Project description:Multiple comprehensive in situ bio-optical investigations were conducted from 2005 to 2010 and covered a large variability of total suspended matter (TSM) in Lake Taihu to calibrate and validate a TSM concentration estimation model based on Medium Resolution Imaging Spectrometer (MERIS) data. The estimation model of the TSM concentration in Lake Taihu was developed using top-of-atmosphere (TOA) radiance of MERIS image data at band 9 in combination with a regional empirical atmospheric correction model, which was strongly correlated with the in situ TSM concentration (r(2) = 0.720, p<0.001, and n = 73). The relative root mean square error (RRMSE) and mean relative error (MRE) were 36.9% and 31.6%, respectively, based on an independent validation dataset that produced reliable estimations of the TSM concentration. The developed algorithm was applied to 50 MERIS images from 2003 to 2011 to obtain a high spatial and temporal heterogeneity of TSM concentrations in Lake Taihu. Seasonally, the highest and lowest TSM concentrations were found in spring and autumn, respectively. Spatially, TSM concentrations were high in the southern part and center of the lake and low in Xukou Bay, East Lake Taihu. The lake topography, including the water depth and distance from the shore, had a significant effect on the TSM spatial distribution. A significant correlation was found between the daily average wind speed and TSM concentration (r(2)= 0.685, p<0.001, and n = 50), suggesting a critical role of wind speed in the TSM variations in Lake Taihu. In addition, a low TSM concentration was linked to the appearance of submerged aquatic vegetation (SAV). Therefore, TSM dynamics were controlled by the lake topography, wind-driven sediment resuspension and SAV distribution.

Project description:MicroRNA have been recently discovered in human milk signifying potentially important functions for both the lactating breast and the infant. Whilst human milk microRNA have started to be explored, little data exist on the evaluation of sample processing, and analysis to ensure that a full spectrum of microRNA can be obtained. Human milk comprises three main fractions: cells, skim milk, and lipids. Typically, the skim milk fraction has been measured in isolation despite evidence that the lipid fraction may contain more microRNA. This study aimed to standardize isolation of microRNA and total RNA from all three fractions of human milk to determine the most appropriate sampling and analysis procedure for future studies. Three different methods from eight commercially available kits were tested for their efficacy in extracting total RNA and microRNA from the lipid, skim, and cell fractions of human milk. Each fraction yielded different concentrations of RNA and microRNA, with the highest quantities found in the cell and lipid fractions, and the lowest in skim milk. The column-based phenol-free method was the most efficient extraction method for all three milk fractions. Two microRNAs were expressed and validated in the three milk fractions by qPCR using the three recommended extraction kits for each fraction. High expression levels were identified in the skim and lipid milk factions for these microRNAs. These results suggest that careful consideration of both the human milk sample preparation and extraction protocols should be made prior to embarking upon research in this area.

Project description:Gross primary production (GPP) is a fundamental ecosystem process that sequesters carbon dioxide (CO2) and forms the resource base for higher trophic levels. Still, the relative contribution of different controls on GPP at the whole-ecosystem scale is far from resolved. Here we show, by manipulating CO2 concentrations in large-scale experimental pond ecosystems, that CO2 availability is a key driver of whole-ecosystem GPP. This result suggests we need to reformulate past conceptual models describing controls of lake ecosystem productivity and include our findings when developing models used to predict future lake ecosystem responses to environmental change.

Project description:Studies of normal human mammary gland development and function have mostly relied on cell culture, limited surgical specimens, and rodent models. Although RNA extracted from human milk has been used to assay the mammary transcriptome non-invasively, this assay has not been adequately validated in primates. Thus, the objectives of the current study were to assess the suitability of lactating rhesus macaques as a model for lactating humans and to determine whether RNA extracted from milk fractions is representative of RNA extracted from mammary tissue for the purpose of studying the transcriptome of milk-producing cells.We confirmed that macaque milk contains cytoplasmic crescents and that ample high-quality RNA can be obtained for sequencing. Using RNA sequencing, RNA extracted from macaque milk fat and milk cell fractions more accurately represented RNA from mammary epithelial cells (cells that produce milk) than did RNA from whole mammary tissue. Mammary epithelium-specific transcripts were more abundant in macaque milk fat, whereas adipose or stroma-specific transcripts were more abundant in mammary tissue. Functional analyses confirmed the validity of milk as a source of RNA from milk-producing mammary epithelial cells.RNA extracted from the milk fat during lactation accurately portrayed the RNA profile of milk-producing mammary epithelial cells in a non-human primate. However, this sample type clearly requires protocols that minimize RNA degradation. Overall, we validated the use of RNA extracted from human and macaque milk and provided evidence to support the use of lactating macaques as a model for human lactation.

