Background

Historical natural history collections are very important for the study of nature and environmental protection of the environment, these being the depository of essential information. The Fondazione Museo Civico di Rovereto holds two major Orthopteroid insect collections that make this Museum a landmark on Italian and Mediterranean Orthoptera diversity. Databasing the Galvagni Collection allows considerations on geographic and taxonomic coverage by specialist researchers.

Global impact of the COVID-19 lockdown on biodiversity data collection.

Project description:The COVID-19 pandemic triggered different governmental responses across borders, with cascading effects on people's movements and on biodiversity data collection. We quantified changes in the number of species occurrence records collected during the first global lockdown (March 15th to May 1st 2020) relative to pre-pandemic levels using data from the Global Biodiversity Information Facility (GBIF). We modelled how such changes relate to the stringency of governmental policy responses, changes in human mobility, and countries' population size and economic class across 129 countries. We further focused on data from the community science project eBird, which constitutes the largest dataset in GBIF, to investigate changes in participation and activity patterns of individual observers (eBirders) during the lockdown. We found that the decreases in GBIF records correlated with declines in numbers of visitors to parks and outdoor areas, and were significantly larger in developing countries compared to developed ones. While the activity ranges of eBirders shrunk across all countries analysed, the number of eBirders in developing and least developed countries declined more than in developed countries, as the lockdown disrupted the influx of international visitors. Our results suggest that community-based, local monitoring programmes are essential to reduce biases in global biodiversity monitoring.

People's expectations and experiences of big data collection in the Saudi context.

Project description:Big data and machine learning technologies facilitate various business intelligence activities for businesses. However, personal data collection can generate adverse effects on consumers. Big data collection can compromise people's sense of autonomy, harming digital privacy, transparency and trust. This research investigates personal data collection, control, awareness, and privacy regulation on people's autonomy in Saudi. This study used a hybrid analytical model that incorporates symmetrical and asymmetrical analysis via fuzzy set qualitative comparative analysis (fsQCA) to analyze consumer sense of autonomy regarding big data collection. The symmetrical shows that 'Control' had the most significant influence on people's autonomy, followed by 'Big data collection' and 'Awareness'. The fsQCA shows 84% of the variation, explaining the people's autonomy.

Harnessing big data for health equity through a comprehensive public database and data collection framework.

Project description:The United States Department of Health and Human Services (HHS) pledged $90 million to help reduce health disparities with data-driven solutions. The funds are being distributed to 1400 community health centers, serving over 30 million Americans. Given these developments, our piece examines the reasons behind the delayed adoption of big data for healthcare equity, recent efforts embracing big data tools, and methods to maximize potential without overburdening physicians. We additionally propose a public database for anonymized patient data, introducing diverse metrics and equitable data collection strategies, providing valuable insights for policymakers and health systems to better serve communities.

Toward global integration of biodiversity big data: a harmonized metabarcode data generation module for terrestrial arthropods.

Project description:Metazoan metabarcoding is emerging as an essential strategy for inventorying biodiversity, with diverse projects currently generating massive quantities of community-level data. The potential for integrating across such data sets offers new opportunities to better understand biodiversity and how it might respond to global change. However, large-scale syntheses may be compromised if metabarcoding workflows differ from each other. There are ongoing efforts to improve standardization for the reporting of inventory data. However, harmonization at the stage of generating metabarcode data has yet to be addressed. A modular framework for harmonized data generation offers a pathway to navigate the complex structure of terrestrial metazoan biodiversity. Here, through our collective expertise as practitioners, method developers, and researchers leading metabarcoding initiatives to inventory terrestrial biodiversity, we seek to initiate a harmonized framework for metabarcode data generation, with a terrestrial arthropod module. We develop an initial set of submodules covering the 5 main steps of metabarcode data generation: (i) sample acquisition; (ii) sample processing; (iii) DNA extraction; (iv) polymerase chain reaction amplification, library preparation, and sequencing; and (v) DNA sequence and metadata deposition, providing a backbone for a terrestrial arthropod module. To achieve this, we (i) identified key points for harmonization, (ii) reviewed the current state of the art, and (iii) distilled existing knowledge within submodules, thus promoting best practice by providing guidelines and recommendations to reduce the universe of methodological options. We advocate the adoption and further development of the terrestrial arthropod module. We further encourage the development of modules for other biodiversity fractions as an essential step toward large-scale biodiversity synthesis through harmonization.

The Anfinsen Dogma: Intriguing Details Sixty-Five Years Later.

Project description:The pioneering experiments of Anfinsen on the oxidative folding of RNase have been revisited discovering some details, which update the statement of his dogma and shed new light on the leading role of the correct disulfide in the attainment of the native structure. CD analysis, mass spectrometry, fluorescence spectroscopy and enzyme activity indicate that native disulfides drive the formation of the secondary and tertiary structures that cannot be entirely formed in their absence. This opposes a common opinion that these structures are first formed and then stabilized by the native disulfides. Our results also indicate that a spontaneous re-oxidation of a reduced RNase cannot produce a complete recovery of activity, as described by many textbooks; this can be obtained only in the presence of a reshuffling solution such as GSH/GSSG.

Limited sampling hampers "big data" estimation of species richness in a tropical biodiversity hotspot.

Project description:Macro-scale species richness studies often use museum specimens as their main source of information. However, such datasets are often strongly biased due to variation in sampling effort in space and time. These biases may strongly affect diversity estimates and may, thereby, obstruct solid inference on the underlying diversity drivers, as well as mislead conservation prioritization. In recent years, this has resulted in an increased focus on developing methods to correct for sampling bias. In this study, we use sample-size-correcting methods to examine patterns of tropical plant diversity in Ecuador, one of the most species-rich and climatically heterogeneous biodiversity hotspots. Species richness estimates were calculated based on 205,735 georeferenced specimens of 15,788 species using the Margalef diversity index, the Chao estimator, the second-order Jackknife and Bootstrapping resampling methods, and Hill numbers and rarefaction. Species richness was heavily correlated with sampling effort, and only rarefaction was able to remove this effect, and we recommend this method for estimation of species richness with "big data" collections.
