Movements of Indian Flying Fox in Myanmar as a Guide to Human-Bat Interface Sites.

ABSTRACT: Frugivorous bats play a vital role in tropical ecosystems as pollinators and seed dispersers but are also important vectors of zoonotic diseases. Myanmar sits at the intersection of numerous bioregions and contains habitats that are important for many endangered and endemic species. This rapidly developing country also forms a connection between hotspots of emerging human diseases. We deployed Global Positioning System collars to track the movements of 10 Indian flying fox (Pteropus giganteus) in the agricultural landscapes of central Myanmar. We used clustering analysis to identify foraging sites and high-utilization areas. As part of a larger viral surveillance study in bats of Myanmar, we also collected oral and rectal swab samples from 29 bats to test for key emerging viral diseases in this colony. There were no positive results detected for our chosen viruses. We analyzed their foraging movement behavior and evaluated selected foraging sites for their potential as human-wildlife interface sites.

Project description:Flying-foxes (Pteropodidae) are large bats capable of long-distance flight. Many species are threatened; some are considered pests. Effective conservation and management of flying-foxes are constrained by lack of knowledge of their ecology, especially of movement patterns over large spatial scales. Using satellite telemetry, we quantified long-distance movements of the grey-headed flying-fox Pteropus poliocephalus among roost sites in eastern Australia. Fourteen adult males were tracked for 2-40 weeks (mean 25 weeks). Collectively, these individuals utilised 77 roost sites in an area spanning 1,075 km by 128 km. Movement patterns varied greatly between individuals, with some travelling long distances. Five individuals travelled cumulative distances >1,000 km over the study period. Five individuals showed net displacements >300 km during one month, including one movement of 500 km within 48 hours. Seasonal movements were consistent with facultative latitudinal migration in part of the population. Flying-foxes shifted roost sites frequently: 64% of roost visits lasted <5 consecutive days, although some individuals remained at one roost for several months. Modal 2-day distances between consecutive roosts were 21-50 km (mean 45 km, range 3-166 km). Of 13 individuals tracked for >12 weeks, 10 moved >100 km in one or more weeks. Median cumulative displacement distances over 1, 10 and 30 weeks were 0 km, 260 km and 821 km, respectively. On average, over increasing time-periods, one additional roost site was visited for each additional 100 km travelled. These findings explain why culling and relocation attempts have had limited success in resolving human-bat conflicts in Australia. Flying-foxes are highly mobile between camps and regularly travel long distances. Consequently, local control actions are likely to have only temporary effects on local flying-fox populations. Developing alternative methods to manage these conflicts remains an important challenge that should be informed by a better understanding of the species' movement patterns.

Project description:Urban expansion is a major threat to natural ecosystems but also creates novel opportunities that adaptable species can exploit. The grey-headed flying-fox (Pteropus poliocephalus) is a threatened, highly mobile species of bat that is increasingly found in human-dominated landscapes, leading to many management and conservation challenges. Flying-fox urbanisation is thought to be a result of diminishing natural foraging habitat or increasing urban food resources, or both. However, little is known about landscape utilisation of flying-foxes in human-modified areas, and how this may differ in natural areas. Here we examine positional data from 98 satellite-tracked P. poliocephalus for up to 5 years in urban and non-urban environments, in relation to vegetation data and published indices of foraging habitat quality. Our findings indicate that human-modified foraging landscapes sustain a large proportion of the P. poliocephalus population year-round. When individuals roosted in non-urban and minor-urban areas, they relied primarily on wet and dry sclerophyll forest, forested wetlands, and rainforest for foraging, and preferentially visited foraging habitat designated as high-quality. However, our results highlight the importance of human-modified foraging habitats throughout the species' range, and particularly for individuals that roosted in major-urban environments. The exact plant species that exist in human-modified habitats are largely undocumented; however, where this information was available, foraging by P. poliocephalus was associated with different dominant plant species depending on whether individuals roosted in 'urban' or 'non-urban' areas. Overall, our results demonstrate clear differences in urban- and non-urban landscape utilisation by foraging P. poliocephalus. However, further research is needed to understand the exact foraging resources used, particularly in human-modified habitats, and hence what attracts flying-foxes to urban areas. Such information could be used to modify the urban foraging landscape, to assist long-term habitat management programs aimed at minimising human-wildlife conflict and maximising resource availability within and outside of urban environments.

