The genome is 132,710 bp long and contains 144 open reading frames. Project description:Fall armyworm (FAW; Spodoptera frugiperda), an exotic moth which recently invaded Africa, is a highly destructive pest of cereals especially maize a highly valued staple crop in Nigeria. The use of natural enemies such as predators or parasitoids for FAW control is more economically viable and environmentally safer than currently recommended synthetic insecticides. Natural enemies to combat the pest have not yet been reported in Nigeria. An exploration for the pests' natural enemies was undertaken by collecting FAW eggs and larvae from maize fields. These were reared in the laboratory for emergence, identification and efficacy as natural enemies. This yielded Euplectrus laphygmae (Hymenoptera: Eulophidae); Telenomus remus (Hymenoptera: Platygastridae) and Trombidium sp. (Acari.: Trombidiidae). Cotesia or Apanteles spp. were inferred to occur since Stictopisthus sp. (Hym.: Ichneumonidae), a secondary parasitoid, that attacks cocoons of Microgasterinae (e.g. Cotesia, Apanteles etc.) also emerged. Species of yet-to-be identified predators were also observed in various niches of maize plants. A positive relationship was found between FAW instar and the number of E. laphygmae eggs/instar ranging, on average, from 1.5 on second instar to 5.5 on fourth instars hosts. Parasitism rate of T. remus on FAW eggs was 100%. Parasitic mite infestation resulted in increasing paleness, reduced feeding, growth and movement as well as death of FAW 1st instars. Thus, the occurrence of FAW natural enemies in Nigeria calls for advocacy campaign to incorporate their use into integrated pest management strategies that attract and allow natural enemies to thrive for FAW management.

Project description:The fall armyworm, Spodoptera frugiperda, is an important agricultural pest native to tropical and subtropical regions of the Western Hemisphere, and has invaded Africa and further spread into most countries of Asia within two years. Here, we analyzed the genetic variation of invaded populations by comparing the nucleotide sequences of two genes: the nuclear Z-chromosome linked gene triose phosphate isomerase (Tpi) and the mitochondrial gene cytochrome oxidase subunit I (COI) of 27 specimens collected in Africa (DR Congo, Tanzania, Uganda, and Zimbabwe) and Asia (Bangladesh, Korea, Nepal, and Vietnam). The results revealed that 25 specimens were from a heterogeneous hybrid (Tpi-corn strain and COI-rice strain; Tpi-C/COI-R) of the corn strain male and rice strain female, but two specimens were from a homogenous corn strain (Tpi-corn strain and COI-corn strain; Tpi-C/COI-C). The further analysis of the fourth exon and the fourth intron sequences of the Tpi gene identified at least four subgroups of the corn strain. These four genetic subgroups were identified in Africa and Asia, suggesting no significant genetic change due to the rapid migration within two years. Our study provides essential information for understanding the genetic diversity of fall armyworm in new habitats.

Project description:In the Americas, the fall armyworm (Spodoptera frugiperda) exists in two genetically distinct strains, the corn (C) and rice (R) strains. Despite their names, these strains are not associated with host plant preferences but have been shown to vary in pheromone composition and male responses. Recently, S. frugiperda was detected in Africa as an invasive species, but knowledge about variation in strain types, pheromone composition and inter-strain mating of populations of the pest in the continent has not been fully examined. Therefore, this study aimed to investigate variations, if any in the pheromone composition of female moths, male moth responses, and mating between C and R mitotypes of S. frugiperda populations in Kenya, as well as their geographic distribution. Strains (mitotypes) of S. frugiperda were identified using mitochondrial DNA (mtDNA) markers, and their pheromonal composition determined by coupled gas chromatography-mass spectrometric (GC-MS) analysis. Male moth responses to these compounds were evaluated using GC-electroantennographic detection (EAD), electroantennogram (EAG), and wind tunnel assays. Oviposition assays were used to determine whether R and C mitotype moths could mate and produce eggs. The results showed that both the R and C mitotypes were present, and there were no statistically significant differences in their distribution across all sampled locations. Five pheromone compounds including (Z)-7-dodecenyl acetate (Z7-12:OAc), (Z)-7-tetradecenyl acetate (Z7-14:OAc), (Z)-9-tetradecenyl acetate (Z9-14:OAc), (Z)-11-tetradecenyl acetate (Z11-14:OAc) and (Z)-11-hexadecenyl acetate (Z11-16:OAc), were detected in the pheromone glands of female moths of both mitotypes, with Z9-14:OAc being the most abundant. The relative percentage composition of Z9-14:OAc was similar in both mitotypes. However, the R mitotype had a 2.7 times higher relative percentage composition of Z7-12:OAc compared to the C mitotype moth, while the C mitotype moth had a 2.4 times higher relative percentage composition of Z11-16:OAc than the R mitotype moth. Male moths of both mitotypes exhibited similar responses to the pheromone compounds, showing the strongest responses to Z9-14:OAc and Z7-12:OAc in electrophysiological and behavioural assays. There was mating between R and C mitotypes with egg production comparable to mating within the same mitotype. Our results revealed that differences between the two S. frugiperda mitotypes are characterized by female moth pheromone composition rather than male moth responses to the pheromones, and that this does not prevent hybridisation between the mitotypes, which may have implications for their management.

