Engineering self-organized criticality in living cells.

ABSTRACT: Complex dynamical fluctuations, from intracellular noise, brain dynamics or computer traffic display bursting dynamics consistent with a critical state between order and disorder. Living close to the critical point has adaptive advantages and it has been conjectured that evolution could select these critical states. Is this the case of living cells? A system can poise itself close to the critical point by means of the so-called self-organized criticality (SOC). In this paper we present an engineered gene network displaying SOC behaviour. This is achieved by exploiting the saturation of the proteolytic degradation machinery in E. coli cells by means of a negative feedback loop that reduces congestion. Our critical motif is built from a two-gene circuit, where SOC can be successfully implemented. The potential implications for both cellular dynamics and behaviour are discussed.

Self-organized criticality in geophysical turbulence.

Project description:Turbulence in geophysical flows tends to organize itself so that the mean flow remains close to a stability boundary in parameter space. That characteristic suggests self-organized criticality (SOC), a statistical property that has been identified in a range of complex phenomena including earthquakes, forest fires and solar flares. This note explores the relationship between the properties of forced, sheared, stratified turbulence (as found in oceans, atmospheres and other geophysical fluids) and those of SOC. Self-organization to the critical state is demonstrated in a wide range of cases drawn mostly (but not entirely) from in situ observations of ocean turbulence. Turbulent events in the ocean also exhibit a second characteristic associated with SOC: their sizes follow a power-law distribution indicating self-similarity. These results suggest SOC as a new conceptual foundation for the study of geophysical turbulence, an explanation for the mixing efficiency of ocean turbulence and a potential for cross-fertilization with other areas of geophysics.

Self-organized criticality theory of autoimmunity.

Project description:BackgroundThe cause of autoimmunity, which is unknown, is investigated from a different angle, i.e., the defect in immune 'system', to explain the cause of autoimmunity.Methodology/principal findingsRepeated immunization with antigen causes systemic autoimmunity in mice otherwise not prone to spontaneous autoimmune diseases. Overstimulation of CD4(+) T cells led to the development of autoantibody-inducing CD4(+) T (aiCD4(+) T) cell which had undergone T cell receptor (TCR) revision and was capable of inducing autoantibodies. The aiCD4(+) T cell was induced by de novo TCR revision but not by cross-reaction, and subsequently overstimulated CD8(+) T cells, driving them to become antigen-specific cytotoxic T lymphocytes (CTL). These CTLs could be further matured by antigen cross-presentation, after which they caused autoimmune tissue injury akin to systemic lupus erythematosus (SLE).Conclusions/significanceSystemic autoimmunity appears to be the inevitable consequence of over-stimulating the host's immune 'system' by repeated immunization with antigen, to the levels that surpass system's self-organized criticality.

Self-organized criticality in developing neuronal networks.

Project description:Recently evidence has accumulated that many neural networks exhibit self-organized criticality. In this state, activity is similar across temporal scales and this is beneficial with respect to information flow. If subcritical, activity can die out, if supercritical epileptiform patterns may occur. Little is known about how developing networks will reach and stabilize criticality. Here we monitor the development between 13 and 95 days in vitro (DIV) of cortical cell cultures (n = 20) and find four different phases, related to their morphological maturation: An initial low-activity state (≈19 DIV) is followed by a supercritical (≈20 DIV) and then a subcritical one (≈36 DIV) until the network finally reaches stable criticality (≈58 DIV). Using network modeling and mathematical analysis we describe the dynamics of the emergent connectivity in such developing systems. Based on physiological observations, the synaptic development in the model is determined by the drive of the neurons to adjust their connectivity for reaching on average firing rate homeostasis. We predict a specific time course for the maturation of inhibition, with strong onset and delayed pruning, and that total synaptic connectivity should be strongly linked to the relative levels of excitation and inhibition. These results demonstrate that the interplay between activity and connectivity guides developing networks into criticality suggesting that this may be a generic and stable state of many networks in vivo and in vitro.

Avalanches and criticality in self-organized nanoscale networks.

Project description:Current efforts to achieve neuromorphic computation are focused on highly organized architectures, such as integrated circuits and regular arrays of memristors, which lack the complex interconnectivity of the brain and so are unable to exhibit brain-like dynamics. New architectures are required, both to emulate the complexity of the brain and to achieve critical dynamics and consequent maximal computational performance. We show here that electrical signals from self-organized networks of nanoparticles exhibit brain-like spatiotemporal correlations and criticality when fabricated at a percolating phase transition. Specifically, the sizes and durations of avalanches of switching events are power law distributed, and the power law exponents satisfy rigorous criteria for criticality. These signals are therefore qualitatively and quantitatively similar to those measured in the cortex. Our self-organized networks provide a low-cost platform for computational approaches that rely on spatiotemporal correlations, such as reservoir computing, and are an important step toward creating neuromorphic device architectures.

Scaling and self-organized criticality in proteins II.

Project description:The complexity of proteins is substantially simplified by regarding them as archetypical examples of self-organized criticality (SOC). To test this idea and to elaborate it, this article applies the Moret-Zebende (MZ) SOC hydrophobicity scale to transport repeat proteins of the HEAT superfamily, importin beta, and transportin, as well as the export protein Cse1p, and their ubiquitous cargo manager Ran. The difference between the MZ scale and conventional hydrophobicity scales reflects long-range conformational forces that are central to protein functionality. These compete with long-range Coulomb forces associated with cationic and anionic side chains in a revealing way.

