Multiple states in ongoing neural activity in the rat visual cortex.

ABSTRACT: The brain continuously produces internal activity in the absence of afferently salient sensory input. Spontaneous neural activity is intrinsically defined by circuit structures and associated with the mode of information processing and behavioral responses. However, the spatiotemporal dynamics of spontaneous activity in the visual cortices of behaving animals remain almost elusive. Using a custom-made electrode array, we recorded 32-site electrocorticograms in the primary and secondary visual cortex of freely behaving rats and determined the propagation patterns of spontaneous neural activity. Nonlinear dimensionality reduction and unsupervised clustering revealed multiple discrete states of the activity patterns. The activity remained stable in one state and suddenly jumped to another state. The diversity and dynamics of the internally switching cortical states would imply flexibility of neural responses to various external inputs.

Separable gain control of ongoing and evoked activity in the visual cortex by serotonergic input.

Project description:Controlling gain of cortical activity is essential to modulate weights between internal ongoing communication and external sensory drive. Here, we show that serotonergic input has separable suppressive effects on the gain of ongoing and evoked visual activity. We combined optogenetic stimulation of the dorsal raphe nucleus (DRN) with wide-field calcium imaging, extracellular recordings, and iontophoresis of serotonin (5-HT) receptor antagonists in the mouse visual cortex. 5-HT1A receptors promote divisive suppression of spontaneous activity, while 5-HT2A receptors act divisively on visual response gain and largely account for normalization of population responses over a range of visual contrasts in awake and anesthetized states. Thus, 5-HT input provides balanced but distinct suppressive effects on ongoing and evoked activity components across neuronal populations. Imbalanced 5-HT1A/2A activation, either through receptor-specific drug intake, genetically predisposed irregular 5-HT receptor density, or change in sensory bombardment may enhance internal broadcasts and reduce sensory drive and vice versa.

Pulsed infrared light alters neural activity in rat somatosensory cortex in vivo.

Project description:Pulsed infrared light has shown promise as an alternative to electrical stimulation in applications where contact free or high spatial precision stimulation is desired. Infrared neural stimulation (INS) is well characterized in the peripheral nervous system; however, to date, research has been limited in the central nervous system. In this study, pulsed infrared light (λ=1.875 μm, pulse width=250 μs, radiant exposure=0.01-0.55 J/cm(2), fiber size=400 μm, repetition rate=50-200 Hz) was used to stimulate the somatosensory cortex of anesthetized rats, and its efficacy was assessed using intrinsic optical imaging and electrophysiology techniques. INS was found to evoke an intrinsic response of similar magnitude to that evoked by tactile stimulation (0.3-0.4% change in intrinsic signal magnitude). A maximum deflection in the intrinsic signal was measured to range from 0.05% to 0.4% in response to INS, and the activated region of cortex measured approximately 2mm in diameter. The intrinsic signal magnitude increased with faster laser repetition rates and increasing radiant exposures. Single unit recordings indicated a statistically significant decrease in neuronal firing that was observed at the onset of INS stimulation (0.5s stimulus) and continued up to 1s after stimulation onset. The pattern of neuronal firing differed from that observed during tactile stimulation, potentially due to a different spatial integration field of the pulsed infrared light compared to tactile stimulation. The results demonstrate that INS can be used safely and effectively to manipulate neuronal firing.

Neural activity in human visual cortex is transformed by learning real world size.

