Consistent tumorigenesis with self-assembled hydrogels enables high-powered murine cancer studies.

ABSTRACT: Preclinical cancer research is heavily dependent on allograft and xenograft models, but current approaches to tumor inoculation yield inconsistent tumor formation and growth, ultimately wasting valuable resources (e.g., animals, time, and money) and limiting experimental progress. Here we demonstrate a method for tumor inoculation using self-assembled hydrogels to reliably generate tumors with low variance in growth. The observed reduction in model variance enables smaller animal cohorts, improved effect observation and higher powered studies.

Self-assembled nematic colloidal motors powered by light.

Project description:Biological motors are marvels of nature that inspire creation of their synthetic counterparts with comparable nanoscale dimensions, high efficiency and diverse functions. Molecular motors have been synthesized, but obtaining nanomotors through self-assembly remains challenging. Here we describe a self-assembled colloidal motor with a repetitive light-driven rotation of transparent micro-particles immersed in a liquid crystal and powered by a continuous exposure to unstructured ~1 nW light. A monolayer of azobenzene molecules defines how the liquid crystal's optical axis mechanically couples to the particle's surface, as well as how they jointly rotate as the light's polarization changes. The rotating particle twists the liquid crystal, which changes polarization of traversing light. The resulting feedback mechanism yields a continuous opto-mechanical cycle and drives the unidirectional particle spinning, with handedness and frequency robustly controlled by polarization and intensity of light. Our findings may lead to opto-mechanical devices and colloidal machines compatible with liquid crystal display technology.

High-performance compliant thermoelectric generators with magnetically self-assembled soft heat conductors for self-powered wearable electronics.

Project description:Softening of thermoelectric generators facilitates conformal contact with arbitrary-shaped heat sources, which offers an opportunity to realize self-powered wearable applications. However, existing wearable thermoelectric devices inevitably exhibit reduced thermoelectric conversion efficiency due to the parasitic heat loss in high-thermal-impedance polymer substrates and poor thermal contact arising from rigid interconnects. Here, we propose compliant thermoelectric generators with intrinsically stretchable interconnects and soft heat conductors that achieve high thermoelectric performance and unprecedented conformability simultaneously. The silver-nanowire-based soft electrodes interconnect bismuth-telluride-based thermoelectric legs, effectively absorbing strain energy, which allows our thermoelectric generators to conform perfectly to curved surfaces. Metal particles magnetically self-assembled in elastomeric substrates form soft heat conductors that significantly enhance the heat transfer to the thermoelectric legs, thereby maximizing energy conversion efficiency on three-dimensional heat sources. Moreover, automated additive manufacturing paves the way for realizing self-powered wearable applications comprising hundreds of thermoelectric legs with high customizability under ambient conditions.

Tunable Pentapeptide Self-Assembled β-Sheet Hydrogels.

Project description:Oligopeptide-based supramolecular hydrogels hold promise in a range of applications. The gelation of these systems is hard to control, with minor alterations in the peptide sequence significantly influencing the self-assembly process. We explored three pentapeptide sequences with different charge distributions and discovered that they formed robust, pH-responsive hydrogels. By altering the concentration and charge distribution of the peptide sequence, the stiffness of the hydrogels could be tuned across two orders of magnitude (2-200 kPa). Also, through reassembly of the β-sheet interactions the hydrogels could self-heal and they demonstrated shear-thin behavior. Using spectroscopic and cryo-imaging techniques, we investigated the relationship between peptide sequence and molecular structure, and how these influence the mechanical properties of the hydrogel. These pentapeptide hydrogels with tunable morphology and mechanical properties have promise in tissue engineering, injectable delivery vectors, and 3D printing applications.

Self-assembled hydrogels utilizing polymer-nanoparticle interactions.

Project description:Mouldable hydrogels that flow on applied stress and rapidly self-heal are increasingly utilized as they afford minimally invasive delivery and conformal application. Here we report a new paradigm for the fabrication of self-assembled hydrogels with shear-thinning and self-healing properties employing rationally engineered polymer-nanoparticle (NP) interactions. Biopolymer derivatives are linked together by selective adsorption to NPs. The transient and reversible interactions between biopolymers and NPs enable flow under applied shear stress, followed by rapid self-healing when the stress is relaxed. We develop a physical description of polymer-NP gel formation that is utilized to design biocompatible gels for drug delivery. Owing to the hierarchical structure of the gel, both hydrophilic and hydrophobic drugs can be entrapped and delivered with differential release profiles, both in vitro and in vivo. The work introduces a facile and generalizable class of mouldable hydrogels amenable to a range of biomedical and industrial applications.

Co-adsorbed self-assembled monolayer enables high-performance perovskite and organic solar cells.

Project description:Self-assembled monolayers (SAMs) have become pivotal in achieving high-performance perovskite solar cells (PSCs) and organic solar cells (OSCs) by significantly minimizing interfacial energy losses. In this study, we propose a co-adsorb (CA) strategy employing a novel small molecule, 2-chloro-5-(trifluoromethyl)isonicotinic acid (PyCA-3F), introducing at the buried interface between 2PACz and the perovskite/organic layers. This approach effectively diminishes 2PACz's aggregation, enhancing surface smoothness and increasing work function for the modified SAM layer, thereby providing a flattened buried interface with a favorable heterointerface for perovskite. The resultant improvements in crystallinity, minimized trap states, and augmented hole extraction and transfer capabilities have propelled power conversion efficiencies (PCEs) beyond 25% in PSCs with a p-i-n structure (certified at 24.68%). OSCs employing the CA strategy achieve remarkable PCEs of 19.51% based on PM1:PTQ10:m-BTP-PhC6 photoactive system. Notably, universal improvements have also been achieved for the other two popular OSC systems. After a 1000-hour maximal power point tracking, the encapsulated PSCs and OSCs retain approximately 90% and 80% of their initial PCEs, respectively. This work introduces a facile, rational, and effective method to enhance the performance of SAMs, realizing efficiency breakthroughs in both PSCs and OSCs with a favorable p-i-n device structure, along with improved operational stability.

