Regularities in human mortality after age 105.

ABSTRACT: Empirical research on human mortality and extreme longevity suggests that the risk of death among the oldest-old ceases to increase and levels off at age 110. The universality of this finding remains in dispute because of two main reasons: i) high uncertainty around statistical estimates generated from scarce data, and ii) the lack of country-specific comparisons. In this article, we estimate age patterns of mortality above age 105 using data from the International Database on Longevity, an exceptionally large and recently updated database comprising more than 13,000 validated records of long-lived individuals from eight populations. We show that, in all of them, similar mortality trajectories arise, suggesting that the risk of dying levels off after age 105. As more high-quality data become available, there is more evidence in support of a levelling-off of the risk of dying as a regularity of longevous populations.

Project description:We use a combination of extreme value statistics, survival analysis and computer-intensive methods to analyse the mortality of Italian and French semi-supercentenarians. After accounting for the effects of the sampling frame, extreme-value modelling leads to the conclusion that constant force of mortality beyond 108 years describes the data well and there is no evidence of differences between countries and cohorts. These findings are consistent with use of a Gompertz model and with previous analysis of the International Database on Longevity and suggest that any physical upper bound for the human lifespan is so large that it is unlikely to be approached. Power calculations make it implausible that there is an upper bound below 130 years. There is no evidence of differences in survival between women and men after age 108 in the Italian data and the International Database on Longevity, but survival is lower for men in the French data.

Partial Correlation between Spatial and Temporal Regularities of Human Mobility.

Project description:The regularity of human mobility has been extensively studied because of its prominent applications in a considerable number of important areas. Entropy, in addition to many other measures, has long been used to quantify the regularity of human mobility. We adopt the commonly used spatial entropy and develop an analogical temporal entropy to separately investigate the spatial and temporal regularities of human mobility. The underlying data are from an automated transit fare collection system operated by a metropolitan public transit authority in China. The distributions of both spatial and temporal entropies and their dependences on several widely used statistics are examined. The spatial and temporal entropies present a statistically significant correlation, which has not previously been reported to the best of our knowledge.

Multiple mechanisms regulate statistical learning of orthographic regularities in school-age children: Neurophysiological evidence.

Project description:Using event-related potentials (ERPs), this study investigated how the brains of Chinese children of different ages extract and encode relational patterns contained in orthographic input. Ninety-nine Chinese children in Grades 1-3 performed an artificial orthography statistical learning task that comprised logographic components embedded in characters with high (100%), moderate (80%), and low (60%) positional consistency. The behavioral results indicated that across grades, participants more accurately recognized characters with high rather than low consistency. The neurophysiological results revealed that in each grade, the amplitude of some ERP components differed, with a larger P1 effect in the high consistency condition and a larger N170 and left-lateralized P300 effect in the low consistency condition. A smaller N170 effect occurred in Grade 3 than in Grade 1, and a larger P300 effect occurred in Grade 1 than in either Grade 2 or 3. These findings suggest the dynamic nature of statistical learning by showing that neural adaptation associated with N170, and attention and working memory related to P1 and P300, regulate different types of structural input, and that children's abilities to prioritize these mechanisms vary with context and age.

Rapid encoding of task regularities in the human hippocampus guides sensorimotor timing.

Project description:The brain encodes the statistical regularities of the environment in a task-specific yet flexible and generalizable format. Here, we seek to understand this process by bridging two parallel lines of research, one centered on sensorimotor timing, and the other on cognitive mapping in the hippocampal system. By combining functional magnetic resonance imaging (fMRI) with a fast-paced time-to-contact (TTC) estimation task, we found that the hippocampus signaled behavioral feedback received in each trial as well as performance improvements across trials along with reward-processing regions. Critically, it signaled performance improvements independent from the tested intervals, and its activity accounted for the trial-wise regression-to-the-mean biases in TTC estimation. This is in line with the idea that the hippocampus supports the rapid encoding of temporal context even on short time scales in a behavior-dependent manner. Our results emphasize the central role of the hippocampus in statistical learning and position it at the core of a brain-wide network updating sensorimotor representations in real time for flexible behavior.

Modulation of TLR2 protein expression by miR-105 in human oral keratinocytes.

Project description:Mammalian biological processes such as inflammation, involve regulation of hundreds of genes controlling onset and termination. MicroRNAs (miRNAs) can translationally repress target mRNAs and regulate innate immune responses. Our model system comprised primary human keratinocytes, which exhibited robust differences in inflammatory cytokine production (interleukin-6 and tumor necrosis factor-alpha) following specific Toll-like receptor 2 and 4 (TLR-2/TLR-4) agonist challenge. We challenged these primary cells with Porphyromonas gingivalis (a Gram-negative bacterium that triggers TLR-2 and TLR-4) and performed miRNA expression profiling. We identified miRNA (miR)-105 as a modulator of TLR-2 protein translation in human gingival keratinocytes. There was a strong inverse correlation between cells that had high cytokine responses following TLR-2 agonist challenge and miR-105 levels. Knock-in and knock-down of miR-105 confirmed this inverse relationship. In silico analysis predicted that miR-105 had complementarity for TLR-2 mRNA, and the luciferase reporter assay verified this. Further understanding of the role of miRNA in host responses may elucidate disease susceptibility and suggest new anti-inflammatory therapeutics.

Shaping of object representations in the human medial temporal lobe based on temporal regularities.