Conversely, higher C/P ratio increased the storage yield but lowered the productivity (1.55 gPHA L-1 h-1), compromising sludge settleability and reactor stability, indicating impaired microbial functionality. Transmission electron microscopy further confirmed the presence of intracellular PHA granules and polyphosphate reserves, reinforcing the connection between nutrient limitation and adaptive microbial strategies. Overall, these findings highlight the critical role of the C/P ratio in shaping the performance of mixed microbial cultures, demonstrating that a well-balanced nutrient supply can enhance PHA production while maintaining microbial community stability. Project description:Interest is growing for the development of inclusive seed production models. However, there is limited understanding of gender-based roles and constraints and how these might influence gender relations in seed production. Through a case study on sweetpotato seed production in Lake Zone Tanzania, this article examines men's and women's roles in seed production with the introduction of specialized seed practices and a commercial orientation. The study uses data from 17 field-based plot observations and eight sex disaggregated focus group discussions (FGDs) with 33 (51% women and 48% men) decentralized vine multipliers (DVMs). Participatory, gender-based analytical tools were used to obtain an in-depth understanding of gender dimensions and implications of new seed production practices, the resources required and access to those resources. Our findings show that men and women have complementary roles in specialized seed production, and that men increased their involvement in production and commercialization, especially when larger monetary inputs and transactions took place. Women gained new tangible (income) and intangible (knowledge) assets, which enhanced their community status. Women's contributions to household income became more visible. In conclusion male-takeover did not take place. There were changes in the perceptions around sweetpotato production and gender relations. As women's contributions to household income became more visible, they were able to negotiate with their husbands on access to key resources to maintain this household revenue stream. We discuss how the new knowledge and skills related to seed production enhanced women's status in the community. Project description:Widespread lake drainage can lead to large-scale drying in Arctic lake-rich areas, affecting hydrology, ecosystems and permafrost carbon dynamics. To date, the spatio-temporal distribution, driving factors, and post-drainage dynamics of lake drainage events across the Arctic remain unclear. Using satellite remote sensing and surface water products, we identify over 35,000 (~0.6% of all lakes) lake drainage events in the northern permafrost zone between 1984 and 2020, with approximately half being relatively understudied non-thermokarst lakes. Smaller, thermokarst, and discontinuous permafrost area lakes are more susceptible to drainage compared to their larger, non-thermokarst, and continuous permafrost area counterparts. Over time, discontinuous permafrost areas contribute more drained lakes annually than continuous permafrost areas. Following drainage, vegetation rapidly colonizes drained lake basins, with thermokarst drained lake basins showing significantly higher vegetation growth rates and greenness levels than their non-thermokarst counterparts. Under warming, drained lake basins are likely to become more prevalent and serve as greening hotspots, playing an important role in shaping Arctic ecosystems.

Project description:Eastern equine encephalitis virus (EEEV) infection results in high mortality in infected horses and humans. Florida has been identified as an important source of EEEV epidemics to other states in the United States. In this study, we further characterized the epidemiological and evolutionary dynamics of EEEV in Florida. Epidemiological analysis of sentinel chicken seroconversion rates to EEEV infections during 2005-2016 suggested significant seasonality of EEEV activity in Florida. We observed significant annual activity of EEEV in the North and North Central regions, with little significant seasonality in the Panhandle region. Phylogenetic analysis of complete EEEV genome sequences from different host sources and regions in Florida during 1986-2014 revealed extensive genetic diversity and spatial dispersal of the virus within Florida and relatively more clustering of the viruses in the Panhandle region. We found no significant association between EEEV genetic variation and host source. Overall, our study revealed a complex epidemiological dynamic of EEEV within Florida, implicating the Panhandle region as a possible source of the virus with sustained year-round transmission. These findings will help in implementing targeted control measures that can have the most impact in reducing or eliminating EEEV and other mosquito-borne viral infections within Florida and in the rest of the United States.
