Amino Acids Supplementation for the Milk and Milk Protein Production of Dairy Cows.

ABSTRACT: As the preference of consumers for casein products has increased, the protein content of milk from dairy cows is drawing more attention. Protein synthesis in the milk of dairy cows requires a proper supply of dietary protein. High protein supplementation may help to produce more milk protein, but residues in feces and urine cause environmental pollution and increase production costs. As such, previous studies have focused on protein supplements and amino acid (AA) supply. This review concerns AA nutrition for enhancing milk protein in dairy cows, and mainly focuses on three AAs: methionine, lysine, and histidine. AA supplementation for promoting protein synthesis is related to the mammalian target of rapamycin (mTOR) complex and its downstream pathways. Each AA has different stimulating effects on the mTOR translation initiation pathway, and thus manifests different milk protein yields. This review will expand our understanding of AA nutrition and the involved pathways in relation to the synthesis of milk protein in dairy cows.

Project description:Limiting amino acids (AAs) for milk production in dairy cows fed on a concentrate diet of corn grain and soybean meal was evaluated in this study. Four lactating and multiparous Holstein cows (in third or fourth parities, with an average body weight of 633 ± 49.2 kg), 8 to 9 weeks into their lactation period, were used in a 4 × 4 Latin square design. The experiment comprised four dietary treatments: (1) no intravenous infusion (control); (2) control plus intravenous infusion of an AA mixture of 6 g/d methionine, 19.1 g/d lysine, 13.8 g/d isoleucine, and 15.4 g/d valine (4AA); (3) control plus intravenous infusion of the AA mixture without methionine (no-Met); and (4) control plus intravenous infusion of the AA mixture without lysine (no-Lys). All animals were fed on a controlled diet (1 kg/d alfalfa hay, 10 kg/d silage, 14 kg/d concentrate mixture, ad libitum timothy hay). The AA composition of the diet and blood were determined using an automatic AA analyzer. Milk composition (protein, fat, lactose, urea nitrogen, and somatic cell counts) was determined using a MilkoScan. The results showed that feed intake for milk production did not differ from that of intravenous infusion using a limiting AA mixture. The 4AA treatment numerically had the highest milk yield (32.4 kg/d), although there was no difference when compared with the control (31.2 kg/d), no-Met (31.3 kg/d), and no-Lys (31.7 kg/d) treatments. The concentration of AAs in blood plasma of cows in all treatments, mainly isoleucine and valine, increased significantly compared with that of control. The no-Met treatment increased (p < 0.05) the concentration of lysine in the blood relative to the control and no-Lys treatments, whereas the no-Lys treatment increased (p < 0.05) the concentration of methionine relative to the control and no-Met treatments. In conclusion, milk production increased when feeding 10 g/d methionine to the cows, together with their concentrate diet of corn grain and soybean meal.

Project description:BACKGROUND: Oriental theileriosis is a tick-borne, protozoan disease of cattle caused by members of the Theileria orientalis-complex. Recent outbreaks of this disease in eastern Australia have caused major concerns to the dairy and beef farming communities, but there are no published studies of the economic impact of this disease. On a farm in Victoria, Australia, we assessed whether oriental theileriosis has an impact on milk production and reproductive performance in dairy cows. METHODS: Blood samples collected from all 662 cows on the farm were tested using an established molecular test. For individual cows, milk production and reproductive performance data were collected. A clinical assessment of individual cows was performed. Based on clinical findings and molecular test results, the following groups of cows were classified: group 1, with cardinal clinical signs of oriental theileriosis and molecular test-positive for T. orientalis; group 2, with mild or suspected signs of theileriosis and test-positive; group 3, with no clinical signs and test-positive; and group 4, with no clinical signs and test-negative. Milk production and reproductive performance data for groups 1, 2 and 3 were each compared with those for group 4 using linear and logistic regression analyses, respectively. RESULTS: At 100 days of lactation, group 1 cows produced significantly less milk (288 l; P = 0.001), milk fat (16.8 kg; P < 0.001) and milk protein (12.6 kg; P < 0.001) compared with group 4. At this lactation point, group 2 also produced significantly less milk fat (13.6 kg; P = 0.002) and milk protein (8.6 kg; P = 0.005) than group 4. At 305 days of lactation, group 1 cows produced significantly less milk (624 l; P = 0.004), milk fat (42.9 kg; P < 0.001) and milk protein (26.0 kg; P < 0.001) compared with group 4 cows. Group 2 cows also produced significantly less milk fat (21.2 kg; P = 0.033) at this lactation point. No statistically significant difference in reproductive performance was found upon pairwise comparisons of groups 1-3 with group 4 cows. CONCLUSIONS: The present findings demonstrate that clinical oriental theileriosis can cause significant milk production losses in dairy cattle.

