The complete mitochondrial genome of Cochylimorpha cultana (Lederer, 1855) (Lepidoptera: Tortricidae).

ABSTRACT: The complete mitochondrial genome of Cochylimorpha cultana (Lederer) (Lepidoptera: Tortricidae) was 15,348 bps in size, and comprised 37 genes, which were 13 PCGs, 22 tRNA genes and two rRNA genes. Most PCGs used the conventional ATN start codon, except for cox1 initiating with CGA. Four genes (cox1, cox2, nad4 and nad5) used single T residue as stop condon. 21 out of 22 tRNAs are folded into the cloverleaf secondary structure, except for trnS1. The phylogenetic analysis based on maximum-likelihood (ML) method revealed that the evolutionary status of C. cultana in Tortricinae at the molecular level, which agrees well with the classical taxonomy.

Project description:We describe the complete mitochondrial genome of Phiaris dolosana. It is 15,562 bp in length, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rRNAs), and a 612 bp D-Loop. All PCGs start with ATN codon except for COI gene, which uses CGA as the initiation codon. Nine of 13 PCGs use a typical stop codon of TAA, the rest use incomplete stop codon of T or TA. Phylogenetic analysis of P. dolosana with other 17 leaf rollers is conducted with neighbor-joining method, the result is consistent with the conventional classification.

Project description:Choristoneura metasequoiacola Liu, 1983 is an important caterpillar species that specifically infests the leaves and branches of Metasequoia glyptostroboides Hu & W. C. Cheng 1948 with short larval infestations, long-term dormancy, and has a limited distribution in Lichuan, Hubei, China. The complete mitochondria genome of C. metasequoiacola was determined by using Illumina NovaSeq, and analyzed based on previously annotated sibling species. In total, we obtained mitochondria genome with 15,128 bp in length, circular in shape with a double-stranded closed ring structure, including 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and an AT-rich region. Of which the nucleotide composition was highly A + T biased, accounting for 81.98% of the whole mitogenome. Thirteen protein-coding genes (PCGs) were 11,142 bp; Twenty-two tRNA genes and AT-rich region were 1,472 and 199 bp, respectively. Phylogenetically, the relationship between Choristoneura spp. (containing C. metasequoiacola) and Adoxophyes spp. was closer than any other two genera from Tortricidae, and the relationship between C. metasequoiacola and C.murinana was the closest among nine sibling species from that genus, which helps to explain species evolution within the family Tortricidae.

The genome is 15,433 bp long with a high A + T content (80.6%), and consists of 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and a noncoding control region. A phylogenetic analysis of 18 tortricid species for which mitogenes are available showed strong support for the monophyly of Tortricinae.

The complete mitochondrial genome of a leaf roller, Eudemis lucina (Lepidoptera: Tortricidae).

Project description:The leaf roller, Eudemis lucina, is a potential pest of Quercus in East Asia. In this study, we described the complete mitochondrial genome of this species by high-throughput sequencing. The mitochondrial genome is found to be a circular molecule of 16,056 bp in length, which consisted of 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes, and a non-coding control region (A + T-rich region). The A + T content is 80.5% for the whole mitogenome. All PCGs are initiated by ATN codons, except for COI which is initiated by the CGA codon. Eight PCGs use a typical stop codon of TAA, whereas the remaining PCGs use incomplete stop codon of T-- or TA-. The non-coding control region is 1013 bp and located between s-rRNA and Met-tRNA.

The complete mitochondrial genome of Clepsis pallidana (Lepidoptera: Tortricidae) with phylogenetic implications.

Project description:The leaf roller, Clepsis pallidana (Fabricius, 1776) (Lepidoptera: Tortricidae), is an important pest of many crops in China. The circular genome is 15,679 bp in length, consisting of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a non-coding AT-rich region. The base composition of the whole mitogenome is 41.2% A, 40.1% T, 11.1% C, and 7.7% G, which shows a strong AT bias. Phylogenetic analysis based on 13 PCGs showed that C. pallidana was closely related to Epiphyas postvittana and clustered within Archipini clade, which was consistent with the traditional classification.

The complete mitochondrial genome of the oriental fruit moth Grapholita molesta (Busck) (Lepidoptera: Tortricidae).

