On the recognition of the long neglected Vitis adenoclada Hand.-Mazz. (Vitaceae) from southern China.

ABSTRACT: This study reports the recognition of Vitis adenoclada Hand.-Mazz. from southern China. The species was not recognized in the Flora Reipublicae Ropularis Sinicicae and Flora of China treatments. Recent field studies and examination of herbarium collections including the type material suggest that Vitis adenoclada is morphologically similar to V. heyneana, in their densely arachnoid tomentose abaxial leaves, yet it can be easily distinguished from the latter by its red-purple glandular hairs on the young branches (vs. glandular hairs absent in V. heyneana) and inflorescences usually subtended by a tendril at the base (vs. only occasionally with a tendril in V. heyneana). Vitis adenoclada may be a species of hybrid origin, with the highly tomentose Vitis heyneana as one of the parental species, and likely the glandular-hair bearing V. davidii as the other parental species. Vitis adenoclada is recorded from southern China in Guangdong, Guangxi, Guizhou, Hunan and Zhejiang provinces.

The complete chloroplast genome of Ampelopsis grossedentata (Hand.-Mazz.) W. T. Wang (Family: Vitaceae) and its phylogenetic analysis.

Project description:Ampelopsis grossedentata (Hand.-Mazz.) W. T. Wang is rich in flavonoids and also displays excellent pharmacological activities. The phylogenetic relationship between A. grossedentata and other related Vitaceae family members remains unclear. The chloroplast (cp) genome is a useful model for assessing genome evolution. In this study, we assembled the cp genome of A. grossedentata using the high-throughput Illumina pair-end sequencing data and characterized the genome to providing useful information for future genetic studies. The circular cp genome was 162,147 bp in size, including a large single-copy (LSC) region of 89,244 bp and a small single-copy (SSC) region of 18,439 bp, which were separated by two inverted repeat (IR) regions (27,232 bp each). A total of 135 genes were predicted, including 8 ribosomal RNAs (rRNAs), 37 transfer RNAs (tRNAs), and 90 protein-coding genes (PCGs). Furthermore, phylogenetic analysis revealed that A. grossedentata within Ampelopsis genus and formed a different clade from other three congeneric species. This study provides useful information for future genetic study of A. grossedentata.

Morphology, Structure, and Ontogeny of Trichomes of the Grape Genus (Vitis, Vitaceae).

Project description:Trichomes are widely distributed on surfaces of different organs in the grape genus Vitis and are of taxonomic utility. To explore the morphology, structure and ontogeny of Vitis trichomes, we investigated the diversity and distribution of trichomes in 34 species of Vitis. Two main types of trichomes in Vitis are documented: non-glandular and glandular. Within non-glandular trichomes, ribbon and simple trichomes are found on different vegetative plant organs. The morphology and ontogeny of these types of trichomes are further examined with light microscopy and scanning electron microscopy. The ultrastructure of the glandular trichomes is explored with transmission electron microscopy. The ribbon trichomes are twisted, greatly elongated and unicellular, and this trichome type may be a morphological synapomorphy of Vitis and its closest tropical relative Ampelocissus and Pterisanthes in Vitaceae. The simple trichomes are documented in most species sampled in the genus. The glandular trichomes are multicellular, non-vascularized and composed of both epidermis and subjacent layers. We show that prickles occurring along the stems and petioles of Vitis davidii are modified glandular trichomes. We observed that glandular trichomes of V. romanetii secrete mucilage and volatile substances which trap insectes on the glands. Transmission electron microscopy indicates that metabolic products accumulate in vacuoles, the cytoplasm and intercellular spaces. We infer that glandular trichomes and young prickles are involved in the secretion of these metabolic products and the intercellular spaces may be the places of temporary storage of these secretions.

Diterpenoid Alkaloids from the Aerial Parts of Aconitum flavum Hand.-Mazz.

Project description:Sixteen diterpenoid alkaloids (DAs), including six aconitine-type alkaloids (5 and 9 - 13), seven 7,17-seco-aconitine-type alkaloids (1 - 4, 6 - 8), two napelline-type alkaloids (14 and 15) as well as one veatchine-type alkaloid (16), were isolated from the aerial parts of Aconitum flavum Hand.-Mazz. In which, flavumolines A - D (1 - 4) were four new ones, and flavumoline E (5) was reported as natural compound for the first time. Their chemical structures were elucidated by the analysis of extensive spectroscopic data. The inhibitory activities of these isolates on Cav3.1 low voltage-gated Ca2+ channel, NO production in LPS-activated RAW264.7cells, five human tumor cell lines, as well as acetylcholinesterase (AChE) were tested.

