Poly(ADP-ribose) polymerase (PARP) enzymes initiate (mt)DNA repair mechanisms and use nicotinamide adenine dinucleotide (NAD<sup>+</sup>) as energy source. Prolonged PARP activity can drain cellular NAD<sup>+</sup> reserves, leading to de-regulation of important molecular processes.
