ABSTRACT: We conducted an online national survey using the South Oaks Gambling Screen (SOGS) on 42,880 people in Japan to identify gambling addiction prevalence differences with regard to gambling type and region. This study included 14,780 individuals (valid responses) who engaged in gambling activities in the past year. There was a large difference in mean SOGS score by gambling type: public gambling and casinos score ≈ 4.0, whereas lotteries (including numbers, etc.) ≈ 1.43. SOGS scores were much higher for those who participated in more than one gambling type than for those who participated in only one type. Regional differences in SOGS scores were also confirmed, with more populous prefectures having lower and less populous having higher SOGS scores. Integrating SOGS scores with existing data concerning hobbies and recreational behaviors across regions indicated that regions characterized by lower SOGS scores were also characterized by greater activity for hobbies and recreational behaviors.

Project description:Recent large genome-wide association studies have identified multiple confident risk loci linked to addiction-associated behavioral traits. Most genetic variants linked to addiction-associated traits lie in noncoding regions of the genome, likely disrupting cis-regulatory element (CRE) function. CREs tend to be highly cell type-specific and may contribute to the functional development of the neural circuits underlying addiction. Yet, a systematic approach for predicting the impact of risk variants on the CREs of specific cell populations is lacking. To dissect the cell types and brain regions underlying addiction-associated traits, we applied stratified linkage disequilibrium score regression to compare genome-wide association studies to genomic regions collected from human and mouse assays for open chromatin, which is associated with CRE activity. We found enrichment of addiction-associated variants in putative CREs marked by open chromatin in neuronal (NeuN+) nuclei collected from multiple prefrontal cortical areas and striatal regions known to play major roles in reward and addiction. To further dissect the cell type-specific basis of addiction-associated traits, we also identified enrichments in human orthologs of open chromatin regions of female and male mouse neuronal subtypes: cortical excitatory, D1, D2, and PV. Last, we developed machine learning models to predict mouse cell type-specific open chromatin, enabling us to further categorize human NeuN+ open chromatin regions into cortical excitatory or striatal D1 and D2 neurons and predict the functional impact of addiction-associated genetic variants. Our results suggest that different neuronal subtypes within the reward system play distinct roles in the variety of traits that contribute to addiction.SIGNIFICANCE STATEMENT We combine statistical genetic and machine learning techniques to find that the predisposition to for nicotine, alcohol, and cannabis use behaviors can be partially explained by genetic variants in conserved regulatory elements within specific brain regions and neuronal subtypes of the reward system. Our computational framework can flexibly integrate open chromatin data across species to screen for putative causal variants in a cell type- and tissue-specific manner for numerous complex traits.

Project description:Investigating neurophysiological markers linked to impulsivity in individuals with gambling and methamphetamine addiction using resting-state EEG data offers valuable insights into the underlying neurophysiological mechanisms associated with impulsivity in individuals with addiction. This study aims to use resting-state EEG to explore the connection between various types of addiction and different aspects of impulsivity. Participants from the methamphetamine, gambling, and healthy control groups (abbreviation: MA, GB, HC) underwent EEG recordings and completed measures of impulsivity. Group differences in trait scores and behavioral tendencies were analyzed. Abnormal connections with node linkage and importance changes were analyzed through the resting-state EEG power spectral and network analyses. Further, relationships between impulsivity scores and connectivity differences in groups were explored through correlation analysis. Finally, these abnormal connections related to impulsivity were tested for their effect of distinguishing individuals with addiction from healthy controls through the ROC analysis. Results revealed that GB displayed the highest trait impulsivity on the overall score, while MA exhibited greater attentional impulsivity. Variations in behavioral impulsivity were reflected in response times. Resting-state EEG analysis showed higher beta power in GB. Specific channel pairs demonstrated abnormal connections and altered connectivity patterns in the beta band, with MA displaying a less efficient network compared to GB. Correlation analyses uncovered associations between impulsivity scores and connectivity, which were influenced by group differences. Furthermore, resting-state EEG connections effectively differentiated individuals with addiction from healthy controls. Overall, this study contributes valuable insights into the neural mechanisms of addiction-related impulsivity, emphasizing the potential of resting-state EEG connections as an important neurophysiological correlate.