Project description:The way that our brain processes visual information is directly affected by our experience. Repeated exposure to a visual stimulus triggers experience-dependent plasticity in the visual cortex of many species. Humans also have the unique ability to acquire visual knowledge through instruction. We introduced human participants to the real-world size of previously unfamiliar species, and to the functional motion of novel tools, during a functional magnetic resonance imaging scan. Using machine learning, we compared activity patterns evoked by images of the new items, before and after participants learned the animals' real-world size or tools' motion. We found that, after acquiring size information, participants' visual activity patterns for the new animals became more confusable with activity patterns evoked by similar-sized known animals in early visual cortex, but not in ventral temporal cortex, reflecting an influence of new size knowledge on posterior, but not anterior, components of the ventral stream. In contrast, learning the functional motion of new tools did not lead to an equivalent change in recorded activity. Finally, the time-points marked by evidence of new size information in early visual cortex were more likely to show size information and greater activation in the right angular gyrus, a key hub of semantic knowledge and spatial cognition. Overall, these findings suggest that learning an item's real-world size by instruction influences subsequent activity in visual cortex and in a region that is central to semantic and spatial brain systems.

Layer-specific entrainment of ?-band neural activity by the ? rhythm in monkey visual cortex.

Project description:Although the mammalian neocortex has a clear laminar organization, layer-specific neuronal computations remain to be uncovered. Several studies suggest that gamma band activity in primary visual cortex (V1) is produced in granular and superficial layers and is associated with the processing of visual input. Oscillatory alpha band activity in deeper layers has been proposed to modulate neuronal excitability associated with changes in arousal and cognitive factors. To investigate the layer-specific interplay between these two phenomena, we characterized the coupling between alpha and gamma band activity of the local field potential in V1 of the awake macaque. Using multicontact laminar electrodes to measure spontaneous signals simultaneously from all layers of V1, we found a robust coupling between alpha phase in the deeper layers and gamma amplitude in granular and superficial layers. Moreover, the power in the two frequency bands was anticorrelated. Taken together, these findings demonstrate robust interlaminar cross-frequency coupling in the visual cortex, supporting the view that neuronal activity in the alpha frequency range phasically modulates processing in the cortical microcircuit in a top-down manner.

Reduced neural activity but improved coding in rodent higher-order visual cortex during locomotion.

Project description:Running profoundly alters stimulus-response properties in mouse primary visual cortex (V1), but its effect in higher-order visual cortex is under-explored. Here we systematically investigate how visual responses vary with locomotive state across six visual areas and three cortical layers using a massive dataset from the Allen Brain Institute. Although previous work has shown running speed to be positively correlated with neural activity in V1, here we show that the sign of correlations between speed and neural activity varies across extra-striate cortex, and is even negative in anterior extra-striate cortex. Nevertheless, across all visual cortices, neural responses can be decoded more accurately during running than during stationary periods. We show that this effect is not attributable to changes in population activity structure, and propose that it instead arises from an increase in reliability of single-neuron responses during locomotion.

Functional ultrasound imaging of the spreading activity following optogenetic stimulation of the rat visual cortex.

Project description:Optogenetics has revolutionized neurosciences by allowing fine control of neuronal activity. An important aspect for this control is assessing the activation and/or adjusting the stimulation, which requires imaging the entire volume of optogenetically-induced neuronal activity. An ideal technique for this aim is fUS imaging, which allows one to generate brain-wide activation maps with submesoscopic spatial resolution. However, optical stimulation of the brain with blue light might lead to non-specific activations at high irradiances. fUS imaging of optogenetic activations can be obtained at these wavelengths using lower light power (< 2mW) but it limits the depth of directly activatable neurons from the cortical surface. Our main goal was to report that we can detect specific optogenetic activations in V1 even in deep layers following stimulation at the cortical surface. Here, we show the possibility to detect deep optogenetic activations in anesthetized rats expressing the red-shifted opsin ChrimsonR in V1 using fUS imaging. We demonstrate the optogenetic specificity of these activations and their neuronal origin with electrophysiological recordings. Finally, we show that the optogenetic response initiated in V1 spreads to downstream (LGN) and upstream (V2) visual areas.

Locking of correlated neural activity to ongoing oscillations.

