Plasmonic Nanotweezers and Nanosensors for Point-of-Care Applications.

ABSTRACT: The capabilities of manipulating and analyzing biological cells, bacteria, viruses, DNAs, and proteins at high resolution are significant in understanding biology and enabling early disease diagnosis. We discuss progress in developments and applications of plasmonic nanotweezers and nanosensors where the plasmon-enhanced light-matter interactions at the nanoscale improve the optical manipulation and analysis of biological objects. Selected examples are presented to illustrate their design and working principles. In the context of plasmofluidics, which merges plasmonics and fluidics, the integration of plasmonic nanotweezers and nanosensors with microfluidic systems for point-of-care (POC) applications is envisioned. We provide our perspectives on the challenges and opportunities in further developing and applying the plasmofluidic POC devices.

Femtosecond-pulsed plasmonic nanotweezers.

Project description:We demonstrate for the first time plasmonic nanotweezers based on Au bowtie nanoantenna arrays (BNAs) that utilize a femtosecond-pulsed input source to enhance trapping of both Rayleigh and Mie particles. Using ultra-low input power densities, we demonstrate that the high-peak powers associated with a femtosecond source augment the trap stiffness to 2x that of nanotweezers employing a continuous-wave source, and 5x that of conventional tweezers using a femtosecond source. We show that for trapped fluorescent microparticles the two-photon response is enhanced by 2x in comparison to the response without nanoantennas. We also demonstrate tweezing of 80-nm diameter Ag nanoparticles, and observe an enhancement of the second-harmonic signal of ~3.5x for the combined nanoparticle-BNA system compared to the bare BNAs. Finally, under select illumination conditions, fusing of Ag nanoparticles to the BNAs is observed which holds potential for in situ fabrication of three-dimensional, bimetallic nanoantennas.

Dispersion and shape engineered plasmonic nanosensors.

Project description:Biosensors based on the localized surface plasmon resonance (LSPR) of individual metallic nanoparticles promise to deliver modular, low-cost sensing with high-detection thresholds. However, they continue to suffer from relatively low sensitivity and figures of merit (FOMs). Herein we introduce the idea of sensitivity enhancement of LSPR sensors through engineering of the material dispersion function. Employing dispersion and shape engineering of chiral nanoparticles leads to remarkable refractive index sensitivities (1,091 nm RIU(-1) at λ=921 nm) and FOMs (>2,800 RIU(-1)). A key feature is that the polarization-dependent extinction of the nanoparticles is now characterized by rich spectral features, including bipolar peaks and nulls, suitable for tracking refractive index changes. This sensing modality offers strong optical contrast even in the presence of highly absorbing media, an important consideration for use in complex biological media with limited transmission. The technique is sensitive to surface-specific binding events which we demonstrate through biotin-avidin surface coupling.

From saliva to SNP: non-invasive, point-of-care genotyping for precision medicine applications using recombinase polymerase amplification and giant magnetoresistive nanosensors.

Project description:Genetic testing is considered a cornerstone of the precision medicine paradigm. Genotyping of single nucleotide polymorphisms (SNPs) has been shown to provide insights into several important issues, including therapy selection and drug responsiveness. However, a scarcity of widely deployable and cost-effective genotyping tools has limited the integration of precision medicine into routine clinical practice. The objective of our work was to develop a portable, cost-effective, and automated platform that performs SNP genotyping at the point-of-care (POC). Using recombinase polymerase amplification (RPA) and giant magnetoresistive (GMR) nanosensors, we present a highly automated and multiplexed point-of-care platform that utilizes direct saliva for the qualitative genotyping of four SNPs (rs4633, rs4680, rs4818, rs6269) along the catechol-O-methyltransferase gene (COMT), which is associated with the modulation of pain sensitivity and perioperative opioid use. Using this approach, we successfully amplify, detect, and genotype all four of the SNPs, demonstrating 100% accordance between the experimental results obtained using the automated RPA and GMR genotyping assay and the results obtained using a COMT PCR genotyping assay that was formerly validated using pyrosequencing. This automated, portable, and multiplexed RPA and GMR assay shows great promise as a solution for SNP genotyping at the POC and reinforces the broad applications of magnetic nanotechnology in biomedicine.

Point of Care 3D Ultrasound for Various Applications: A Pilot Study

Project description:Summary 1. Purpose and Objective: The purpose of this study is to test the feasibility of rapid acquisition of point of care 3D ultrasound in obtaining abdominal and/or pelvic images. The study will use a newly developed acquisition method and post-processing technique to create three dimensional image models of the abdomen and/or pelvis. 2. Study activities and population group. The study population will be a convenience sample of patients of any age presenting to the Emergency Department with complaints necessitating a clinical abdominal and/or pelvic imaging. The study intervention includes acquisition of research ultrasound images, which will not be used for clinical care, and comparison of these images with clinically obtained images. Other clinical data such as surgical and pathology reports will also be reviewed. 3.Data analysis and risk/safety issues. This is a pilot study intended to determine feasibility and to refine image reconstruction algorithms. Research images will be compared to clinical images. Comparison of research images with final diagnosis will also occur. The research intervention, an ultrasound exam, has no known safety risks. The only risk to subjects is loss of confidentiality. This study is observational, not interventional, because the experimental ultrasound will be performed in all subjects and will not be used in the clinical care of patients (consequently, will not have the opportunity to affect clinical outcomes). Experimental images will be reviewed after completion of clinical care and will not be provided to the clinicians caring for the subjects. The investigators are not measuring the effect of the ultrasound examination on the subjects’ outcomes.

Dual-Enhanced Plasmonic Biosensing for Point-of-Care Sepsis Detection.