Project description:Regularities are gradually represented in cortex after extensive experience, and yet they can influence behavior after minimal exposure. What kind of representations support such rapid statistical learning? The medial temporal lobe (MTL) can represent information from even a single experience, making it a good candidate system for assisting in initial learning about regularities. We combined anatomical segmentation of the MTL, high-resolution fMRI, and multivariate pattern analysis to identify representations of objects in cortical and hippocampal areas of human MTL, assessing how these representations were shaped by exposure to regularities. Subjects viewed a continuous visual stream containing hidden temporal relationships--pairs of objects that reliably appeared nearby in time. We compared the pattern of blood oxygen level-dependent activity evoked by each object before and after this exposure, and found that perirhinal cortex, parahippocampal cortex, subiculum, CA1, and CA2/CA3/dentate gyrus (CA2/3/DG) encoded regularities by increasing the representational similarity of their constituent objects. Most regions exhibited bidirectional associative shaping, whereas CA2/3/DG represented regularities in a forward-looking predictive manner. These findings suggest that object representations in MTL come to mirror the temporal structure of the environment, supporting rapid and incidental statistical learning.

Attention is spontaneously biased toward regularities.

Project description:Knowledge about regularities in the environment can be used to facilitate perception, memory, and language acquisition. Given this usefulness, we hypothesized that statistically structured sources of information receive attentional priority over noisier sources, independent of their intrinsic salience or goal relevance. We report three experiments that support this hypothesis. Experiment 1 shows that regularities bias spatial attention: Visual search was facilitated at a location containing temporal regularities, even though these regularities did not predict target location, timing, or identity. Experiments 2 and 3 show that regularities bias feature attention: Attentional capture doubled in magnitude when singletons appeared, respectively, in a color or dimension with temporal regularities among task-irrelevant stimuli. Prioritization of the locations and features of regularities is not easily accounted for in the conventional dichotomy between stimulus-driven and goal-directed attention. This prioritization may in turn promote further statistical learning, helping the mind to acquire knowledge about stable aspects of the environment.

Impact of delayed effects on human old-age mortality<sub>1</sub>.

Project description:BackgroundThere is growing empirical evidence supporting theories of developmental origins of health and disease (DOHaD). However, the implications of DOHaD conjectures for aggregate population patterns of human disease, disability, mortality and aging are poorly understood.ObjectiveWe empirically test two predictions derived from a formal model of aggregate population-level impacts of DOHaD. This model predicts that populations potentially influenced by delayed effects should experience singularities in their adult mortality patterns that can be empirically detected from aggregate data.MethodsWe test predictions using a large mortality database for populations in the Latin American and Caribbean region (LAC) spanning nearly one hundred years of mortality history.ResultsResults are consistent. within explicit bounds of uncertainty, with expected patterns. We find that younger cohorts in countries whose mortality decline starts more recently experience deceleration in survival gains at older ages, attenuation of the rate of aging at older ages and a decline in the association between early childhood and adult mortality.ConclusionsResults point to the importance of adverse early conditions for human longevity. Future research should shed light on the impact on morbidity, disability and healthy life expectancy.ContributionTo our knowledge this is the first time that implications of DOHaD conjectures for populations' mortality patterns are formulated precisely and empirically tested with aggregate population data.

Age-associated microRNA expression in human peripheral blood is associated with all-cause mortality and age-related traits.

Project description:Recent studies provide evidence of correlations of DNA methylation and expression of protein-coding genes with human aging. The relations of microRNA expression with age and age-related clinical outcomes have not been characterized thoroughly. We explored associations of age with whole-blood microRNA expression in 5221 adults and identified 127 microRNAs that were differentially expressed by age at P < 3.3 × 10-4 (Bonferroni-corrected). Most microRNAs were underexpressed in older individuals. Integrative analysis of microRNA and mRNA expression revealed changes in age-associated mRNA expression possibly driven by age-associated microRNAs in pathways that involve RNA processing, translation, and immune function. We fitted a linear model to predict 'microRNA age' that incorporated expression levels of 80 microRNAs. MicroRNA age correlated modestly with predicted age from DNA methylation (r = 0.3) and mRNA expression (r = 0.2), suggesting that microRNA age may complement mRNA and epigenetic age prediction models. We used the difference between microRNA age and chronological age as a biomarker of accelerated aging (Δage) and found that Δage was associated with all-cause mortality (hazards ratio 1.1 per year difference, P = 4.2 × 10-5 adjusted for sex and chronological age). Additionally, Δage was associated with coronary heart disease, hypertension, blood pressure, and glucose levels. In conclusion, we constructed a microRNA age prediction model based on whole-blood microRNA expression profiling. Age-associated microRNAs and their targets have potential utility to detect accelerated aging and to predict risks for age-related diseases.

EnhancerAtlas: a resource for enhancer annotation and analysis in 105 human cell/tissue types.

Project description:MotivationMultiple high-throughput approaches have recently been developed and allowed the discovery of enhancers on a genome scale in a single experiment. However, the datasets generated from these approaches are not fully utilized by the research community due to technical challenges such as lack of consensus enhancer annotation and integrative analytic tools.ResultsWe developed an interactive database, EnhancerAtlas, which contains an atlas of 2,534,123 enhancers for 105 cell/tissue types. A consensus enhancer annotation was obtained for each cell by summation of independent experimental datasets with the relative weights derived from a cross-validation approach.