Project description:Milk production by dairy cows is sensitive to increased levels of stress hormones such as glucocorticoids (GC) that also regulate the transcription of several genes required for milk synthesis. Whereas previous studies identified that an exogenous GC such as dexamethasone (DEX) transiently suppresses milk yield in several species without any pronounced effect on milk protein or fat percentage, the mechanism underlying this effect has not been established. In this study we sought to establish changes within the mammary glands of non-pregnant dairy cows in their second lactation (n = 3-4; 648-838 kg) following a single dose of exogenous DEX. Changes in the udder were monitored by serial biopsy of alternating quarters, concurrent with quarter-level monitoring of milk yield and composition. Dexamethasone increased serum glucose levels from 12-36 h (p <0 .05), reduced milk yield from 12-48 h (p <0 .05), increased % milk protein content at 24 h post-DEX, and transiently decreased both milk lactose and α-lactalbumin content, while not altering the level of milk fat. After 72 h, all aspects of milk production had returned to pre-treatment levels. Transcriptomic changes in the mammary glands in response to DEX were identified by RNA sequencing followed by differential gene expression analysis. Coincident with the milk yield and composition changes was the differential expression of 519 and 320 genes at 12 and 24 h after DEX (adjusted p <0 .05), respectively, with the return of all gene expression to baseline levels by 72 h. Among the transcriptomic changes in response to DEX, there was notable downregulation of elements in the lactose synthesis pathway, specifically AQP3, GALE and LALBA (α-lactalbumin) at 12 h, and sustained downregulation of LALBA at 24 h. One gene in the pathway, UGP2, was upregulated at 12-24 h post-DEX. This work supports the hypothesis that there is a direct relationship between the response to DEX and the concurrent suppression of milk yield due to the reduced synthesis of α-lactalbumin and lactose by the mammary epithelium. The ability of glucocorticoids to modulate the homeorrhetic requirements for glucose during stressful states concurrent with immune activation bears significance for dairy animals as well as a broad range of lactating mammals.

Project description:IntroductionChromium (Cr) is an essential mineral that has been demonstrated to enhance milk production in dairy cows. This study aims to evaluate the effects of dietary Cr supplementation on dry matter intake (DMI), milk production and composition using a meta-analysis based on existing literature.MethodsA random effects meta-analysis was performed to investigate the effects of dietary Cr supplementation on DMI, milk production and composition. The heterogeneity was assessed using the I 2 statistic and Q test, while Egger's test was used to evaluate publication bias.ResultsThe meta-analysis discovered that Cr-supplemented cows had a significantly higher DMI compared to those not supplemented, with an increase of 0.72 kg/day [95% confidence interval (CI), 0.46-0.97]. The regression model indicated that DMI significantly increased by 0.9 g/kg of body weight (BW) and by 80.5 g for an increase of 1 mg of Cr supplement. The supplementation phase was associated with an increase in DMI, with an increase of 0.4582 kg/day for BFP (before parturition) and 0.853 kg/day for AFP (after parturition). The methionine and yeast forms of Cr increased DMI by 0.714 and 1.137 kg/day, respectively. The DMI was increased by 2.137 and 0.620 kg/day for multiparous (MP)+ primiparous (PP) cows and MP cows, respectively. Milk production was also increased by Cr supplementation, with an increase of 1.20 kg/day (95% CI, 0.65-1.76). The regression model indicated that milk production increased by 2.3 g/day for an increase of 1 kg of BW and by 122.4 g/day for an increase of 1 mg of Cr supplement. Milk production also increased with the duration of the experiment and days in milk. The amino acid and methionine forms of Cr complexes increased milk production by 1.645 and 1.448 kg/day, respectively. Milk production increased by 1.087 and 1.920 kg/day for MP and PP cows, respectively. Milk composition was not significantly affected by Cr supplementation. Egger's test for publication biases was not significant for all responses of interest.DiscussionThe meta-analysis showed that Cr supplementation improves DMI and milk production in dairy cows. The results suggest that the supplementation phase, form of Cr, and parity should be considered when supplementing dairy cows with Cr. The results have important implications for the dairy industry and can contribute to the development of more effective feeding strategies for dairy cows.
