A Machine Vision Approach for Bioreactor Foam Sensing.

ABSTRACT: Machine vision is a powerful technology that has become increasingly popular and accurate during the last decade due to rapid advances in the field of machine learning. The majority of machine vision applications are currently found in consumer electronics, automotive applications, and quality control, yet the potential for bioprocessing applications is tremendous. For instance, detecting and controlling foam emergence is important for all upstream bioprocesses, but the lack of robust foam sensing often leads to batch failures from foam-outs or overaddition of antifoam agents. Here, we report a new low-cost, flexible, and reliable foam sensor concept for bioreactor applications. The concept applies convolutional neural networks (CNNs), a state-of-the-art machine learning system for image processing. The implemented method shows high accuracy for both binary foam detection (foam/no foam) and fine-grained classification of foam levels.

A Vision-Based Sensing Approach for a Spherical Soft Robotic Arm.

Project description:Sensory feedback is essential for the control of soft robotic systems and to enable deployment in a variety of different tasks. Proprioception refers to sensing the robot's own state and is of crucial importance in order to deploy soft robotic systems outside of laboratory environments, i.e. where no external sensing, such as motion capture systems, is available. A vision-based sensing approach for a soft robotic arm made from fabric is presented, leveraging the high-resolution sensory feedback provided by cameras. No mechanical interaction between the sensor and the soft structure is required and consequently the compliance of the soft system is preserved. The integration of a camera into an inflatable, fabric-based bellow actuator is discussed. Three actuators, each featuring an integrated camera, are used to control the spherical robotic arm and simultaneously provide sensory feedback of the two rotational degrees of freedom. A convolutional neural network architecture predicts the two angles describing the robot's orientation from the camera images. Ground truth data is provided by a motion capture system during the training phase of the supervised learning approach and its evaluation thereafter. The camera-based sensing approach is able to provide estimates of the orientation in real-time with an accuracy of about one degree. The reliability of the sensing approach is demonstrated by using the sensory feedback to control the orientation of the robotic arm in closed-loop.

Multiplex Sensing of Complex Mixtures by Machine Vision Analysis of TLC-SERS Images.

Project description:Thin layer chromatography in tandem with surface-enhanced Raman scattering (TLC-SERS) has demonstrated tremendous potentials as a new analytical chemistry tool to detect a wide range of substances from real-world samples. However, it still faces significant challenges of multiplex sensing from complex mixtures due to the imperfect separation by TLC and the resulting interference of SERS detection. In this article, we propose a multiplex sensing method of complex mixtures by machine vision analysis of the scanning image of the TLC-SERS results. Briefly, various pure substances in solution and the complex mixture solution are separated by TLC followed by one-dimensional SERS scanning of the entire TLC plate, which generates TLC-SERS images of all target substances along the chromatography path. After that, a machine vision method is employed to extract the template images from the TLC-SERS images of pure substance solutions. Finally, we apply a feature point matching strategy based on the Winner-take-all principle, which matches the template image of each pure substance with the mixture image to confirm the existence and derive the position of each target substance in the TLC plate, respectively. Our experimental results based on the mixture solution of five different substances show that the proposed machine vision analysis is highly selective, sensitive and does not require artificial analysis of the SERS spectra. Therefore, we envision that the proposed machine vision analysis of the TLC-SERS imaging is an objective, accurate, and efficient method for multiplex sensing of trace level of target substances from complex mixtures.

A machine-vision approach for automated pain measurement at millisecond timescales.

Project description:Objective and automatic measurement of pain in mice remains a barrier for discovery in neuroscience. Here, we capture paw kinematics during pain behavior in mice with high-speed videography and automated paw tracking with machine and deep learning approaches. Our statistical software platform, PAWS (Pain Assessment at Withdrawal Speeds), uses a univariate projection of paw position over time to automatically quantify seven behavioral features that are combined into a single, univariate pain score. Automated paw tracking combined with PAWS reveals a behaviorally divergent mouse strain that displays hypersensitivity to mechanical stimuli. To demonstrate the efficacy of PAWS for detecting spinally versus centrally mediated behavioral responses, we chemogenetically activated nociceptive neurons in the amygdala, which further separated the pain-related behavioral features and the resulting pain score. Taken together, this automated pain quantification approach will increase objectivity in collecting rigorous behavioral data, and it is compatible with other neural circuit dissection tools for determining the mouse pain state.

