The complete mitochondrial genome of leafhopper Koreocerus koreanus (Matsumura, 1915) (Hemiptera: Cicadellidae).

ABSTRACT: In this study, the complete mitochondrial genome (mitogenome) of Koreocerus koreanus (Mastsumura, 1915) was sequenced with 16,428 bp in length, which consisted of 37 genes including 22 transfer RNAs genes (tRNAs), 13 protein coding genes (PCGs) and 2 ribosomal RNAs genes (rRNAs). Except COX1 and ATP6 initiated with the TTG, other PCGs started with ATN. The COX1, ND4 and ND4L terminated with an incomplete T, while the remaining PCGs stopped with TAA. Excluding tRNA-Ser1 lacking a DHU arm, 21 tRNA genes with secondary structures were typical of cloverleaf structures. Phylogenetic analysis based on the complete mitochondrial sequences of 28 species (26 Cicadellidae and 2 outgroups) showed that Koreocerus koreanus was clustered into the subfamily Idiocerinae, the complete mitogenome of Koreocerus koreanus provides information for further study of Cicadellidae evolution.

Project description:We have sequenced the complete mitochondrial genome (mitogenome) of the leafhopper Idioscopus clypealis in Idiocerinae, the mitochondrial genome sequences to be 15,393 bp with an A + T content of 78.3%. The mitogenome includes 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNA (tRNA) genes, and one non-coding [A + T]-rich region. These genes are in the same order as in the Idioscopus nitidulus. All protein-coding genes have ATN as the start codon, TAA and single T as the stop codon. The phylogenetic tree confirms that I. clypealis and I. nitidulus are clustered into a clade, and provides the relationships between the Megophthalminae, Deltocephalinae, Idiocerinae, Cicadellinae, and Typhloeybinae.

The complete mitochondrial genome of the leafhopper Evacanthus acuminatus (Hemiptera: Cicadellidae: Evacanthinae).

Project description:The complete mitogenome of Evacanthus acuminatus was described in this study. The circular molecule is 14,793 bp in length (GenBank accession no. MK948205), and contains 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and an A + T-rich region. The base composition values were A (40.0%), C (11.2%), G (10.0%), and T (38.8%), with an A + T bias (78.8%). ATT, ATG, ATA, TTG were initiation codons and TAA, TAG, T were termination codons. Phylogenetic tree based on concatenated amino acid sequences of 13 PCGs shows that E. acuminatu and Idiocerinae clade form sister groups, and all species of different subfamilies congregate with each other, which is consistent with the traditional taxonomy.

Complete Mitogenome Analysis of Five Leafhopper Species of Idiocerini (Hemiptera: Cicadellidae).

Project description:Insect mitochondrial genomes (mitogenomes) are of great interest in exploring molecular evolution, phylogenetics, and biogeography. So far, only 12 mitogenomes of the leafhopper tribe Idiocerini have been released in GenBank, although the tribe comprises 488 known species including some agricultural, forestry, and horticultural pests. In order to compare and analyze the mitochondrial genome structure of Idiocerini and even the selective pressure of 13 protein-coding genes (PCGs) of the family Cicadellidae, the complete mitogenomes of five species including Nabicerus dentimus, Sahlbergotettix salicicola, Podulmorinus opacus, Podulmorinus consimilis, and a new species of a new genus were determined by next-generation sequencing. The size of the newly determined mitogenomes ranged from 14,733 bp to 15,044 bp, comprising the standard set of 13 PCGs, 22 transfer RNA genes, two ribosomal RNA genes, and a long non-coding control region (CR). The extent of purifying selection presented different pictures in the tribe and the family. The less pronounced genes (0.5 < dN/dS < 1) were nad5 and nad4l in Idiocerin, whereas in the family Cicadellidae including the sequences of Idiocerin, nad1-nad6 and cox1 genes were less pronounced. The codon encoding leucine was the most common in all species, and the codon encoding serine 1 was the most common in all species except for P. opacus. Interestingly, in P. opacus, another of the most common codons is that encoding serine 2. Among the 17 examined species of the Idiocerini, 14 species contained the tandem repeats, and 11 species of them contained the motif "TTATA". These findings will promote research on the structure and evolution of the mitochondrial genome and highlight the need for more mitogenomes in Cicadellidae.

The phylogenetic analyses based on concatenated thirteen protein-coding genes of mitogenomes recover the monophyly of Auchenorrhyncha (Fulgoromorpha + Cicadomorpha) and Sternorrhyncha as a sister group to Auchenorrhyncha. Project description:The complete mitochondrial genome of the Aster leafhopper Macrosteles quadrilineatus was sequenced using an Illumina-based next-generation sequencing approach. The genome is 16,626 bp in length with 78.0% AT content. It encodes 37 typical mitochondrial genes including 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 1 A + T-rich region. Two tandem repeats were identified within the A + T-rich region. One tRNA gene rearrangement (trnW-trnC-trnY→trnC-trnW-trnY) was found between nd2 and cox1.