Project description:Rapid, sensitive, simultaneous quantification of multiple biomarkers in point-of-care (POC) settings could improve the diagnosis and management of sepsis, a common, potentially life-threatening condition. Compared to high-end commercial analytical systems, POC systems are often limited by low sensitivity, limited multiplexing capability, or low throughput. Here, we report an ultrasensitive, multiplexed plasmonic sensing technology integrating chemifluorescence signal enhancement with plasmon-enhanced fluorescence detection. Using a portable imaging system, the dual chemical and plasmonic amplification enabled rapid analysis of multiple cytokine biomarkers in 1 h with sub-pg/mL sensitivities. Furthermore, we also developed a plasmonic sensing chip based on nanoparticle-spiked gold nanodimple structures fabricated by wafer-scale batch processes. We used the system to detect six cytokines directly from clinical plasma samples (n = 20) and showed 100% accuracy for sepsis detection. The described technology could be employed in rapid, ultrasensitive, multiplexed plasmonic sensing in POC settings for myriad clinical conditions.

Metal-Organic Frameworks for Preserving the Functionality of Plasmonic Nanosensors.

Project description:Preserving the functionality of nanosensors is critical for their reliable performance under harsh environmental conditions. Biofunctionalized plasmonic nanostructures are an important class of bionanoconjugates for biosensing, bioimaging, and nanotherapeutics. Plasmonic nanostructures and biomolecules exhibit poor thermal stability over time. Here, we report a class of metal-organic framework, zeolitic imidazolate framework-8 (ZIF-8), as a protective coating for preserving plasmonic nanostructures and plasmonic bionanoconjugates at elevated temperature. Gold nanobipyramids (AuNBPs) with sharp tips are attractive plasmonic nanotransducers with high sensitivity but are prone to structural change and loss of sensitivity. This work reports the first observation that ZIF-8 can preserve the structure of AuNBPs and their corresponding strong electromagnetic field enhancement and high refractive index sensitivity. In addition, ZIF-8 coating enables nearly 100% retention of biorecognition capability of antibodies immobilized on the AuNBP surface after exposure to 60 °C for 48 h. The efficacy, versatility, and facile implementation of ZIF-8 coating offer great promise for the preservation of nanosensors.

Buoyant particulate strategy for few-to-single particle-based plasmonic enhanced nanosensors.

Project description:Detecting matter at a single-molecule level is the ultimate target in many branches of study. Nanosensors based on plasmonics have garnered significant interest owing to their ultrahigh sensitivity even at single-molecule level. However, currently, plasmonic-enhanced nanosensors have not achieved excellent performances in practical applications and their detection at femtomolar or attomolar concentrations remains highly challenging. Here we show a plasmonic sensing strategy, called buoyant plasmonic-particulate-based few-to-single particle-nanosensors. Large-sized floating particles combined with a slippery surface may prevent the coffee-ring effect and enhance the spatial enrichment capability of the analyte in plasmonic sensitive sites via the aggregation and lifting effect. Dimer and single particle-nanosensors demonstrate an enhanced surface-enhanced Raman spectroscopy (SERS) and a high fluorescence sensitivity with an enrichment factor up to an order of ∼104 and the limit of detection of CV molecules down to femto- or attomolar levels. The current buoyant particulate strategy can be exploited in a wide range of plasmonic enhanced sensing applications for a cost-effective, simple, fast, flexible, and portable detection.

Thermally-actuated microfluidic membrane valve for point-of-care applications.

Project description:Microfluidics has enabled low volume biochemistry reactions to be carried out at the point-of-care. A key component in microfluidics is the microfluidic valve. Microfluidic valves are not only useful for directing flow at intersections but also allow mixtures/dilutions to be tuned real-time and even provide peristaltic pumping capabilities. In the transition from chip-in-a-lab to lab-on-a-chip, it is essential to ensure that microfluidic valves are designed to require less peripheral equipment and that they are transportable. In this paper, a thermally-actuated microfluidic valve is presented. The valve itself is fabricated with off-the-shelf components without the need for sophisticated cleanroom techniques. It is shown that multiple valves can be controlled and operated via a power supply and an Arduino microcontroller; an important step towards transportable microfluidic devices capable of carrying out analytical assays at the point-of-care. It is been calculated that a single actuator costs less than $1, this highlights the potential of the presented valve for scaling out. The valve operation is demonstrated by adjusting the ratio of a water/dye mixture in a continuous flow microfluidic chip with Y-junction channel geometry. The power required to operate one microfluidic valve has been characterised both theoretically and experimentally. Cyclical operation of the valve has been demonstrated for 65 h with 585 actuations. The presented valve is capable of actuating rectangular microfluidic channels of 500 μm × 50 μm with an expected temperature increase of up to 5 °C. The fastest actuation times achieved were 2 s for valve closing (heating) and 9 s for valve opening (cooling).

Opto-thermoelectric nanotweezers.

Project description:Optical manipulation of plasmonic nanoparticles provides opportunities for fundamental and technical innovation in nanophotonics. Optical heating arising from the photon-to-phonon conversion is considered as an intrinsic loss in metal nanoparticles, which limits their applications. We show here that this drawback can be turned into an advantage, by developing an extremely low-power optical tweezing technique, termed opto-thermoelectric nanotweezers (OTENT). Through optically heating a thermoplasmonic substrate, alight-directed thermoelectric field can be generated due to spatial separation of dissolved ions within the heating laser spot, which allows us to manipulate metal nanoparticles of a wide range of materials, sizes and shapes with single-particle resolution. In combination with dark-field optical imaging, nanoparticles can be selectively trapped and their spectroscopic response can be resolved in-situ. With its simple optics, versatile low-power operation, applicability to diverse nanoparticles, and tuneable working wavelength, OTENT will become a powerful tool in colloid science and nanotechnology.
