Innovative finding of 266-nm laser regulating CD90 levels in SDSCs.

ABSTRACT: We used light to irradiate skin-derived stem cells and tried to find any cellular protein alterations 24 h after illumination. A 266-nm laser with four intensities was used, and of the nine cell markers that were surveyed in our trials, only CD90 was downregulated at an intensity of 20 μJ for 10 s. Repeated illuminations from the 266-nm laser at seven intensities revealed that CD90 expression was downregulated 14.6-28.8%, depending on light intensity. The maximal effect was noted at an intensity of 30 μJ for 2 s. This innovative finding reveals that a 266-nm laser can regulate protein expression in skin-derivative stem cells.

Evaluating the efficacy of Nd:YAG fourth harmonic (266 nm) in comparison with ArF excimer (193 nm) in laser corneal reshaping: ex vivo pilot study.

Project description:PurposeLaser corneal reshaping is a common eye surgery utilized to overcome many vision disorders. Different UV laser wavelengths can be effective in the treatment. However, the ArF excimer laser (193 nm) is the most commonly used due to its high absorption in the cornea. In the current study, we investigate the efficacy of applying a solid-state laser (Nd:YAG fourth harmonic at 266 nm) for the corneal reshaping procedure.MethodsThe utilized laser is generated using an optical setup based on a BBO nonlinear crystal which converts the Q-switched laser (532 nm) to its fourth harmonic (266 nm). Different pulse energies were applied with the same number of the shoots on ex vivo rabbit corneas, and the histological effect is studied. Moreover, the possible thermal damage on the treated corneal tissues was inspected via electron microscope. Additionally, the DNA damage on the corneal cells due to the application of the proposed laser was examined and compared with the existing technology (ArF Excimer laser at 193 nm) using the comet assay.ResultsThe histological examination revealed an appropriate ablation result with the minimum thermal effect at 1.5 mJ and 2.0 mJ. The overall results show that applying 50-shoots of the 1.5-mJ pulse energy using the proposed 266-nm solid-state laser produces the optimum ablation effect with the minimum thermal damage, and almost the same DNA damage occurred using the commercial 193-nm ArF excimer laser.ConclusionSolid-state laser at 266 nm could be a good alternative to the common 193-nm excimer laser for corneal reshaping procedures.

Clusters formation and fragmentation of nitromethane at 266 nm.

Project description:We carry out experiments on the fragmentation of nitromethane by multiphoton absorption at the wavelength 266 nm. This was conducted in a reflectron (Jordan), modified in the laboratory. Due to the large number of fragments, special care has been taken into the calibration of the system, in the simultaneity between the laser pulse and the sample, and the associated electronics to ensure that produced fragment spectra arise from the interaction laser-sample. We emphasize the next aspects of the method:•Simple design for introducing a gas sample at laser interaction region to facilitate the cluster formation•Astonishing number of fragments produced by multiphoton absorption.

Ultrafast inactivation of SARS-CoV-2 with 266 nm lasers.

Project description:Disinfection eliminates pathogenic microorganisms and ensures a biosafe environment for human beings. The rapid spread of COVID-19 is challenging traditional disinfection methods in terms of reducing harmful side effects and conducting faster processes. Spraying large-scale chemical disinfectants is harmful to individuals and the environment, while UV lamp and light-emitting diode (LED) disinfection still requires a long exposure time due to the low irradiance and highly divergent beam characteristics. Given that a laser maintains a high irradiance over a long distance, we studied the effectiveness of lasers as a new disinfection method, and the results show the capability for ultrafast inactivation of SARS-CoV-2 virus with a 266 nm laser. This work confirms UV lasers as a good candidate for disinfection.

Histological Effects of an Innovative 445 Nm Blue Laser During Oral Soft Tissue Biopsy.

Project description:Continuously evolving laser devices can be used in various fields; they are an alternative to the traditional cold blade surgery to perform biopsies of oral soft tissues. The aspect focused on in this paper is the possibility to use the 445 nm diode laser (Eltech K-Laser srl, Treviso, Italy) in complete safety, by evaluating its thermal effects during microscopy. A histological evaluation of the alteration of the peri-incisional edges on 10 samples was realized. All excisional biopsies were related to clinically unsuspected lesions and performed by the same expert operator. The surgical procedure was performed with the same laser parameters and the same pathologist evaluated the thermal effect on the samples. An average value of the detected tissue alteration was calculated; the average damage of the epithelium was 650.93 ?m, while in the connective tissue it was 468.07 ?m. In all the cases a clear diagnosis was possible, and no clinical complications were observed; so, the 445 nm diode laser proved to be a device that can be safely used for biopsies of clinically unsuspicious lesions. Due to the small number of cases, this in vivo preliminary experience needs to be extended.