Project description:Indian fruit bats, flying fox Pteropus medius was identified as an asymptomatic natural host of recently emerged Nipah virus, which is known to induce a severe infectious disease in humans. The absence of P. medius genome sequence presents an important obstacle for further studies of virus-host interactions and better understanding of mechanisms of zoonotic viral emergence. Generation of the high-quality genome sequence is often linked to a considerable effort associated to elevated costs. Although secondary scaffolding methods have reduced sequencing expenses, they imply the development of new tools for the integration of different data sources to achieve more reliable sequencing results. We initially sequenced the P. medius genome using the combination of Illumina paired-end and Nanopore sequencing, with a depth of 57.4x and 6.1x, respectively. Then, we introduced the novel scaff2link software to integrate multiple sources of information for secondary scaffolding, allowing to remove the association with discordant information among two sources. Different quality metrics were next produced to validate the benefits from secondary scaffolding. The P. medius genome, assembled by this method, has a length of 1,985 Mb and consists of 33,613 contigs and 16,113 scaffolds with an NG50 of 19 Mb. At least 22.5% of the assembled sequences is covered by interspersed repeats already described in other species and 19,823 coding genes are annotated. Phylogenetic analysis demonstrated the clustering of P. medius genome with two other Pteropus bat species, P. alecto and P. vampyrus, for which genome sequences are currently available. SARS-CoV entry receptor ACE2 sequence of P. medius was 82.7% identical with ACE2 of Rhinolophus sinicus bats, thought to be the natural host of SARS-CoV. Altogether, our results confirm that a lower depth of sequencing is enough to obtain a valuable genome sequence, using secondary scaffolding approaches and demonstrate the benefits of the scaff2link application. The genome sequence is now available to the scientific community to (i) proceed with further genomic analysis of P. medius, (ii) to characterize the underlying mechanism allowing Nipah virus maintenance and perpetuation in its bat host, and (iii) to monitor their evolutionary pathways toward a better understanding of bats' ability to control viral infections.

Project description:Bats of the genus Pteropus (flying-foxes) are the natural host of Hendra virus (HeV) which periodically causes fatal disease in horses and humans in Australia. The increased urban presence of flying-foxes often provokes negative community sentiments because of reduced social amenity and concerns of HeV exposure risk, and has resulted in calls for the dispersal of urban flying-fox roosts. However, it has been hypothesised that disturbance of urban roosts may result in a stress-mediated increase in HeV infection in flying-foxes, and an increased spillover risk. We sought to examine the impact of roost modification and dispersal on HeV infection dynamics and cortisol concentration dynamics in flying-foxes. The data were analysed in generalised linear mixed models using restricted maximum likelihood (REML). The difference in mean HeV prevalence in samples collected before (4.9%), during (4.7%) and after (3.4%) roost disturbance was small and non-significant (P = 0.440). Similarly, the difference in mean urine specific gravity-corrected urinary cortisol concentrations was small and non-significant (before = 22.71 ng/mL, during = 27.17, after = 18.39) (P= 0.550). We did find an underlying association between cortisol concentration and season, and cortisol concentration and region, suggesting that other (plausibly biological or environmental) variables play a role in cortisol concentration dynamics. The effect of roost disturbance on cortisol concentration approached statistical significance for region, suggesting that the relationship is not fixed, and plausibly reflecting the nature and timing of disturbance. We also found a small positive statistical association between HeV excretion status and urinary cortisol concentration. Finally, we found that the level of flying-fox distress associated with roost disturbance reflected the nature and timing of the activity, highlighting the need for a 'best practice' approach to dispersal or roost modification activities. The findings usefully inform public discussion and policy development in relation to Hendra virus and flying-fox management.