Project description:BackgroundLittle is known about the genome sequences of lepidopteran insects, although this group of insects has been studied extensively in the fields of endocrinology, development, immunity, and pathogen-host interactions. In addition, cell lines derived from Spodoptera frugiperda and other lepidopteran insects are routinely used for baculovirus foreign gene expression. This study reports the results of an expressed sequence tag (EST) sequencing project in cells from the lepidopteran insect S. frugiperda, the fall armyworm.ResultsWe have constructed an EST database using two cDNA libraries from the S. frugiperda-derived cell line, SF-21. The database consists of 2,367 ESTs which were assembled into 244 contigs and 951 singlets for a total of 1,195 unique sequences.ConclusionS. frugiperda is an agriculturally important pest insect and genomic information will be instrumental for establishing initial transcriptional profiling and gene function studies, and for obtaining information about genes manipulated during infections by insect pathogens such as baculoviruses.

Project description:With the development of globalization and agriculture trade, as well as its own strong migratory capacity, fall armyworm (FAW) (Spodoptera frugiperda) (J.E. Smith) has invaded more than 70 countries, posing a serious threat to the production of major crops in these areas. FAW has now also been detected in Egypt in North Africa, putting Europe, which is separated from it only by the Mediterranean Sea, at high risk of invasion. Therefore, this study integrated multiple factors of insect source, host plant, and environment to provide a risk analysis of the potential trajectories and time periods of migration of FAW into Europe in 2016~2022. First, the CLIMEX model was used to predict the annual and seasonal suitable distribution of FAW. The HYSPLIT numerical trajectory model was then used to simulate the possibility of the FAW invasion of Europe through wind-driven dispersal. The results showed that the risk of FAW invasion between years was highly consistent (P<0.001). Coastal areas were most suitable for the expansion of the FAW, and Spain and Italy had the highest risk of invasion, with 39.08% and 32.20% of effective landing points respectively. Dynamic migration prediction based on spatio-temporal data can enable early warning of FAW, which is important for joint multinational pest management and crop protection.

Project description:The fall armyworm (FAW), Spodoptera frugiperda (Lepidoptera: Noctuidae) is a recent invasive pest species that has successfully established across sub-Saharan Africa where it continues to disrupt agriculture, particularly smallholder cereal production. Management of FAW in its native range in the Americas has led to the development of resistance to many commercial pesticides before its arrival in Africa. Pesticide use may therefore be ineffective for FAW control in Africa, so new and more sustainable approaches to pest management are required that can help reduce the impact of this exotic pest. Pesticidal plants provide an effective and established approach to pest management in African smallholder farming and recent research has shown that their use can be cost-beneficial and sustainable. In order to optimize the use of botanical extracts for FAW control, we initially screened ten commonly used plant species. In laboratory trials, contact toxicity and feeding bioassays showed differential effects. Some plant species had little to no effect when compared to untreated controls; thus, only the five most promising plant species were selected for more detailed study. In contact toxicity tests, the highest larval mortality was obtained from Nicotiana tabacum (66%) and Lippia javanica (66%). Similarly, in a feeding bioassay L. javanica (62%) and N. tabacum (60%) exhibited high larval mortality at the highest concentration evaluated (10% w/v). Feeding deterrence was evaluated using glass-fibre discs treated with plant extracts, which showed that Cymbopogon citratus (36%) and Azadirachta indica (20%) were the most potent feeding deterrents among the pesticidal plants evaluated. In a screenhouse experiment where living maize plants infested with fall armyworm larvae were treated with plant extracts, N. tabacum and L. javanica were the most potent species at reducing foliar damage compared to the untreated control whilst the synthetic pesticide chlorpyrifos was the most effective in reducing fall armyworm foliar damage. Further field trial evaluation is recommended, particularly involving smallholder maize fields to assess effectiveness across a range of contexts.

