorbed to align harvest regimes and biological units.

Establishment of a fish model to study gas-bubble lesions.

Project description:Decompression sickness (DCS) is a clinical syndrome caused by the formation of systemic intravascular and extravascular gas bubbles. The presence of these bubbles in blood vessels is known as gas embolism. DCS has been described in humans and animals such as sea turtles and cetaceans. To delve deeper into DCS, experimental models in terrestrial mammals subjected to compression/decompression in a hyperbaric chamber have been used. Fish can suffer from gas bubble disease (GBD), characterized by the formation of intravascular and extravascular systemic gas bubbles, similarly to that observed in DCS. Given these similarities and the fact that fish develop this disease naturally in supersaturated water, they could be used as an alternative experimental model for the study of the pathophysiological aspect of gas bubbles. The objective of this study was to obtain a reproducible model for GBD in fish by an engineering system and a complete pathological study, validating this model for the study of the physiopathology of gas related lesions in DCS. A massive and severe GBD was achieved by exposing the fish for 18 h to TDG values of 162-163%, characterized by the presence of severe hemorrhages and the visualization of massive quantities of macroscopic and microscopic gas bubbles, systemically distributed, circulating through different large vessels of experimental fish. These pathological findings were the same as those described in small mammals for the study of explosive DCS by hyperbaric chamber, validating the translational usefulness of this first fish model to study the gas-bubbles lesions associated to DCS from a pathological standpoint.

Fish can use coordinated fin motions to recapture their own vortex wake energy.

Project description:During swimming, many fishes use pectoral fins for propulsion and, in the process, move substantial amounts of water rearward. However, the effect that this upstream wake has on the caudal fin remains largely unexplored. By coordinating motions of the caudal fin with the pectoral fins, fishes have the potential to create constructive flow interactions which may act to partially recapture the upstream energy lost in the pectoral fin wake. Using experimentally derived velocity and pressure fields for the silver mojarra (Eucinostomus argenteus), we show that pectoral-caudal fin (PCF) coordination enables the circulation and interception of pectoral fin wake vortices by the caudal fin. This acts to transfer energy to the caudal fin and enhance its hydrodynamic efficiency at swimming speeds where this behaviour occurs. We also find that mojarras commonly use PCF coordination in nature. The results offer new insights into the evolutionary drivers and behavioural plasticity of fish swimming as well as for developing more capable bioinspired underwater vehicles.

Single-nucleus RNA-seq and FISH identify coordinated transcriptional activity in mammalian myofibers.

Project description:Skeletal muscle fibers are large syncytia but it is currently unknown whether gene expression is coordinately regulated in their numerous nuclei. Here we show by snRNA-seq and snATAC-seq that slow, fast, myotendinous and neuromuscular junction myonuclei each have different transcriptional programs, associated with distinct chromatin states and combinations of transcription factors. In adult mice, identified myofiber types predominantly express either a slow or one of the three fast isoforms of Myosin heavy chain (MYH) proteins, while a small number of hybrid fibers can express more than one MYH. By snRNA-seq and FISH, we show that the majority of myonuclei within a myofiber are synchronized, coordinately expressing only one fast Myh isoform with a preferential panel of muscle-specific genes. Importantly, this coordination of expression occurs early during post-natal development and depends on innervation. These findings highlight a previously undefined mechanism of coordination of gene expression in a syncytium.

Binding, Release and Functionalization of Intact Pnictogen Tetrahedra Coordinated to Dicopper Complexes.

Project description:The bridging MeCN ligand in the dicopper(I) complexes [(DPFN)Cu2 (μ,η1 : η1 -MeCN)][X]2 (X=weakly coordinating anion, NTf2 (1 a), FAl[OC6 F10 (C6 F5 )]3 (1 b), Al[OC(CF3 )3 ]4 (1 c)) was replaced by white phosphorus (P4 ) or yellow arsenic (As4 ) to yield [(DPFN)Cu2 (μ,η2 : η2 -E4 )][X]2 (E=P (2 a-c), As (3 a-c)). The molecular structures in the solid state reveal novel coordination modes for E4 tetrahedra bonded to coinage metal ions. Experimental data and quantum chemical computations provide information concerning perturbations to the bonding in coordinated E4 tetrahedra. Reactions with N-heterocyclic carbenes (NHCs) led to replacement of the E4 tetrahedra with release of P4 or As4 and formation of [(DPFN)Cu2 (μ,η1 : η1 -Me NHC)][X]2 (4 a,b) or to an opening of one E-E bond leading to an unusual E4 butterfly structural motif in [(DPFN)Cu2 (μ,η1 : η1 -E4 Dipp NHC)][X]2 (E=P (5 a,b), E=As (6)). With a cyclic alkyl amino carbene (Et CAAC), cleavage of two As-As bonds was observed to give two isomers of [(DPFN)Cu2 (μ,η2 : η2 -As4 Et CAAC)][X]2 (7 a,b) with an unusual As4 -triangle+1 unit.

Tilapia fish microbial spoilage monitored by a single optical gas sensor.

Project description:As consumption of fish and fish-based foods increases, non-destructive monitoring of fish freshness also becomes more prominent. Fish products are very perishable and prone to microbiological growth, not always easily detected by organoleptic evaluation. The analysis of the headspace of fish specimens through gas sensing is an interesting approach to monitor fish freshness. Here we report a gas sensing method for monitoring Tilapia fish spoilage based on the application of a single gas sensitive gel material coupled to an optical electronic nose. The optical signals of the sensor and the extent of bacterial growth were followed over time, and results indicated good correlation between the two determinations, which suggests the potential application of this simple and low cost system for Tilapia fish freshness monitoring.

Diastereomeric resolution directed towards chirality determination focussing on gas-phase energetics of coordinated sodium dissociation.

Project description:Defining chiral centres is addressed by introducing a pair of chiral auxiliary groups. Ions of diastereomeric pairs of molecules could be distinguished utilising energy-resolved mass spectrometry, and the applicability of the method to a series of compounds carrying amine, carboxylic acid, alcohol, and all the amino acids was verified. The method was further strengthened by distinguishing diastereomeric ions that did not undergo fragmentation. Mass spectrometric evaluation of the dissociation process of adducted sodium cations from the diastereomeric precursors agreed with the theoretical calculations, indicating the potential usefulness of the method for the determination of absolute configurations.
