Development of a Gas Chromatography-Time-of-Flight Method for Detecting Glucosinolate Metabolites and Volatile Organic Compounds in Kimchi.

ABSTRACT: This study examined the volatile organic compounds (VOCs) and metabolites of glucosinolate (GLS) contained in kimchi and analyzed GLS using myrosinase. The analysis was conducted using gas chromatography-time of flight (GC-TOF), and VOC and the metabolite quantities were detected and analyzed. Based on 22 samples, tests were conducted, and 12 metabolites and 52 VOCs were found. When the detected metabolites were compared in general, the rate of isothiocyanate, which is well known for its anticancer effects and various other activities, was the highest. A total of 52 VOCs, including 15 aliphatic hydrocarbons, 7 acids, and 6 alcohols, were detected by GC-TOF. Therefore, the analytical methods provide a good basis to examine VOC and GLS metabolites; furthermore, the methods are of great help to secure excellent kimchi and evaluate its quality.

Project description:Volatile organic compounds (VOCs) are byproducts from metabolic pathways that can be detected in exhaled breath and have been reported as biomarkers for different diseases. The gold standard for analysis is gas chromatography-mass spectrometry (GC-MS), which can be coupled with various sampling methods. The current study aims to develop and compare different methods for sampling and preconcentrating VOCs using solid-phase microextraction (SPME). An in-house sampling method, direct-breath SPME (DB-SPME), was developed to directly extract VOCs from breath using a SPME fiber. The method was optimized by exploring different SPME types, the overall exhalation volume, and breath fractionation. DB-SPME was quantitatively compared to two alternative methods involving the collection of breath in a Tedlar bag. In one method, VOCs were directly extracted from the Tedlar bag (Tedlar-SPME) and in the other, the VOCs were cryothermally transferred from the Tedlar bag to a headspace vial (cryotransfer). The methods were verified and quantitatively compared using breath samples (n = 15 for each method respectively) analyzed by GC-MS quadrupole time-of-flight (QTOF) for compounds including but not limited to acetone, isoprene, toluene, limonene, and pinene. The cryotransfer method was the most sensitive, demonstrating the strongest signal for the majority of the VOCs detected in the exhaled breath samples. However, VOCs with low molecular weights, including acetone and isoprene, were detected with the highest sensitivity using the Tedlar-SPME. On the other hand, the DB-SPME was less sensitive, although it was rapid and had the lowest background GC-MS signal. Overall, the three breath-sampling methods can detect a wide variety of VOCs in breath. The cryotransfer method may be optimal when collecting a large number of samples using Tedlar bags, as it allows the long-term storage of VOCs at low temperatures (-80 °C), while Tedlar-SPME may be more effective when targeting relatively small VOCs. The DB-SPME method may be the most efficient when more immediate analyses and results are required.

Project description:Volatile and semivolatile organic compounds in ambient air and occupational settings are of great concern due to their associated adverse human health and environmental impacts. Novel graphene wool samplers have been developed and tested to overcome limitations of commercially available sorbents that can only be used once and typically require solvent extraction. Graphene wool (GW) was synthesized by non-catalytic chemical vapor deposition with optimized conditions, resulting in a novel fibrous graphene wool that is very easy to manage and less rigid than other forms of graphene, lending itself to a wide range of potential applications. Here, the air pollutant sampling capabilities of the GW were of interest. The optimal packing weight of GW inside a glass tube (length 178 mm, i.d. 4 mm, o.d. 6 mm) was investigated by the adsorption of vaporized alkane standards on the GW, using a condensation aerosol generator in a temperature-controlled chamber and subsequent detection using a flame ionization detector. The optimized GW packing density was found to be 0.19 mg mm-3 at a flow rate of 500 mL min-1, which provided a gas collection efficiency of >90% for octane, decane, and hexadecane. The humidity uptake of the sampler is less than 1% (m/m) for ambient humidities <70%. Breakthrough studies showed the favorable adsorption of polar molecules, which is attributed to the defective nature of the graphene and the inhomogeneous coating of the graphene layers on the quartz wool, suggesting that the polar versus non-polar uptake potential of the GW can be tuned by varying the graphene layering on the quartz wool substrate during synthesis. Oxidized domains at the irregular edges of the graphene layers, due to a broken, non-pristine sp2 carbon network, allow for adsorption of polar molecules. The GW was applied and used in a combustion sampling campaign where the samplers proved to be comparable to frequently used polydimethylsiloxane sorbents in terms of sampling and thermal desorption of non-polar semivolatile organic compounds. The total alkane concentrations detected after thermal desorption of GW and PDMS samplers were found to be 17.96 ± 13.27 and 18.30 ± 16.42 μg m-3, respectively; thus, the difference in the alkane sampling concentration between the two sorbent systems was negligible. GW provides a new, exciting possibility for the monitoring of organic air pollutants with numerous advantages, including high sampling efficiencies, simple and cost-effective synthesis of the thermally stable GW, solvent-free and environmentally friendly analysis, and, importantly, the reusability of samplers.

Project description:BackgroundNon-invasive diagnostic strategies aimed at identifying biomarkers of cancer are of great interest for early cancer detection. Urine is potentially a rich source of volatile organic metabolites (VOMs) that can be used as potential cancer biomarkers. Our aim was to develop a generally reliable, rapid, sensitive, and robust analytical method for screening large numbers of urine samples, resulting in a broad spectrum of native VOMs, as a tool to evaluate the potential of these metabolites in the early diagnosis of cancer.MethodsTo investigate urinary volatile metabolites as potential cancer biomarkers, urine samples from 33 cancer patients (oncological group: 14 leukaemia, 12 colorectal and 7 lymphoma) and 21 healthy (control group, cancer-free) individuals were qualitatively and quantitatively analysed. Dynamic solid-phase microextraction in headspace mode (dHS-SPME) using a carboxen-polydimethylsiloxane (CAR/PDMS) sorbent in combination with GC-qMS-based metabolomics was applied to isolate and identify the volatile metabolites. This method provides a potential non-invasive method for early cancer diagnosis as a first approach. To fulfil this objective, three important dHS-SPME experimental parameters that influence extraction efficiency (fibre coating, extraction time and temperature of sampling) were optimised using a univariate optimisation design. The highest extraction efficiency was obtained when sampling was performed at 50°C for 60 min using samples with high ionic strengths (17% sodium chloride, w v(-1)) and under agitation.ResultsA total of 82 volatile metabolites belonging to distinct chemical classes were identified in the control and oncological groups. Benzene derivatives, terpenoids and phenols were the most common classes for the oncological group, whereas ketones and sulphur compounds were the main classes that were isolated from the urine headspace of healthy subjects. The results demonstrate that compound concentrations were dramatically different between cancer patients and healthy volunteers. The positive rates of 16 patients among the 82 identified were found to be statistically different (P<0.05). A significant increase in the peak area of 2-methyl-3-phenyl-2-propenal, p-cymene, anisole, 4-methyl-phenol and 1,2-dihydro-1,1,6-trimethyl-naphthalene in cancer patients was observed. On average, statistically significant lower abundances of dimethyl disulphide were found in cancer patients.ConclusionsGas chromatographic peak areas were submitted to multivariate analysis (principal component analysis and supervised linear discriminant analysis) to visualise clusters within cases and to detect the volatile metabolites that are able to differentiate cancer patients from healthy individuals. Very good discrimination within cancer groups and between cancer and control groups was achieved.
