Fate and transformation of silver nanoparticles in different biological conditions.

ABSTRACT: The exploitation of silver nanoparticles (AgNPs) in biomedicine represents more than one third of their overall application. Despite their wide use and significant amount of scientific data on their effects on biological systems, detailed insight into their in vivo fate is still lacking. This study aimed to elucidate the biotransformation patterns of AgNPs following oral administration. Colloidal stability, biochemical transformation, dissolution, and degradation behaviour of different types of AgNPs were evaluated in systems modelled to represent biological environments relevant for oral administration, as well as in cell culture media and tissue compartments obtained from animal models. A multimethod approach was employed by implementing light scattering (dynamic and electrophoretic) techniques, spectroscopy (UV-vis, atomic absorption, nuclear magnetic resonance) and transmission electron microscopy. The obtained results demonstrated that AgNPs may transform very quickly during their journey through different biological conditions. They are able to degrade to an ionic form and again reconstruct to a nanoparticulate form, depending on the biological environment determined by specific body compartments. As suggested for other inorganic nanoparticles by other research groups, AgNPs fail to preserve their specific integrity in in vivo settings.

Project description:PurposeThe biomedical applications of silver nanoparticles (AgNPs) are heavily investigated due to their cytotoxic and antimicrobial properties. However, the scientific literature is lacking in data on the aggregation behavior of nanoparticles, especially regarding its impact on biological activity. Therefore, to assess the potential of AgNPs in therapeutic applications, two different AgNP samples were compared under biorelevant conditions.MethodsCitrate-capped nanosilver was produced by classical chemical reduction and stabilization with sodium citrate (AgNP@C), while green tea extract was used to produce silver nanoparticles in a green synthesis approach (AgNP@GTs). Particle size, morphology, and crystallinity were characterized using transmission electron microscopy. To observe the effects of the most important biorelevant conditions on AgNP colloidal stability, aggregation grade measurements were carried out using UV-Vis spectroscopy and dynamic light scatterig, while MTT assay and a microdilution method were performed to evaluate the effects of aggregation on cytotoxicity and antimicrobial activity in a time-dependent manner.ResultsThe aggregation behavior of AgNPs is mostly affected by pH and electrolyte concentration, while the presence of biomolecules can improve particle stability due to the biomolecular corona effect. We demonstrated that high aggregation grade in both AgNP samples attenuated their toxic effect toward living cells. However, AgNP@GT proved less prone to aggregation thus retained a degree of its toxicity.ConclusionTo our knowledge, this is the first systematic examination regarding AgNP aggregation behavior with simultaneous measurements of its effect on biological activity. We showed that nanoparticle behavior in complex systems can be estimated by simple compounds like sodium chloride and glutamine. Electrostatic stabilization might not be suitable for biomedical AgNP applications, while green synthesis approaches could offer new frontiers to preserve nanoparticle toxicity by enhancing colloidal stability. The importance of properly selected synthesis methods must be emphasized as they profoundly influence colloidal stability, and therefore biological activity.

Project description:Silver nanoparticles (AgNPs) are excellent antimicrobial agents and promising candidates for preventing or treating bacterial infections caused by antibiotic resistant strains. However, their increasing use in commercial products raises concerns about their environmental impact. In addition, traditional physicochemical approaches often involve harmful agents and excessive energy consumption, resulting in AgNPs with short-term colloidal stability and silver ion leaching. To address these issues, we designed stable hybrid lignin-silver nanoparticles (AgLigNPs) intended to effectively hit bacterial envelopes as a main antimicrobial target. The lignin nanoparticles (LigNPs), serving as a reducing and stabilizing agent for AgNPs, have a median size of 256 nm and a circularity of 0.985. These LigNPs were prepared using the dialysis solvent exchange method, producing spherical particles stable under alkaline conditions and featuring reducing groups oriented toward a wrinkled surface, facilitating AgNPs synthesis and attachment. Maximum accumulation of silver on the LigNP surface was observed at a mass reaction ratio mAg:mLig of 0.25, at pH 11. The AgLigNPs completely inhibited suspension growth and reduced biofilm development by 50% in three tested strains of Pseudomonas aeruginosa at a concentration of 80/9.5 (lignin/silver) mg L-1. Compared to unattached AgNPs, AgLigNPs required two to eight times lower silver concentrations to achieve complete inhibition. Additionally, our silver-containing nanosystems were effective against bacteria at safe concentrations in HEK-293 and HaCaT tissue cultures. Stability experiments revealed that the nanosystems tend to aggregate in media used for bacterial cell cultures but remain stable in media used for tissue cultures. In all tested media, the nanoparticles retained their integrity, and the presence of lignin facilitated the prevention of silver ions from leaching. Overall, our data demonstrate the suitability of AgLigNPs for further valorization in the biomedical sector.