Project description:Population-wide oscillations are ubiquitously observed in mesoscopic signals of cortical activity. In these network states a global oscillatory cycle modulates the propensity of neurons to fire. Synchronous activation of neurons has been hypothesized to be a separate channel of signal processing information in the brain. A salient question is therefore if and how oscillations interact with spike synchrony and in how far these channels can be considered separate. Experiments indeed showed that correlated spiking co-modulates with the static firing rate and is also tightly locked to the phase of beta-oscillations. While the dependence of correlations on the mean rate is well understood in feed-forward networks, it remains unclear why and by which mechanisms correlations tightly lock to an oscillatory cycle. We here demonstrate that such correlated activation of pairs of neurons is qualitatively explained by periodically-driven random networks. We identify the mechanisms by which covariances depend on a driving periodic stimulus. Mean-field theory combined with linear response theory yields closed-form expressions for the cyclostationary mean activities and pairwise zero-time-lag covariances of binary recurrent random networks. Two distinct mechanisms cause time-dependent covariances: the modulation of the susceptibility of single neurons (via the external input and network feedback) and the time-varying variances of single unit activities. For some parameters, the effectively inhibitory recurrent feedback leads to resonant covariances even if mean activities show non-resonant behavior. Our analytical results open the question of time-modulated synchronous activity to a quantitative analysis.

Dynamics of the cerebral blood flow response to brief neural activity in human visual cortex.

Project description:The blood oxygen-level dependent (BOLD) functional magnetic resonance imaging (fMRI) signal depends on an interplay of cerebral blood flow (CBF), oxygen metabolism, and cerebral blood volume. Despite wide usage of BOLD fMRI, it is not clear how these physiological components create the BOLD signal. Here, baseline CBF and its dynamics evoked by a brief stimulus (2 s) in human visual cortex were measured at 3T. We found a stereotypical CBF response: immediate increase, rising to a peak a few second after the stimulus, followed by a significant undershoot. The BOLD hemodynamic response function (HRF) was also measured in the same session. Strong correlations between HRF and CBF peak responses indicate that the flow responses evoked by neural activation in nearby gray matter drive the early HRF. Remarkably, peak CBF and HRF were also strongly modulated by baseline perfusion. The CBF undershoot was reliable and significantly correlated with the HRF undershoot. However, late-time dynamics of the HRF and CBF suggest that oxygen metabolism can also contribute to the HRF undershoot. Combined measurement of the CBF and HRF for brief neural activation is a useful tool to understand the temporal dynamics of neurovascular and neurometabolic coupling.

UP states protect ongoing cortical activity from thalamic inputs.

Project description:Cortical neurons in vitro and in vivo fluctuate spontaneously between two stable membrane potentials: a depolarized UP state and a hyperpolarized DOWN state. UP states temporally correspond with multineuronal firing sequences which may be important for information processing. To examine how thalamic inputs interact with ongoing cortical UP state activity, we used calcium imaging and targeted whole-cell recordings of activated neurons in thalamocortical slices of mouse somatosensory cortex. Whereas thalamic stimulation during DOWN states generated multineuronal, synchronized UP states, identical stimulation during UP states had no effect on the subthreshold membrane dynamics of the vast majority of cells or on ongoing multineuronal temporal patterns. Both thalamocortical and corticocortical PSPs were significantly reduced and neuronal input resistance was significantly decreased during cortical UP states -- mechanistically consistent with UP state insensitivity. Our results demonstrate that cortical dynamics during UP states are insensitive to thalamic inputs.

Projections between visual cortex and pulvinar in the rat.

Project description:The extrageniculate visual pathway, which carries visual information from the retina through the superficial layers of the superior colliculus and the pulvinar, is poorly understood. The pulvinar is thought to modulate information flow between cortical areas, and has been implicated in cognitive tasks like directing visually guided actions. In order to better understand the underlying circuitry, we performed retrograde injections of modified rabies virus in the visual cortex and pulvinar of the Long-Evans rat. We found a relatively small population of cells projecting to primary visual cortex (V1), compared to a much larger population projecting to higher visual cortex. Reciprocal corticothalamic projections showed a similar result, implying that pulvinar does not play as big a role in directly modulating rodent V1 activity as previously thought.