Developing neuronal networks: self-organized criticality predicts the future.

Project description:Self-organized criticality emerged in neural activity is one of the key concepts to describe the formation and the function of developing neuronal networks. The relationship between critical dynamics and neural development is both theoretically and experimentally appealing. However, whereas it is well-known that cortical networks exhibit a rich repertoire of activity patterns at different stages during in vitro maturation, dynamical activity patterns through the entire neural development still remains unclear. Here we show that a series of metastable network states emerged in the developing and "aging" process of hippocampal networks cultured from dissociated rat neurons. The unidirectional sequence of state transitions could be only observed in networks showing power-law scaling of distributed neuronal avalanches. Our data suggest that self-organized criticality may guide spontaneous activity into a sequential succession of homeostatically-regulated transient patterns during development, which may help to predict the tendency of neural development at early ages in the future.

Failure of adaptive self-organized criticality during epileptic seizure attacks.

Project description:Critical dynamics are assumed to be an attractive mode for normal brain functioning as information processing and computational capabilities are found to be optimal in the critical state. Recent experimental observations of neuronal activity patterns following power-law distributions, a hallmark of systems at a critical state, have led to the hypothesis that human brain dynamics could be poised at a phase transition between ordered and disordered activity. A so far unresolved question concerns the medical significance of critical brain activity and how it relates to pathological conditions. Using data from invasive electroencephalogram recordings from humans we show that during epileptic seizure attacks neuronal activity patterns deviate from the normally observed power-law distribution characterizing critical dynamics. The comparison of these observations to results from a computational model exhibiting self-organized criticality (SOC) based on adaptive networks allows further insights into the underlying dynamics. Together these results suggest that brain dynamics deviates from criticality during seizures caused by the failure of adaptive SOC.

Quantum coherence-driven self-organized criticality and nonequilibrium light localization.

Project description:Self-organized criticality emerges in dynamical complex systems driven out of equilibrium and characterizes a wide range of classical phenomena in physics, geology, and biology. We report on a quantum coherence-controlled self-organized critical transition observed in the light localization behavior of a coherence-driven nanophotonic configuration. Our system is composed of a gain-enhanced plasmonic heterostructure controlled by a coherent drive, in which photons close to the stopped-light regime interact in the presence of the active nonlinearities, eventually synchronizing their dynamics. In this system, on the basis of analytical and corroborating full-wave Maxwell-Bloch computations, we observe quantum coherence-controlled self-organized criticality in the emergence of light localization arising from the synchronization of the photons. It is associated with two first-order phase transitions: one pertaining to the synchronization of the dynamics of the photons and the second pertaining to an inversionless lasing transition by the coherent drive. The so-attained light localization, which is robust to dissipation, fluctuations, and many-body interactions, exhibits scale-invariant power laws and absence of finely tuned control parameters. We also found that, in this nonequilibrium dynamical system, the effective critical "temperature" of the system drops to zero, whereupon one enters the quantum self-organized critical regime.

Growth, collapse, and self-organized criticality in complex networks.

Project description:Network growth is ubiquitous in nature (e.g., biological networks) and technological systems (e.g., modern infrastructures). To understand how certain dynamical behaviors can or cannot persist as the underlying network grows is a problem of increasing importance in complex dynamical systems as well as sustainability science and engineering. We address the question of whether a complex network of nonlinear oscillators can maintain its synchronization stability as it expands. We find that a large scale avalanche over the entire network can be triggered in the sense that the individual nodal dynamics diverges from the synchronous state in a cascading manner within a relatively short time period. In particular, after an initial stage of linear growth, the network typically evolves into a critical state where the addition of a single new node can cause a group of nodes to lose synchronization, leading to synchronization collapse for the entire network. A statistical analysis reveals that the collapse size is approximately algebraically distributed, indicating the emergence of self-organized criticality. We demonstrate the generality of the phenomenon of synchronization collapse using a variety of complex network models, and uncover the underlying dynamical mechanism through an eigenvector analysis.

DOCK8-expressing T follicular helper cells newly generated beyond self-organized criticality cause systemic lupus erythematosus.

Project description:Pathogens including autoantigens all failed to induce systemic lupus erythematosus (SLE). We, instead, studied the integrity of host's immune response that recognized pathogen. By stimulating TCR with an antigen repeatedly to levels that surpass host's steady-state response, self-organized criticality, SLE was induced in mice normally not prone to autoimmunity, wherein T follicular helper (Tfh) cells expressing the guanine nucleotide exchange factor DOCK8 on the cell surface were newly generated. DOCK8+Tfh cells passed through TCR re-revision and induced varieties of autoantibody and lupus lesions. They existed in splenic red pulp and peripheral blood of active lupus patients, which subsequently declined after therapy. Autoantibodies and disease were healed by anti-DOCK8 antibody in the mice including SLE-model (NZBxNZW) F1 mice. Thus, DOCK8+Tfh cells generated after repeated TCR stimulation by immunogenic form of pathogen, either exogenous or endogenous, in combination with HLA to levels that surpass system's self-organized criticality, cause SLE.