Dissociative Photoionization of Dimethylpyridines and Trimethylpyridine at 266 nm: Dynamics of Methyl Radical Release.

Project description:The 266 nm photolysis of various positional isomers of dimethylpyridines and trimethylpyridine was investigated by measuring the translational energy distribution of the methyl radical following {sp2}C-C{sp3} bond dissociation. The observed translational energy distribution is attributed to the dissociative photoionization in the cationic ground state following [1 + 1 + 1] three-photon absorption. The translational energy distribution profiles of the methyl radical were broad with the maximum translation energy in excess of 2 eV, which originates due to the dissociation of {sp2}C-C{sp3} bond ortho to the N atom in the ring. The dynamics of {sp2}C-C{sp3} bond dissociation in the cationic ground state of methylpyridines is marginally dependent on the number and position of the methyl groups; similar to xylenes, however, it is site-selective with the preferential cleavage of C-C bond in the ortho position to the pyridinic nitrogen atom, which is attributed to the relative stability of the resulting radical cation.

Chromophore Optimization in Organometallic Au(III) Cys Arylation of Peptides and Proteins for 266 nm Photoactivation.

Project description:Cysteine is the most reactive naturally occurring amino acid due to the presence of a free thiol, presenting a tantalizing handle for covalent modification of peptides/proteins. Although many mass spectrometry experiments could benefit from site-specific modification of Cys, the utility of direct arylation has not been thoroughly explored. Recently, Spokoyny and co-workers reported a Au(III) organometallic reagent that robustly arylates Cys and tolerates a wide variety of solvents and conditions. Given the chromophoric nature of aryl groups and the known susceptibility of carbon-sulfur bonds to photodissociation, we set out to identify an aryl group that could efficiently cleave Cys carbon-sulfur bonds at 266 nm. A streamlined workflow was developed to facilitate rapid examination of a large number of aryls with minimal sample using a simple test peptide, RAAACGVLK. We were able to identify several aryl groups that yield abundant homolytic photodissociation of the adjacent Cys carbon-sulfur bonds with short activation times (<10 ms). In addition, we characterized the radical products created by photodissociation by subjecting the product ions to further collisional activation. Finally, we tested Cys arylation with human hemoglobin, identified reaction conditions that facilitate efficient modification of intact proteins, and evaluated the photochemistry and activation of these large radical ions.

Effects of 445 nm, 520 nm, and 638 nm Laser Irradiation on the Dermal Cells.

Project description:Background: The invention of non-ionizing emission devices revolutionized science, medicine, industry, and the military. Currently, different laser systems are commonly used, generating the potential threat of excessive radiation exposure, which can lead to adverse health effects. Skin is the organ most exposed to laser irradiation; therefore, this study aims to evaluate the effects of 445 nm, 520 nm, and 638 nm non-ionizing irradiation on keratinocytes and fibroblasts. Methods: Keratinocytes and fibroblasts were exposed to a different fluency of 445 nm, 520 nm, and 638 nm laser irradiation. In addition, viability, type of cell death, cell cycle distribution, and proliferation rates were investigated. Results: The 445 nm irradiation was cytotoxic to BJ-5ta (≥58.7 J/cm2) but not to Ker-CT cells. Exposure influenced the cell cycle distribution of Ker-CT (≥61.2 J/cm2) and BJ-5ta (≥27.6 J/cm2) cells, as well as the Bj-5ta proliferation rate (≥50.5 J/cm2). The 520 nm irradiation was cytotoxic to BJ-5ta (≥468.4 J/cm2) and Ker-CT (≥385.7 J/cm2) cells. Cell cycle distribution (≥27.6 J/cm2) of Ker-CT cells was also affected. The 638 nm irradiation was cytotoxic to BJ-5ta and Ker-CT cells (≥151.5 J/cm2). The proliferation rate and cell cycle distribution of BJ-5ta (≥192.9 J/cm2) and Ker-CT (13.8 and 41.3 J/cm2) cells were also affected. Conclusions: At high fluences, 455 nm, 520 nm, and 638 nm irradiation, representing blue, green, and red light spectra, are hazardous to keratinocytes and fibroblasts. However, laser irradiation may benefit the cells at low fluences by modulating the cell cycle and proliferation rate.