Novel anti-inflammatory activity of chlorojanerin isolated from Sausurrea heteromalla (D. Don) Hand.-Mazz.

Project description:Medicinal plants have shown great promise as a source of novel drug compounds for the treatment of inflammatory disorders. In our search for new entities with anti-inflammatory potential, the extracts of the whole plant of Saussurea heteromalla (family-Asteraceae), collected from Himalayas, were evaluated in the high throughput screen for TNF-α and IL-6 inhibitors. The plant has been found as a new source of chlorojanerin, a guaianolide type of sesquiterpene lactone. This is the first report on the potent anti-inflammatory activity of the compound. Chlorojanerin was shown to be significantly effective in inhibiting TNF-α and IL-6 production in LPS induced THP-1 cells (TNF-α, IC50 = 3µM and IL-6, IC50 = 0.8µM). The compound also blocked TNF-α and IL-6 production from LPS induced human monocytes and synovial cells from RA patients. Chlorojanerin also inhibited the binding of NF-κB in a GFP reporter assay system. Transcriptional profiling of the LPS stimulated THP-1 cells revealed that chlorojanerin exerted its anti-inflammatory effect by inhibiting the expression of genes involved in activating the transcription factor – NF-κB. Real time analysis of these genes validated the effect of chlorojanerin on the classical downstream targets of NF-κB. Thus, this study clearly delineated 8 targets specific for the effect of chlorojanerin on NF-κB induced signaling at the mRNA level. This work is a step towards the isolation and characterization of lead anti-inflammatory agents from the extract of Saussurea heteromalla, which can be developed into better therapeutic molecules targeted towards some specific inflammatory diseases.

Visualized Exploratory Spatiotemporal Analysis of Hand-Foot-Mouth Disease in Southern China.

Project description:ObjectivesIn epidemiological research, major studies have focused on theoretical models; however, few methods of visual analysis have been used to display the patterns of disease distribution.DesignFor this study, a method combining the space-time cube (STC) with space-time scan statistics (STSS) was used to analyze the pattern of incidence of hand-foot-mouth disease (HFMD) in Guangdong Province from May 2008 to March 2009. In this research, STC was used to display the spatiotemporal pattern of incidence of HFMD, and STSS were used to detect the local aggregations of the disease.SettingThe hand-foot-mouth disease data were obtained from Guangdong Province from May 2008 to March 2009, with a total of 68,130 cases.ResultsThe STC analysis revealed a differential pattern of HFMD incidence among different months and cities and also showed that the population density and average precipitation are correlated with the incidence of HFMD. The STSS analysis revealed that the most likely aggregation includes the Shenzhen, Foshan and Dongguan populations, which are the most developed regions in Guangdong Province.ConclusionBoth STC and STSS are efficient tools for the exploratory data analysis of disease transmission. STC clearly displays the spatiotemporal patterns of disease. Using the maximum likelihood ratio, the STSS model precisely locates the most likely aggregation.

De novo transcriptome assembly and EST-SSR markers development for Zelkova schneideriana Hand.-Mazz. (Ulmaceae).

Project description:Zelkova schneideriana Hand.-Mazz. of the Ulmaceae family is a Tertiary relict and economically deciduous tree species endemic to Central and Southern China. In this study, we performed a transcriptome sequencing of Z. schneideriana using high-throughput sequencing approach to detect polymorphic expressed sequence tag-simple sequence repeats (EST-SSR) markers. A total of 3,235 microsatellite loci were detected from 53,517 unigenes. A set of 30 microsatellite markers were randomly selected to validate in 41 individuals from three populations, of which 10 were polymorphic. The number of alleles per locus ranged from 3 to 11. The observed heterozygosity and expected heterozygosity ranged from 0.366 to 0.829 and 0.439 to 0.848, respectively. These polymorphic SSR primers showed good transferability across different Zelkova species, and are valuable for future studies on genetic diversity, conservation, phylogeography, and species delimitation in Z. schneideriana, as well as other Zelkova species.Supplementary informationThe online version contains supplementary material available at 10.1007/s13205-021-02968-5.