Production of rhamnolipids by integrated foam adsorption in a bioreactor system.

Project description:Biosurfactants offer environmental as well as health benefits over traditionally used chemical surfactants and heterologous production from engineered microorganisms has been demonstrated, offering containable as well as scalable production of these alternative chemicals. Low product titers and cost intensive downstream processing are the main hurdles for economical biosurfactant production at industrial scales. Increased biosurfactant concentrations are found in the liquid fraction of the foam formed during fermentation of producing microbes. Adsorption of biosurfactants from foam fractions in cultivations may offer a simple concentration and purification method which could enable their cost-effective production. Here, foam adsorption was applied as an in situ method for separation of the rhamnolipid biosurfactants during fermentation of Pseudomonas putida EM383. An integrated process was designed to capture the produced rhamnolipids on hydrophobic adsorbent in packed bed units while minimizing the impact of adsorption on the productivity of the system by recirculating cell-containing collapsed foam flow-through back into the reactor vessel. A stable rhamnolipid production by P. putida EM383 on glucose was performed coupled to this adsorption strategy for 82 h, after which no remaining rhamnolipids were found in the cultivation broth and 15.5 g of rhamnolipids could be eluted from the adsorbent. Rhamnolipid yield from glucose feed was 0.05 g g-1, when up to 2 g L-1 glucose pulse feeding was applied. After solvent evaporation, a product purity of 96% was obtained. The results indicate that the integrated adsorption method can be efficient for simultaneous production and recovery of rhamnolipid biosurfactants from microbial fermentations.

Agile Detection of Chemical Warfare Agents by Machine Vision: a Supramolecular Approach.

Project description:The supramolecular detection by image analysis of a simulant chemical warfare agent on a solid device containing a selective molecular sensor based on a BODIPY scaffold is reported. The recognition properties were investigated in solution, demonstrating high affinity (log K 6.60) and sensitivity (LOD 10 ppt). A test strip also confirmed the sensing properties in gas phase. Image analysis of the solid device allows quantitative information about the simulant to be obtained, recovering the sensor almost 5 times and thus confirming the goal of the supramolecular approach.

New Continuous Process for the Production of Lipopeptide Biosurfactants in Foam Overflowing Bioreactor.

Project description:In this work, an original culture process in bioreactor named overflowing continuous culture (O-CC) was developed to produce and recover continuously mycosubtilin, a lipopeptide antifungal biosurfactant of major interest. The lipopeptide production was first investigated in shake conical flasks in different culture media [ammonium citrate sucrose (ACS), Difco sporulation medium (DSM), and Landy], followed by a pH condition optimization using 3-(N-morpholino)propanesulfonic acid (MOPS) and 2-(N-morpholino)ethanesulfonic acid (MES) buffered media. A simple theoretical modeling of the biomass evolution combined with an experimental setup was then proposed for O-CC processed in stirred tank reactor at laboratory scale. Seven O-CC experiments were done in modified Landy medium at the optimized pH 6.5 by applying dilution rates comprised between 0.05 and 0.1 h-1. The O-CC allowed the continuous recovery of the mycosubtilin contained in the foam overflowing out of the reactor, achieving a remarkable in situ product removal superior to 99%. The biomass concentration in the overflowing foam was found to be twofold lower than the biomass concentration in the reactor, relating advantageously this process to a continuous one with biomass feedback. To evaluate its performances regarding the type of lipopeptide produced, the O-CC process was tested with strain BBG116, a mycosubtilin constitutive overproducing strain that also produces surfactin, and strain BBG125, its derivative strain obtained by deleting surfactin synthetase operon. At a dilution rate of 0.1 h-1, specific productivity of 1.18 mg of mycosubtilin⋅g-1(DW)⋅h-1 was reached. Compared with other previously described bioprocesses using almost similar culture conditions and strains, the O-CC one allowed an increase of the mycosubtilin production rate by 2.06-fold.

Machine-Vision-Enabled Salt Dissolution Analysis.