Ultraviolet 320 nm laser excitation for flow cytometry.

Project description:Although multiple lasers and high-dimensional analysis capability are now standard on advanced flow cytometers, ultraviolet (UV) lasers (usually 325-365 nm) remain an uncommon excitation source for cytometry. This is primarily due to their cost, and the small number of applications that require this wavelength. The development of the Brilliant Ultraviolet (BUV fluorochromes, however, has increased the importance of this formerly niche excitation wavelength. Historically, UV excitation was usually provided by water-cooled argon- and krypton-ion lasers. Modern flow cytometers primary rely on diode pumped solid state lasers emitting at 355 nm. While useful for all UV-excited applications, DPSS UV lasers are still large by modern solid state laser standards, and remain very expensive. Smaller and cheaper near UV laser diodes (NUVLDs) emitting at 375 nm make adequate substitutes for 355 nm sources in many situations, but do not work as well with very short wavelength probes like the fluorescent calcium chelator indo-1. In this study, we evaluate a newly available UV 320 nm laser for flow cytometry. While shorter in wavelength that conventional UV lasers, 320 is close to the 325 nm helium-cadmium wavelength used in the past on early benchtop cytometers. A UV 320 nm laser was found to excite almost all Brilliant Ultraviolet dyes to nearly the same level as 355 nm sources. Both 320 nm and 355 nm sources worked equally well for Hoechst and DyeCycle Violet side population analysis of stem cells in mouse hematopoetic tissue. The shorter wavelength UV source also showed excellent excitation of indo-1, a probe that is not compatible with NUVLD 375 nm sources. In summary, a 320 nm laser module made a suitable substitute for conventional 355 nm sources. This laser technology is available in a smaller form factor than current 355 nm units, making it useful for small cytometers with space constraints. © 2017 International Society for Advancement of Cytometry.

A Comparative Study of Laser-Induced Graphene by CO2 Infrared Laser and 355 nm Ultraviolet (UV) Laser.

Project description:Laser-induced graphene (LIG) is an emerging technique for producing few-layer graphene or graphene-like material that has recently received increasing attention, due to its unique advantages. Subsequently, a variety of lasers and materials have been used to fabricate LIG using this technique. However, there is a lack of understanding of how different lasers (wavelengths) perform differently in the LIG conversion process. In this study, the produced LIG on polyimide (PI) under a locally water-cooled condition using a 10.6 μm CO2 infrared laser and a 355 nm ultraviolet (UV) laser are compared. The experimental investigations reveal that under the same UV and CO2 laser fluence, the ablation of PI show different results. Surface morphologies with micron-sized and nanometer pores were formed by the UV laser under different laser fluences, whereas micron-sized pores and sheet structure with fewer pores were produced by the CO2 laser. Energy dispersive spectrometry and three-dimensional topography characterization indicate that the photochemical effects were also involved in the LIG conversion with UV laser irradiation. It is also observed through experiments that the photothermal effect contributed to the formation of LIG under both lasers, and the LIG formed on PI substrates by the CO2 laser showed better quality and fewer layers.

Efficacy and safety of 1565-nm non-ablative fractional laser versus long-pulsed 1064-nm Nd:YAG laser in treating enlarged facial pores.

Project description:Facial pores are visible openings of pilosebaceous follicles, and they are one of the major factors influencing facial skin appearance. This article aims to evaluate and compare the efficacy and safety of 1565-nm non-ablative fractional laser (NAFL) and long-pulsed 1064-nm Nd:YAG laser (LPNY) in treating enlarged facial pores. All subjects were treated with NAFL on their left faces and LPNY on their right. Five treatments were administered at 2-week intervals, with one follow-up session 2 months after the final treatment. Treatment efficacy was evaluated by subjective (pore improvement and subject satisfaction ratings) assessments and objective (pore number) assessments. At each appointment, any side effects or complications were recorded to evaluate the safety of the two lasers. A total of 18 individuals participated in this study. At the 2-month follow-up, NAFL and LPNY sides had significant reduction in pores (p < 0.0001 and p < 0.0001, respectively). However, there was no statistically significant difference in the mean number of pore reductions on either side (p > 0.05). There was no significant difference in pore improvement ratings and satisfaction ratings between the two sides (p > 0.05 and p > 0.05, respectively). Both lasers showed minimal side effects. Both lasers effectively treated enlarged facial pores and were well tolerated. The side effects of the 1064-nm LPNY were less severe than those of the 1565-nm NAFL.
