Distinct Particle Films Impacts on Olive Leaf Optical Properties and Plant Physiology.

ABSTRACT: The olive fruit fly is worldwide considered a major harmful pest of the olive agroecosystem. In Italy, the fruit fly infestation is traditionally countered by spraying chemical insecticides (e.g., dimethoate), but due to the recent ban of dimethoate by the Reg EU2019/1090 and the increasing awareness of consumers of food sustainability, the interest in developing chemical-free alternatives to pesticides, such as the use of particle-films, is rising. A field experiment was conducted to assess the effect of different particle films (kaolin-base and zeolitite-base) on leaf gas exchanges and leaf optical properties. Results showed that with the dust accumulation on the leaves' surface, photosynthesis, stomatal conductance, transpiration and water use efficiency were significantly lower in kaolin-treated olive trees compared to those treated with zeolitite and to the control, while olive trees treated with zeolitite showed physiological parameters similar to the untreated plants. Microstructural differences of different particle film on the leaf and olive surfaces emerged by ESEM observations also influenced leaf optical properties. Oils produced by zeolitite-treated plants show higher intensities of gustatory and olfactory secondary flavors compared to kaolin and test oils.

Project description:In this study, developing mucoadhesive oral films with optimized olive leaf extract (OLE) and microencapsulated thyme essential oil (TEO) contents was aimed to create a novel oral delivery system with desirable film properties as well as enhanced bioactivities. For this, TEO-containing sodium alginate/okra gum microcapsules were first fabricated at optimized electrospray conditions, where optimization was performed for three parameters, which are the flow rate of the emulsion solution, the potential difference, and the distance between the nozzle of the sample container and the collector plate, through three-level centered Box-Behnken design. Prior to the inclusion of the OLE in custom-designed mucoadhesive films, polymeric components of the films were also optimized through a separate multi-variable, three-level Box-Behnken experimental design by which the effect of mixing ratios of Carbopol 934, hydroxypropyl methylcellulose and polyethylene glycol on viscosity, solution pH, film pH, disintegration time, thickness, and weight were elaboratively investigated. The resulting films, containing OLE and microencapsulated TEO, obtained through solvent casting were characterized for pH, viscosity, mass uniformity and thickness, dispersion, tensile strength, folding endurance, contact angle, in vitro mucoadhesion time, and ex vivo mucoadhesion force. Results showed that the film had good tensile strength, swelling capacity, compatibility with oral mucosa, and flexibility. It also demonstrated desirable stability and adhesion ability. Regarding bioactivity, the film was found to have desirable antioxidant capacity and it was non-toxic to fibroblast cells when diluted to certain concentration. Moreover, antiviral studies indicated reductions of 32.39% and 54.29% against Poliovirus Type 1 and Murine Norovirus, respectively, while preliminary antibacterial tests showed no significant reduction in certain bacteria within 1 min, but a logarithmic reduction in Staphylococcus aureus was observed. Despite favorable mucoadhesion properties and physical attributes, further investigation is needed to enhance the antimicrobial effectiveness of the films for their potential utilization in drug delivery systems.

Project description:Waterlogging is expected to increase as a consequence of global climate change, constraining crop production in various parts of the world. This study assessed tolerance to 14-days of early- or late-stage waterlogging of the major winter crops wheat, barley, rapeseed and field pea. Aerenchyma formation in adventitious roots, leaf physiological parameters (net photosynthesis, stomatal and mesophyll conductances, chlorophyll fluorescence), shoot and root growth during and after waterlogging, and seed production were evaluated. Wheat produced adventitious roots with 20-22% of aerenchyma, photosynthesis was maintained during waterlogging, and seed production was 86 and 71% of controls for early- and late-waterlogging events. In barley and rapeseed, plants were less affected by early- than by late-waterlogging. Barley adventitious roots contained 19% aerenchyma, whereas rapeseed did not form aerenchyma. In barley, photosynthesis was reduced during early-waterlogging mainly by stomatal limitations, and by non-stomatal constraints (lower mesophyll conductance and damage to photosynthetic apparatus as revealed by chlorophyll fluorescence) during late-waterlogging. In rapeseed, photosynthesis was mostly reduced by non-stomatal limitations during early- and late-waterlogging, which also impacted shoot and root growth. Early-waterlogged plants of both barley and rapeseed were able to recover in growth upon drainage, and seed production reached ca. 79-85% of the controls, while late-waterlogged plants only attained 26-32% in seed production. Field pea showed no ability to develop root aerenchyma when waterlogged, and its photosynthesis (and stomatal and mesophyll conductances) was rapidly decreased by the stress. Consequently, waterlogging drastically reduced field pea seed production to 6% of controls both at early- and late-stages with plants being unable to resume growth upon drainage. In conclusion, wheat generates a set of adaptive responses to withstand 14 days of waterlogging, barley and rapeseed can still produce significant yield if transiently waterlogged during early plant stages but are more adversely impacted at the late stage, and field pea is not suitable for areas prone to waterlogging events of 14 days at either growth stage.
