Multiply Charged Helium Droplet Anions.

ABSTRACT: The detection of multiply charged helium droplet anions is reported for the first time. By ionizing droplets of superfluid helium with low energy electrons (up to 25 eV), it was possible to produce droplets containing up to five negative charges, which remain intact on the timescale of the experiment. The appearance sizes for different charge states are determined and are found to be orders of magnitude larger than for the equivalent cationic droplets, starting at 4 million He atoms for dianions. Droplets with He*^- as charge carriers show signs of being metastable, but this effect is quenched by the pickup of water molecules.

Generation of Multiply Charged Protein Anions from Multiply Charged Protein Cations via Gas-Phase Ion/Ion Reactions.

Project description:We report a novel charge inversion ion/ion reaction that converts multiply charged protein cations to multiply charged protein anions via a single ion/ion collision using highly charged anions derived from nanoelectrospray ionization (nESI) of hyaluronic acids (HAs). This type of charge inversion reaction is demonstrated with cations derived from cytochrome c, apo-myoglobin, and carbonic anhydrase (CA) cations. For example, the reaction has been demonstrated to convert the [CA+22H]22+ carbonic anhydrase cation to a distribution of anions as high in absolute charge as [CA-19H]19-. Ion/ion reactions involving multiply charged ions of opposite polarity have previously been observed to result predominantly in the attachment of the reactant ions. All mechanisms for ion/ion charge inversion involving low energy ions proceed via the formation of a long-lived complex. Factors that underlie the charge inversion of protein cations to high anionic charge states in reaction with HA anions are hypothesized to include: (i) the relatively high charge densities of the HA anions that facilitate the extraction of multiple protons from the protein leading to multiply charged protein anions, (ii) the relatively high sum of absolute charges of the reactants that leads to high initial energies in the ion/ion complex, and (iii) the relatively high charge of the ion/ion complex following the multiple proton transfers that tends to destabilize the complex.

Efficient Formation of Size-Selected Clusters upon Pickup of Dopants into Multiply Charged Helium Droplets.

Project description:Properties of clusters often depend critically on the exact number of atomic or molecular building blocks, however, most methods of cluster formation lead to a broad, size distribution and cluster intensity anomalies that are often designated as magic numbers. Here we present a novel approach of breeding size-selected clusters via pickup of dopants into multiply charged helium nanodroplets. The size and charge state of the initially undoped droplets and the vapor pressure of the dopant in the pickup region, determines the size of the dopant cluster ions that are extracted from the host droplets, via evaporation of the helium matrix in a collision cell filled with room temperature helium or via surface collisions. Size distributions of the selected dopant cluster ions are determined utilizing a high-resolution time of flight mass spectrometer. The comparison of the experimental data, with simulations taking into consideration the pickup probability into a shrinking He droplet due to evaporation during the pickup process, provides a simple explanation for the emergence of size distributions that are narrower than Poisson.

193-nm photodissociation of singly and multiply charged peptide anions for acidic proteome characterization.

Project description:193-nm ultraviolet photodissociation (UVPD) was implemented to sequence singly and multiply charged peptide anions. Upon dissociation by this method, a-/x-type, followed by d and w side-chain loss ions, were the most prolific and abundant sequence ions, often yielding 100% sequence coverage. The dissociation behavior of singly and multiply charged anions was significantly different with higher charged precursors yielding more sequence ions; however, all charge states investigated (1- through 3-) produced rich diagnostic information. UVPD at 193 nm was also shown to successfully differentiate and pinpoint labile phosphorylation modifications. The sequence ions were produced with high abundances, requiring limited averaging for satisfactory spectral quality. The intact, charge-reduced radical products generated by UV photoexcitation were also subjected to collision-induced dissociation (termed, activated-electron photodetachment dissociation (a-EPD)), but UVPD alone yielded more predictable and higher abundance sequence ions. With the use of a basic (pH∼11.5), piperidine-modified mobile phase, LC-MS/UVPD was implemented and resulted in the successful analysis of mitogen-activated pathway kinases (MAPKs) using ultrafast activation times (5 ns).

Ion/ion reactions with "onium" reagents: an approach for the gas-phase transfer of organic cations to multiply-charged anions.

Project description:The use of ion/ion reactions to effect gas-phase alkylation is demonstrated. Commonly used fixed-charge "onium" cations are well-suited for ion/ion reactions with multiply deprotonated analytes because of their tendency to form long-lived electrostatic complexes. Activation of these complexes results in an SN2 reaction that yields an alkylated anion with the loss of a neutral remnant of the reagent. This alkylation process forms the basis of a general method for alkylation of deprotonated analytes generated via electrospray, and is demonstrated on a variety of anionic sites. SN2 reactions of this nature are demonstrated empirically and characterized using density functional theory (DFT). This method for modification in the gas phase is extended to the transfer of larger and more complex R groups that can be used in later gas-phase synthesis steps. For example, N-cyclohexyl-N'-(2-morpholinoethyl)carbodiimide (CMC) is used to transfer a carbodiimide functionality to a peptide anion containing a carboxylic acid. Subsequent activation yields a selective reaction between the transferred carbodiimide group and a carboxylic acid, suggesting the carbodiimide functionality is retained through the transfer process. Many different R groups are transferable using this method, allowing for new possibilities for charge manipulation and derivatization in the gas phase.

Size and Velocity Distribution of Negatively Charged Helium Nanodroplets.