Project description:The oriental fruit moth, Grapholita molesta (Busck) (Lepidoptera: Tortricidae) currently is one of the economically most destructive pest species of stone and pome fruits worldwide. Here we sequenced the complete mitochondrial genome of this pest. This genome is 15,776 bp long, with an A + T content of 81.24%, containing 37 typical animal mitochondrial genes and an A + T-rich region. All gene are arranged as hypothesized ancestral gene order of insects except for trnM, which was shuffled from 3' downstream of trnQ to 5' upstream of trnI. cox1 gene uses unusual CGA start codon, as that in all other sequenced lepidopteran mitochondrial genome. The secondary structures for the two rRNA genes were predicted. All helices typically present in insect mitochondrial rRNA genes are generated. A microsatellite sequence was inserted into the region of H2347 in rrnL in G. molesta and two other sequenced tortricid mitochondrial genomes, indicating that the insertion event in this helix might occurred anciently in family Tortricidae. All of the 22 typical animal tRNA genes have a typical cloverleaf structure except for trnS2, in which the D-stem pairings in the DHU arm are absent. An intergenic sequence is present between trnQ and nad2 as well as in other sequenced lepidopteran mitochondrial genomes, which was presumed to be a remnant of trnM gene and its boundary sequences after the duplication of trnM to the upstream of trnI in Lepidoptera. The A + T-rich region is 836 bp, containing six repeat sequences of "TTATTATTATTATTAAATA(G)TTT."

First complete mitochondrial genome of Antheraea pernyi Guerin-Meneville 1855 (Lepidoptera: Saturniidae) from North Korea.

Project description:This study reports the first complete mitochondrial genome of Antheraea pernyi Guérin-Méneville 1855 (Lepidoptera: Saturniidae) strain Dingzhou_1, a silkworm resource serving silk production in North Korea. The mitochondrial genome is circular with 15,573 bp in length encoding 37 typical mitochondrial genes (13 protein-coding genes, two ribosomal RNA genes, and 22 transfer RNA genes). The 553-bp A + T-rich region harbors a repeat region composed of 6 ∼ 38 bp tandem repeat units, as found in other known inbred strains from Chinese populations. The phylogenetic analysis clustered Dingzhou_1 from North Korea together with Chinese Liaoning population, suggesting that oak silkworm in North Korea might be introduced from her neighbor China Liaoning.

The complete mitochondrial genome of the cocoa tussock moth Orgyia postica Walker, 1855 (Lepidoptera: Lymantriidae).

Project description:The high-throughput sequencing method was used for the first time to determined complete mitochondrial genome of Orgyia postica. The complete mitochondrial genome was a circular molecule with 15,258 bp in full-length, including 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs) and 2 ribosomal RNA genes (rRNAs). The nucleotide composition of O. postica mitogenome was A: 37.6%, C: 7.9%, G: 14.8%, T: 39.7%. In addition, the results of the phylogenetic analysis indicate that O. postica had the closest relationship with Laelia suffusa. Our study will contribute to further research on the evolution of Lymantriidae and the identification of larva species.

The complete mitochondrial genome of Rondotia menciana (Lepidoptera: Bombycidae).

Project description:The mulberry white caterpillar, Rondotia menciana Moore (Lepidoptera: Bombycidae) is a species with closest relationship with Bombyx mori and Bombyx mandarina, and the genetic information of R. menciana is important for understanding the diversity of the Bombycidae. In this study, the mitochondrial genome (mitogenome) of R. menciana was amplified by polymerase chain reaction and sequenced. The mitogenome of R. menciana was determined to be 15,301 bp, including 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes, and an AT-rich region. The A+T content (78.87%) was lower than that observed for other Bombycidae insects. All PCGs were initiated by ATN codons and terminated with the canonical stop codons, except for coxII, which was terminated by a single T. All the tRNA genes displayed a typical clover-leaf structure of mitochondrial tRNA. The length of AT-rich region (360 bp) of R. menciana mitogenome is shorter than that of other Bombycidae species. Phylogenetic analysis showed that the R. menciana was clustered on one branch with B. mori and B. mandarina from Bombycidae.