Project description:Salt formation is a well-established method to increase the solubility of ionizable drug candidates. However, possible conversion of salt to its original form of free acid or base-disproportionation-can have a drastic effect on the solubility and consequently the bioavailability of a drug. Therefore, during the salt selection process, the salt dissolution behavior should be well understood. Improved understanding could be achieved by a method that enables simultaneous screening of small sample amounts and detailed dissolution process analysis. Here, we use a machine-vision-based single-particle analysis (SPA) method to successfully determine the pH-solubility profile, intrinsic solubility, common-ion effect, pKa, pHmax, and Ksp values of three model compounds in a fast and low sample consumption (<1 mg) manner. Moreover, the SPA method enables, with a particle-scale resolution, in situ observation of the disproportionation process and its immediate effect on the morphology and solubility of dissolving species. In this study, a potentially higher energy thermodynamic solid-state form of diclofenac free acid and an intriguing conversion to liquid verapamil free base were observed upon disproportionation of the respective salts. As such, the SPA method offers a low sample consumption platform for fast yet elaborate characterization of the salt dissolution behavior.

Widefield Diamond Quantum Sensing with Neuromorphic Vision Sensors.

Project description:Despite increasing interest in developing ultrasensitive widefield diamond magnetometry for various applications, achieving high temporal resolution and sensitivity simultaneously remains a key challenge. This is largely due to the transfer and processing of massive amounts of data from the frame-based sensor to capture the widefield fluorescence intensity of spin defects in diamonds. In this study, a neuromorphic vision sensor to encode the changes of fluorescence intensity into spikes in the optically detected magnetic resonance (ODMR) measurements is adopted, closely resembling the operation of the human vision system, which leads to highly compressed data volume and reduced latency. It also results in a vast dynamic range, high temporal resolution, and exceptional signal-to-background ratio. After a thorough theoretical evaluation, the experiment with an off-the-shelf event camera demonstrated a 13× improvement in temporal resolution with comparable precision of detecting ODMR resonance frequencies compared with the state-of-the-art highly specialized frame-based approach. It is successfully deploy this technology in monitoring dynamically modulated laser heating of gold nanoparticles coated on a diamond surface, a recognizably difficult task using existing approaches. The current development provides new insights for high-precision and low-latency widefield quantum sensing, with possibilities for integration with emerging memory devices to realize more intelligent quantum sensors.

Remote sensing and computer vision for marine aquaculture.

Project description:Aquaculture, the cultivation of aquatic plants and animals, has grown rapidly since the 1990s, but sparse, self-reported, and aggregated production data limit the effective understanding and monitoring of the industry's trends and potential risks. Building on a manual survey of aquaculture production from remote sensing imagery, we train a computer vision model to identify marine aquaculture cages from aerial and satellite imagery and generate a spatially explicit dataset of finfish production locations in the French Mediterranean from 2000 to 2021 including 4010 cages (average cage area, 69 square meters). We demonstrate the value of our method as an easily adaptable, cost-effective approach that can improve the speed and reliability of aquaculture surveys and enables downstream analyses relevant to researchers and regulators. We illustrate its use to compute independent estimates of production and develop a flexible framework to quantify uncertainty in these estimates. Overall, our study presents an efficient, scalable, and adaptable method for monitoring aquaculture production from remote sensing imagery.

Machine learning approach to monitor inkjet jetting status based on the piezo self-sensing.

Project description:One of the advantages of inkjet printing in digital manufacturing is the ability to use multiple nozzles simultaneously to improve the productivity of the processes. However, the use of multiple nozzles makes inkjet status monitoring more difficult. The jetting nozzles must be carefully selected to ensure the quality of printed products, which is challenging for most inkjet processes that use multi-nozzles. In this article, we improved inkjet print head monitoring based on self-sensing signals by using machine learning algorithms. Specifically, supervised machine learning models were used to classify nozzle jetting conditions. For this purpose, the self-sensing signals were acquired, and the feature information was extracted for training. A vision algorithm was developed to label the nozzle status for classification. The trained models showed that the classification accuracy is higher than 99.6% when self-sensing signals are used for monitoring. We also proposed a so-called hybrid monitoring method using trained machine learning models, which divides the feature space into three regions based on predicted jetting probability: certain jetting, certain non-jetting, and doubt regions. Then, the nozzles with uncertain status in the doubt region can be verified by jet visualization to improve the accuracy and efficiency of the monitoring process.