Project description:The fall armyworm (FAW; Spodoptera frugiperda) is one of the major agricultural pest insects. FAW is native to the Americas, and its invasion was first reported in West Africa in 2016. Then it quickly spread through Africa, Asia, and Oceania, becoming one of the main threats to corn production. We analyzed whole genome sequences of 177 FAW individuals from 12 locations on four continents to infer evolutionary processes of invasion. Principal component analysis from the TPI gene and whole genome sequences shows that invasive FAW populations originated from the corn strain. Ancestry coefficient and phylogenetic analyses from the nuclear genome indicate that invasive populations are derived from a single ancestry, distinct from native populations, while the mitochondrial phylogenetic tree supports the hypothesis of multiple introductions. Adaptive evolution specific to invasive populations was observed in detoxification, chemosensory, and digestion genes. We concluded that extant invasive FAW populations originated from the corn strain with potential contributions of adaptive evolution.

Project description:The fall armyworm (FAW), Spodoptera frugiperda J.E. Smith, has caused massive maize losses since its attack on the African continent in 2016, particularly in east Africa. In this study, we predicted the spatial distribution (established habitat) of FAW in five east African countries viz., Kenya, Tanzania, Rwanda, Uganda, and Ethiopia. We used FAW occurrence observations for three years i.e., 2018, 2019, and 2020, the maximum entropy (MaxEnt) model, and bioclimatic, land surface temperature (LST), solar radiation, wind speed, elevation, and landscape structure data (i.e., land use and land cover and maize harvested area) as explanatory variables. The explanatory variables were used as inputs into a variable selection experiment to select the least correlated ones that were then used to predict FAW establishment, i.e., suitability areas (very low suitability - very high suitability). The shared socio-economic pathways, SSP2-4.5 and SSP5-8.5 for the years 2030 and 2050 were used to predict the effect of future climate scenarios on FAW establishment. The results demonstrated that FAW establishment areas in eastern Africa were based on the model strength and true performance (area under the curve: AUC = 0.87), but not randomly. Moreover, ∼27% of eastern Africa is currently at risk of FAW establishment. Predicted FAW risk areas are expected to increase to ∼29% (using each of the SSP2-4.5 and SSP5-8.5 scenarios) in the year 2030, and to ∼38% (using SSP2-4.5) and ∼35% (using SSP5-8.5) in the year 2050 climate scenarios. The LULC, particularly croplands and maize harvested area, together with temperature and precipitation bioclimatic variables provided the highest permutation importance in determining the occurrence and establishment of the pest in eastern Africa. Specifically, the study revealed that FAW was sensitive to isothermality (Bio3) rather than being sensitive to a single temperature value in the year. FAW preference ranges of temperature, precipitation, elevation, and maize harvested area were observed, implying the establishment of a once exotic pest in critical maize production regions in eastern Africa. It is recommended that future studies should thus embed the present study's modeling results into a dynamic platform that provides near-real-time predictions of FAW spatial occurrence and risk at the farm scale.

Project description:The fall armyworm (FAW), Spodoptera frugiperda, is native to the tropical and subtropical areas of the American continent and is one of the world's most destructive insect pests and invaded Africa and spread to most of Asia in two years. Glycerol is generally used as a cryoprotectant for overwintering insects in cold areas. In many studies, the increase in glycerol as a main rapid cold hardening (RCH) factor and enhancing the supercooling point was revealed at low temperatures. There are two genes, including glycerol-3-phosphate dehydrogenase (GPDH) and glycerol kinase (GK), that were identified as being associated with the glycerol synthesis pathway. In this study, one GPDH and two GK sequences (GK1 and GK2) were extracted from FAW transcriptome analysis. RNA interference (RNAi) specific to GPDH or GK1 and GK2 exhibited a significant down-regulation at the mRNA level as well as a reduction in survival rate when the RNAi-treated of FAW larvae post a RCH treatment. Following a cold period, an increase in glycerol accumulation was detected utilizing high-pressure liquid chromatography and colorimetric analysis of glycerol quantity in RCH treated hemolymph of FAW larvae. This research suggests that GPDH and GK isozymes are linked to the production of a high quantity of glycerol as an RCH factor, and glycerol as main cryoprotectant plays an important role in survival throughout the cold period in this quarantine pest studied.