Complete mitochondrial genome of Populicerus confusus (Hemiptera: Cicadellidae: Idiocerinae).

Project description:In this study, we firstly reported the complete mitochondrial genome of Populicerus confuses. The complete mitochondrial genome was 16,395 bp in length which overall base composition was 41.43% A, 36.30% T, 11.54% C, and 10.73% G. It consisted of 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes (12S and 16S rRNA), and a control region (D-loop region). The complete mitochondrial genomes of P. confuses and other 9 species were used for phylogenetic analysis using the Bayesian method. The resulting phylogenetic tree confirms that the Populicerus populi is most closely related to P. confuses. The mitogenome provided the valuable evidence on phylogenetic relationship of the Idiocerinae at the molecular level.

Identification and Evaluation of Sugarcane Cultivars for Antixenosis Resistance to the Leafhopper Yamatotettix flavovittatus Matsumura (Hemiptera: Cicadellidae).

Project description:Understanding the settling preference, feeding behavior, honeydew production, and biophysical factors, such as trichome density, related to Y. flavovittatus leafhopper infestation in sugarcane cultivation is crucial for effective pest management strategies. This study investigated these aspects across nine sugarcane cultivars. Significant variability was observed among cultivars in terms of settling behavior, with KK3 and LK92-11 showing the highest number of settled leafhopper adults. Similarly, honeydew production varied significantly among cultivars, with KK3 and LK92-11 exhibiting the highest production. Employing the electrical penetration graph (EPG) technique provided insights into distinct probing behaviors across cultivars, highlighting correlations between settling preference, honeydew production, and specific EPG waveforms. Principal component analysis (PCA) categorized cultivars into four groups based on settling preference, honeydew production, feeding behavior, and biophysical factors. Strong correlations were found between settling preference, honeydew production, and various EPG waveforms, while negative correlations were observed with the number of silica cells and rows per unit area, indicating their potential role in deterring leafhopper settlement. We concluded that TPJ04-768 and K84-200 are promising for resistance against leafhoppers and, thereby, can be exploited in sugarcane breeding programs with regard to resistance against insects.

Complete mitochondrial genome sequence of Empoascanara dwalata (Hemiptera: Cicadellidae: Typhlocybinae).

Project description:In this study, the complete mitochondrial genome of the leafhopper species Empoascanara dwalata (Hemiptera: Cicadellidae: Typhlocybinae) was sequenced and annotated, and the sequence is available in the GenBank with accession number: MT350235. The circular mitogenome with 15,271 bp in length, including 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and one AT-rich region. The nucleotide composition is as follows: A (41.7%), T (34.7%), G (10.6%), and C (13.0%). 11 PCGs have ATN as the start codon, except for atp8 and nad5 genes have TTG. The conventional termination codons (TAA or TAG) occur in 11 PCGs, while cox2 and nad5 use incomplete codon (T--) as termination codon. Phylogenetic analysis using 13 PCGs showed that E. dwalata was clustered with three other Typhlocybine species, which was consistent with the conventional classification.

The complete mitochondrial genome of Fieberiella septentrionalis (Hemiptera: Cicadellidae: Deltocephalinae).

Project description:The complete mitochondrial genome of Fieberiella septentrionalis was annotated for the first time in the present study. The mitogenome was found to have circular shape, with 16,175 bp in size, containing 13 protein coding genes (PCGs), 22 transfer-RNA genes, 2 ribosomal-RNA genes, and 1 non-coding region. The nucleotide composition biases toward A and T is 77.9% of the entirety, which is a typical structure of Cicadellidae. All PCGs have ATN as the start codon, TAA and single T as the stop codon. The resulting phylogenetic tree confirms that the F. septentrionalis belongs to the subfamily of Deltocephalinae and Fieberiellini as sister to the remaining tribes of this subfamily.

Complete mitochondrial genome of Penthimia melanocephala Motschulsky, 1863 (Hemiptera: Cicadellidae: Deltocephalinae).

Project description:Penthimia is the largest genus in the tribe Penthimiini of the subfamily Deltocephalinae. To date, there are no available mitogenome sequences from Penthimia. In this study, we have sequenced and annotated the complete mitogenome of Penthimia melanocephala (Motschulsky 1863), the mitogenome is 15,308 bp in length, which including 13 protein-coding genes (PCGs), 2 ribosomal RNA genes (rRNAs), 22 transfer RNAs (tRNAs) and a long non-coding region (control Region), base composition of whole sequences are A (50.4%), C (15.3%), G (10.0%), and T (24.3%). All PCGs started with the typical ATN codon and stopped with the typical TAN codon except for COII and COIII stopped with single T. Within phylogenetic tree, Deltocephalinae members were clustered into a clade. The complete mitogenome of P. melanocephala can provide essential DNA molecular data for further evolutionary and phylogenetic analysis.
