The complete mitochondrial genome of a planarian flatworm Girardia tigrina (Tricladida: Dugesiidae).

ABSTRACT: Girardia tigrina, a freshwater planarian species native to America and introduced to other continents, has been usually used as model organism in many research fields of biology. In this study, we determined the complete mitochondrial genome of G. tigrina using next-generation sequencing (NGS). The complete mitogenome was 15,938 bp in length, with 36 genes, including 12 protein-coding genes, 2 ribosomal RNAs and 22 transfer RNAs, and ATP8 was absent in the mitogenome of G. tigrina as in the mitogenomes of some other flatworms. The maximum-likelihood phylogenetic tree suggested that G. tigrina was closely related to genus Dugesia in the clade of Tricladida.

Complete genome sequence of a novel extrachromosomal virus-like element identified in planarian Girardia tigrina.

Project description:BackgroundFreshwater planarians are widely used as models for investigation of pattern formation and studies on genetic variation in populations. Despite extensive information on the biology and genetics of planaria, the occurrence and distribution of viruses in these animals remains an unexplored area of research.ResultsUsing a combination of Suppression Subtractive Hybridization (SSH) and Mirror Orientation Selection (MOS), we compared the genomes of two strains of freshwater planarian, Girardia tigrina. The novel extrachromosomal DNA-containing virus-like element denoted PEVE (Planarian Extrachromosomal Virus-like Element) was identified in one planarian strain. The PEVE genome (about 7.5 kb) consists of two unique regions (Ul and Us) flanked by inverted repeats. Sequence analyses reveal that PEVE comprises two helicase-like sequences in the genome, of which the first is a homolog of a circoviral replication initiator protein (Rep), and the second is similar to the papillomavirus E1 helicase domain. PEVE genome exists in at least two variant forms with different arrangements of single-stranded and double-stranded DNA stretches that correspond to the Us and Ul regions. Using PCR analysis and whole-mount in situ hybridization, we characterized PEVE distribution and expression in the planarian body.ConclusionsPEVE is the first viral element identified in free-living flatworms. This element differs from all known viruses and viral elements, and comprises two potential helicases that are homologous to proteins from distant viral phyla. PEVE is unevenly distributed in the worm body, and is detected in specific parenchyma cells.

Physiological evaluation of the behavior and epidermis of freshwater planarians (Girardia tigrina and Girardia sp.) exposed to stressors.

Project description:Planarians are metazoan freshwater flatworms which are free-living organisms. Their body has pluripotent stem cell promoters of tissue regeneration capacity. The water temperature and the potential of hydrogen (pH) of lentic ecosystems are important factors involved in the distribution and abundance of these animals. Although the pH factor is directly related to the physiology and behavior of planarians, their adaptive and regenerating capacities still remain unknown. The Critical Thermal Maximum (CTM) is a very widespread method used in the evaluation of thermal tolerance. In this study, Girardia tigrina (Girard, 1850) and Girardia sp., a species found in Brazil, which is under study as a new species, had their epidermis assessed by scanning electron microscopy (SEM) to analyze their physiological structures before and after exposure to different stressors. SEM was used as a method to evaluate the planarians' epidermis as a result of the increasing temperature (CTM) and pH alterations, the latter with the use of a new methodology defined as Critical Hydrogen ion concentration Maximum (CHM). In increasing temperatures from 20°C to 37°C, both Girardia tigrina and Girardia sp. proved to be adaptable to thermal stress. Girardia sp. was shown to be more resistant to higher temperatures. However, Girardia tigrina was more resistant to extreme pH conditions (4.0 to 10.0). SEM analysis showed morphological differences among planarian species, such as the arrangement of the structures and cell types of the dorsal epidermis. Moreover, planarians demonstrated the ability to change the surrounding pH of their external environment in order to maintain the function of their physiological mechanisms, suggesting that these animals have a complex survival system, possibly related to protonephridia, flame cells and excretory pores.This article has an associated First Person interview with the first author of the paper.

A new and aberrant species of Dugesia (Platyhelminthes, Tricladida, Dugesiidae) from Madagascar.

Project description:In this paper we report a new species of Dugesia of the family Dugesiidae from Madagascar, representing the fourth species of freshwater planarian known from this global biodiversity hotspot. In some respects the new species is aberrant, when compared with its congeners, being characterized by a head with smoothly rounded auricles, a peculiar course of the oviducts, including the presence of a common posterior extension, and by the asymmetrical openings of the vasa deferentia at about halfway along the seminal vesicle. Further, it is characterized by a ventral course of the ejaculatory duct with a terminal opening, very long spermiducal vesicles and unstalked cocoons. Its diploid chromosome complement with 18 chromosomes represents an uncommon feature among fissiparous species of Dugesia.

The taxonomic status of Dugesiabiblica from Israel and Turkey (Platyhelminthes, Tricladida, Dugesiidae).

Project description:The taxonomic status of Dugesiabiblica (Platyhelminthes, Tricladida, Dugesiidae) from Israel and Turkey is problematic due to its morphological similarity with Dugesiasicula since these nominal species present overlapping characters. In this study we analyzed histological preparations of specimens of these two nominal species and also compared mitochondrial COI gene sequences from Israeli populations to the already known haplotype composition of Dugesiasicula. We concluded that these animals belong to the same species and therefore we consider Dugesiabiblica to be a junior synonym of Dugesiasicula. This implies that the distribution range of Dugesiasicula is even wider than previously thought, and that the species is present all around the Mediterranean Basin and on many of its islands.

Project description:For the first time, we report the complete mitochondrial genome (mitogenome) sequence of the marine triclad species, Obrimoposthia wandeli. The complete mitogenome of O. wandeli was 15,185 bp in length, contains 12 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), and 2 ribosomal RNAs (rRNAs). Compared to previously reported gene orders from the order Tricladida, O. wandeli had unique gene order. We constructed a phylogenetic tree based on the mitogenomes belonging to Rhabdocoela, Polycladida, and Tricladida and confirmed that O. wandeli is located in the basal position in the Tricladida.