Project description:Precharged helium nanodroplets can be used in doping experiments with the advantage that they are amenable to size selection with electrostatic fields, therefore adding a useful tuning parameter for dopant growth. For all these applications, the knowledge of the size distribution of charged droplets is an essential parameter, which we have so far assumed would be equivalent to that of their neutral precursors. Here, this assumption is experimentally investigated for negatively charged clusters for temperatures between 4 and 9 K at a stagnation pressure of 2 MPa. We observe a dependency of the velocity of the droplets on mass per charge, especially at the lowest temperatures of the investigated range, and values 20% lower than those known from the literature. Below 6 K, a large deviation from the literature is also found for the average droplet sizes. This information has to be taken into consideration in future experiments where large, charged droplets are sought to produce large dopant clusters. Possible origins for this deviation are discussed in the text.

Mass Analysis of Macro-molecular Analytes via Multiply-Charged Ion Attachment.

Project description:A novel gas-phase charge and mass manipulation approach is demonstrated to facilitate the mass measurement of high mass complexes within the context of native mass spectrometry. Electrospray ionization applied to solutions generated under native or near-native conditions has been demonstrated to be capable of preserving biologically relevant complexes into the gas phase as multiply charged ions suitable for mass spectrometric analysis. However, charge state distributions tend to be narrow and extensive salt adduction, heterogeneity, and so on tend to lead to significantly broadened peaks. These issues can compromise mass measurement of high mass bio-complexes, particularly when charge states are not clearly resolved. In this work, we show that the attachment of high mass ions of known mass and charge to populations of ions of interest can lead to well-separated signals that can yield confident charge state and mass assignments from otherwise poorly resolved signals.

Separation and Simultaneous Trapping of Multiply Charged and Singly Charged Ions for Mass Spectrometry: Application to Lipid Mixtures.

Project description:Conventional electrospray ionization (ESI) of mixtures can give rise to singly and multiply charged analyte species that overlap in mass-to-charge (m/z) ratios, which can complicate the analysis of individual components. The overlap in m/z for ions of different mass and charge is particularly problematic when ions of low relative abundance are of interest. For example, cardiolipins (CLs) are structurally complex phospholipids present in low relative abundance in the lipidome but play crucial roles in mitochondrial metabolism and various regulatory processes. ESI of CLs in negative ion mode shows abundant doubly deprotonated ions and minor singly deprotonated ions. In the ESI of lipid extracts, highly abundant singly charged phospholipids extensively overlap in m/z space with CL dianions of much lesser abundance, thereby complicating the study of the CLs. To address this challenge, we employed a gas-phase approach to separate singly charged ions from a population of ions of mixed charge states while allowing for the storage of one or both of the separated ion populations. Herein, we describe the considerations for applying enhanced singly charged (ESC) and enhanced multiply charged (EMC) scans to perform a gas-phase separation of singly charged lipids from doubly charged lipids in an Escherichia coli extract. This method allows for improved signal-to-noise (S/N) ratio of low abundance ions with minimal overall signal loss, removal of "chemical noise" arising from singly charged ions, and allows for retention of spatially separated ions within a mass spectrometer.

Structure and formation of copper cluster ions in multiply charged He nanodroplets.

Project description:The structure of cationic and anionic Cu clusters grown in multiply charged superfluid He nanodroplets was investigated using He tagging as a chemical probe. Further, the structure assignment was done based on the magic-numbered ions, representing the most energetically favorable structures. The exact geometry of the cluster and positions of He is verified by calculations. It was found that the structure of the clusters grown in the He droplets is similar to that produced with a laser ablation source and the lowest energy structures predicted by theoretical investigations. The only difference is the structure of the Cu5+, which in our experiments has a twisted-X geometry, rather than a bipyramid or planar half-wheel geometry suggested by previous studies. This might be attributed to the different cluster formation mechanisms, the absence of the Ar-tag and the ultracold environment. It was also found that He tends to bind to partially more electro-negative or positive areas of the anionic or cationic clusters, respectively.

Charge-Dependent Metastable Dissociations of Multiply Charged Decafluorobiphenyl Formed by Femtosecond Laser Pulses.

Project description:Femtosecond laser ionization is a unique means to produce multiply charged organic molecules in the gas phase. The charge-dependent chemical reactions of such electron-deficient molecules are interesting from both fundamental and applied scientific perspectives. We have reported the production of quadruply charged perfluoroaromatics; however, they were so stable that we cannot obtain information about their chemical reactions. In general, it might be difficult to realize the conflicting objectives of observing multiply charged molecular ion themselves and their metastable dissociations. In this study, we report the first example showing metastable dissociations of several charge states within the measurable time range of a time-of-flight mass spectrometer. Metastable dissociations were analyzed by selecting a precursor ion with a Bradbury-Nielsen ion gate followed by time-of-flight analysis using a reflectron. We obtained qualitative information that triply and quadruply charged decafluorobiphenyl survived at least in the acceleration region but completely decomposed before entering a reflectron. In contrast, three dissociation channels for singly and one for doubly charged molecular ions were discriminated by a reflectron and determined with the help of ion trajectory simulations.

Fragmentation of multiply-charged intact protein ions using MALDI TOF-TOF mass spectrometry.

Project description:Top down proteomics in a TOF-TOF instrument was further explored by examining the fragmentation of multiply charged precursors ions generated by matrix-assisted laser desorption ionization. Evaluation of sample preparation conditions allowed selection of solvent/matrix conditions and sample deposition methods to produce sufficiently abundant doubly and triply charged precursor ions for subsequent CID experiments. As previously reported, preferential cleavage was observed at sites C-terminal to acidic residues and N-terminal to proline residues for all ions examined. An increase in nonpreferential fragmentation as well as additional low mass product ions was observed in the spectra from multiply charged precursor ions providing increased sequence coverage. This enhanced fragmentation from multiply charged precursor ions became increasingly important with increasing protein molecular weight and facilitates protein identification using database searching algorithms. The useable mass range for MALDI TOF-TOF analysis of intact proteins has been expanded to 18.2 kDa using this approach.
