ABSTRACT: Raoultella planticola is a Gram-negative opportunistic bacterial pathogen associated with hospital-acquired infections in humans. Here, we report the complete genome sequence of one Raoultella planticola strain isolated from Canadian wastewater treatment facilities containing one chromosome and four plasmids with four antimicrobial resistance (AMR) genes and four metal resistance gene clusters.

Project description:Raoultella ornithinolytica is an emerging pathogen that causes human infections. We report the isolation and genome sequencing of R. ornithinolytica from an oral swab of a Persian pet cat in Dhaka, Bangladesh. The genome length was 5,375,160 bp, with 55.9% G+C content. It carries putative genes associated with resistance to antibiotics and metals.

Complete Genome Sequence of the First Colistin-Resistant Raoultella electrica Strain.

Project description:We present the full genome sequence of a colistin-resistant Raoultella electrica strain (MIC, >4 μg/mL) that was isolated from the stool of a healthy person living in India. The sequence consists of a chromosome and three plasmids (5,455,992-bp and 98,913-bp, 4,232-bp, and 3,961-bp, respectively). No previously described colistin resistance mechanisms were detected.

Draft genome of Raoultella planticola, a high lead resistance bacterium from industrial wastewater.

Project description:Isolation of heavy metals-resistant bacteria from their original habitat is a crucial step in bioremediation. Six lead (Pb) resistant bacterial strains were isolated and identified utilizing 16S rRNA to be Enterobacter ludwigii FACU 4, Shigella flexneri FACU, Microbacterium paraoxydans FACU, Klebsiella pneumoniae subsp. pneumonia FACU, Raoultella planticola FACU 3 and Staphylococcus xylosus FACU. It was determined that all these strains had their Minimum inhibitory concentration (MIC) to be 2500 ppm except R. planticola FACU 3 has a higher maximum tolerance concentration (MTC) up to 2700 ppm. We evaluated the survival of all six strains on lead stress, the efficiency of biosorption and lead uptake. It was found that R. planticola FACU 3 is the highest MTC and S. xylosus FACU was the lowest MTC in this evaluation. Therefore, transmission electron microscopy (TEM) confirmed the difference between the morphological responses of these two strains to lead stress. These findings led to explore more about the genome of R. planticola FACU 3 using illumine Miseq technology. Draft genome sequence analysis revealed the genome size of 5,648,460 bp and G + C content 55.8% and identified 5526 CDS, 75 tRNA and 4 rRNA. Sequencing technology facilitated the identification of about 47 genes related to resistance to many heavy metals including lead, arsenic, zinc, mercury, nickel, silver and chromium of R. planticola FACU 3 strain. Moreover, genome sequencing identified plant growth-promoting genes (PGPGs) including indole acetic acid (IAA) production, phosphate solubilization, phenazine production, trehalose metabolism and 4-hydroxybenzoate production genes and a lot of antibiotic-resistant genes.

Klebsiella pneumoniae produces no histamine: Raoultella planticola and Raoultella ornithinolytica strains are histamine producers.

Project description:Histamine fish poisoning is caused by histamine-producing bacteria (HPB). Klebsiella pneumoniae and Klebsiella oxytoca are the best-known HPB in fish. However, 22 strains of HPB from fish first identified as K. pneumoniae or K. oxytoca by commercialized systems were later correctly identified as Raoultella planticola (formerly Klebsiella planticola) by additional tests. Similarly, five strains of Raoultella ornithinolytica (formerly Klebsiella ornithinolytica) were isolated from fish as new HPB. R. planticola and R. ornithinolytica strains were equal in their histamine-producing capabilities and were determined to possess the hdc genes, encoding histidine decarboxylase. On the other hand, a collection of 61 strains of K. pneumoniae and 18 strains of K. oxytoca produced no histamine.

Biodegradation of pyrene and benzo[a]pyrene in the liquid matrix and soil by a newly identified Raoultella planticola strain.

Project description:In the current study, the PL7 strain was isolated from soil and identified as Raoultella planticola based on its physiological characteristics and 16S rDNA sequence. By the 10th day, the PL7 strain degraded 52.0% of the pyrene (PYR) content and 50.8% of the benzo[a]pyrene (BaP) content in 20 mg L-1 PYR and 10 mg L-1 BaP in the liquid matrix. The half-life of PYR and BaP by PL7 degradation was 8.59 and 9.46 days, respectively. At pH 8.0, the degradation rates of PYR and BaP by PL7 were significantly higher at 30 °C than at 20 and 40 °C. The degradation ability of PL7 differed in red soil, paddy soil and fluvo-aquic soil; red soil produced the fastest degradation rates. The half-life of PYR and BaP by PL7 degradation in red soil was 21.7 and 11.9 days, respectively; however, without PL7 the half-life of PYR in red soil was 91.2 days. This study demonstrated the significant potential of the PL7 strain for bioremediation applications in the liquid matrix and soil contaminated by PAHs.