Ion-Triggered Hydrogels Self-Assembled from Statistical Copolypeptides.

Project description:Statistical copolypeptides comprising lysine and tyrosine with unprecedented ion-induced gelation behavior are reported. Copolypeptides are obtained by one-step N-carboxyanhydride (NCA) ring-opening polymerization. The gelation mechanism is studied by in situ SAXS analyses, in addition to optical spectroscopy and transmission electron microscopy (TEM). It is found that the gelation of these statistically polymerized polypeptides is due to the formation of stable intermolecular β-sheet secondary structures induced by the presence of salt ions as well as the aggregation of an α-helix between the copolypeptides. This behavior is unique to the statistical lysine/tyrosine copolypeptides and was not observed in any other amino acid combination or arrangement. Furthermore, the diffusion and mechanical properties of these hydrogels can be tuned through tailoring the polypeptide chain length and ion strength.

Magnetic Dehydrodipeptide-Based Self-Assembled Hydrogels for Theragnostic Applications.

Project description:Self-assembled peptide hydrogels have emerged in recent years as the new paradigm in biomaterials research. We have contributed to this field the development of hydrogels based on dehydrodipeptides N-capped with naproxen. The dehydrodipeptide hydrogels can be loaded with drugs, thus being potential nanocarriers for drug delivery. In this work novel dehydrodipeptides containing tyrosine and aspartic acid amino acid residues N-capped with naproxen and C-terminal dehydrophenylalanine were prepared and characterized. Superparamagnetic iron oxide nanoparticles (SPIONs) were incorporated into the dehydrodipeptide-based hydrogels and their effect on the self-assembly, structure and rheological and magnetic properties of the hydrogels was studied. Magnetic hydrogels, with incorporated SPIONs, displayed concentration-dependent T₂-MRI contrast enhancement. Moreover, upon magnetic excitation (alternating magnetic field -AMF-) the SPIONs were able to generate a significant amount of heat. Hence, magnetic hyperthermia can be used as a remote trigger for release of drug cargos and SPIONs incorporated into the self-assembled dehydrodipeptide hydrogels.

Self-Assembled Monolayer Enables Slurry-Coating of Li Anode.

Project description:Li metal has long been considered as the ultimate anodic material for high-energy-density batteries. Protection of Li metal in electrochemical cycling and in the manufacturing environment is critical for practical applications. Here, we present the passivation of the Li metal-carbon nanotube (CNT) composite with molecular self-assembly of a long-chain aliphatic phosphonic acid. The dynamics of the self-assembly process is investigated with sum-frequency generation spectroscopy (SFG). The aliphatic phosphonic acid molecules self-assemble on the Li metal surface via the lithium phosphate bonding, while the well-aligned long chains of the molecules help to prevent corrosion of lithium by oxygen and water in the air. As a result, the self-assembled monolayer (SAM) passivated Li-CNT composite displays excellent stability in dry or even humid air, and could be slurry-coated with organic solvents. The resulting slurry-coated Li anode exhibits a high Coulombic efficiency of 98.8% under a 33% depth of discharge (DOD) at a 1C rate in full battery cycling. The concept of molecular self-assembly on Li metal and the stability of the resulting SAM layer open vast possibilities of designed reagents for surface passivation of Li, which may eventually pave the way for practical application of Li metal in secondary batteries.

Self assembled bi-functional peptide hydrogels with biomineralization-directing peptides.

Project description:A peptide-based hydrogel has been designed that directs the formation of hydroxyapatite. MDG1, a twenty-seven residue peptide, undergoes triggered folding to form an unsymmetrical beta-hairpin that self-assembles in response to an increase in solution ionic strength to yield a mechanically rigid, self supporting hydrogel. The C-terminal portion of MDG1 contains a heptapeptide (MLPHHGA) capable of directing the mineralization process. Circular dichroism spectroscopy indicates that the peptide folds and assembles to form a hydrogel network rich in beta-sheet secondary structure. Oscillatory rheology indicates that the hydrogel is mechanically rigid (G' 2500Pa) before mineralization. In separate experiments, mineralization was induced both biochemically and with cementoblast cells. Mineralization-domain had little effect on the mechanical rigidity of the gel. SEM and EDXS show that MDG1 gels are capable of directing the formation of hydroxapatite. Control hydrogels, prepared by peptides either lacking the mineral-directing portion or reversing its sequence, indicated that the heptapeptide is necessary and its actions are sequence specific.

Anthranilamide-based Short Peptides Self-Assembled Hydrogels as Antibacterial Agents.

Project description:In this study, we describe the synthesis and molecular properties of anthranilamide-based short peptides which were synthesised via ring opening of isatoic anhydride in excellent yields. These short peptides were incorporated as low molecular weight gelators (LMWG), bola amphiphile, and C3-symmetric molecules to form hydrogels in low concentrations (0.07-0.30% (w/v)). The critical gel concentration (CGC), viscoelastic properties, secondary structure, and fibre morphology of these short peptides were influenced by the aromaticity of the capping group or by the presence of electronegative substituent (namely fluoro) and hydrophobic substituent (such as methyl) in the short peptides. In addition, the hydrogels showed antibacterial activity against S. aureus 38 and moderate toxicity against HEK cells in vitro.
