Effect of the Rearing Substrate on Total Protein and Amino Acid Composition in Black Soldier Fly.

ABSTRACT: Insects are becoming increasingly relevant as protein sources in food and feed. The Black Soldier Fly (BSF) is one of the most utilized, thanks to its ability to live on many leftovers. Vegetable processing industries produce huge amounts of by-products, and it is important to efficiently rear BSF on different substrates to assure an economical advantage in bioconversion and to overcome the seasonality of some leftovers. This work evaluated how different substrates affect the protein and amino acid content of BSF. BSF prepupae reared on different substrates showed total protein content varying between 35% and 49% on dry matter. Significant lower protein contents were detected in BSF grown on fruit by-products, while higher contents were observed when autumnal leftovers were employed. BSF protein content was mainly correlated to fibre and protein content in the diet. Among amino acids, lysine, valine and leucine were most affected by the diet. Essential amino acids satisfied the Food and Agricultural Organization (FAO) requirements for human nutrition, except for lysine in few cases. BSF could be a flexible tool to bio-convert a wide range of vegetable by-products of different seasonality in a high-quality protein-rich biomass, even if significant differences in the protein fraction were observed according to the rearing substrate.

Project description:Saprophagous fly larvae interact with a rich community of bacteria in decomposing organic matter. Larvae of some species, such as the black soldier fly, can process a wide range of organic residual streams into edible insect biomass and thus produce protein as a sustainable component of livestock feed. The microbiological safety of the insects and substrates remains a point of concern. Substrate-associated bacteria can dominate the larval gut microbiota, but the larvae can also alter the bacterial community in the substrate. However, the relative importance of substrate type and larval density in bacterial community dynamics is unknown. We investigated four larval densities (0 [control], 50, 100, or 200 larvae per container [520 mL; diameter, 75 mm]) and three feed substrates (chicken feed, chicken manure, and camelina substrate [50% chicken feed, 50% camelina oilseed press cake]) and sampled the bacterial communities of the substrates and larvae at three time points over 15 days. Although feed substrate was the strongest driver of microbiota composition over time, larval density significantly altered the relative abundances of several common bacterial genera, including potential pathogens, in each substrate and in larvae fed chicken feed. Bacterial communities of the larvae and substrate differed to a higher degree in chicken manure and camelina than in chicken feed. This supports the substrate-dependent impact of black soldier fly larvae on bacteria both within the larvae and in the substrate. This study indicates that substrate composition and larval density can alter bacterial community composition and might be used to improve insect microbiological safety. IMPORTANCE Black soldier fly larvae can process organic side streams into nutritious insect biomass, yielding a sustainable ingredient of animal feed. In processing such organic residues, the larvae impact the substrate and its microbiota. However, their role relative to the feed substrate in shaping the bacterial community is unknown. This may be important for the waste management industry to determine whether pathogens can be controlled by manipulating the larval density and the timing of harvest. We investigated how the type of feed substrate and the larval density (number of larvae per container) interacted to influence bacterial community composition in the substrates and larvae over time. Substrate type was the strongest driver of bacterial community composition, and the magnitude of the impact of the larvae depended on the substrate type and larval density. Thus, both substrate composition and larval density may be used to improve the microbiological safety of the larvae as animal feed.

Project description:Significant economic, environmental, and social impacts are associated with the avoidable disposal of foods worldwide. Mass-rearing of black soldier fly (Hermetia illucens) larvae using organic wastes and food- and agro-industry side products is promising for recycling resources within the food system. One current challenge of this approach is ensuring a reliable and high conversion performance of larvae with inherently variable substrates. Research has been devoted to increasing rearing performance by optimizing substrate nutrient contents and ratios, while the potential of the substrate and larval gut microbiota to increase rearing performance remains untapped. Since previous research has focused on gut microbiota, here, we describe bacterial dynamics in the residue (i.e., the mixture of frass and substrate) of black soldier fly larvae reared on two food wastes (i.e., canteen and household waste). To identify members of the substrate and residue microbiota, potentially associated with rearing performance, bacterial dynamics were also studied in the canteen waste without larvae, and after inactivation by irradiation of the initial microbiota in canteen waste. The food waste substrates had similar microbiota; both were dominated by common lactic acid bacteria. Inactivation of the canteen waste microbiota, which was dominated by Leuconostoc, Bacillus, and Staphylococcus, decreased the levels of all rearing performance indicators by 31-46% relative to canteen waste with the native microbiota. In both food waste substrates, larval rearing decreased the bacterial richness and changed the physicochemical residue properties and composition over the rearing period of 12 days, and typical members of the larval intestinal microbiota (i.e., Providencia, Dysgonomonas, Morganella, and Proteus) became more abundant, suggesting their transfer into the residue through excretions. Future studies should isolate members of these taxa and elucidate their true potential to influence black soldier fly mass-rearing performance.

Project description:BackgroundControlling the substrate moisture is a significant challenge in black soldier fly (BSF) farming. Many substrates have a high moisture content, which results in a low BSF biomass and a high mortality. One potential solution involves incorporating dry substrates into the food mix to mitigate the excessive moisture. However, little information about the types and quantities of dry substrates is available.MethodsSix different dry materials-rice husk (RH), rice bran (RB), rice husk ash (RHA), coconut coir dust (CC), rubberwood sawdust (RSD), and spent coffee grounds (SCGs)-were evaluated by combining with pure minced mixed vegetables in varying proportions (0%, 5%, 10%, 15%, 25%, and 50% by weight). This study encompassed both small-scale and medium-scale experiments to comprehensively assess the effects of the addition of each of these different dry substrates and their quantities on aspects of the development of BSF, such as BSF biomass, larval duration, mortality rates, adult sex ratio, and the moisture removal efficiency of each substrate mixture.ResultsEach dry substrate had specific properties. Although RB emerged as a favorable dry substrate owing to its nutritional content and substantial water-holding capacity, excessive use of RB (>15% by weight) resulted in elevated temperatures and subsequent desiccation of the substrate, potentially leading to larval mortality. In contrast, RH demonstrated the ability to support improved larval duration and growth, permitting its utilization in higher proportions (up to 50%). On the other hand, CC, RHA, and SCG are better suited for inclusion in BSF larval substrates in smaller quantities.DiscussionSome dry substrates require a pretreatment process to eliminate toxic substances prior to their incorporation into substrate mixtures, such as CC and SCG. A potential alternative solution involves employing a combination of various dry substrates. This approach aims to enhance the substrate moisture control and subsequently improve the BSF rearing performance.

Project description:The growing demands on protein producers and the dwindling available resources have made Hermetia illucens (the black soldier fly, BSF) an economically important species. Insights into the genome of this insect will better allow for robust breeding protocols, and more efficient production to be used as a replacement of animal feed protein. The use of microRNA as a method to understand how gene regulation allows insect species to adapt to changes in their environment, has been established in multiple species. The baseline and life stage expression levels established in this study, allow for insight into the development and sex-linked microRNA regulation in BSF. To accomplish this, microRNA was extracted and sequenced from 15 different libraries with each life stage in triplicate. Of the total 192 microRNAs found, 168 were orthologous to known arthropod microRNAs and 24 microRNAs were unique to BSF. Twenty-six of the 168 microRNAs conserved across arthropods had a statistically significant (p < 0.05) differential expression between Egg to Larval stages. The development from larva to pupa was characterized by 16 statistically significant differentially expressed microRNA. Seven and 9 microRNA were detected as statistically significant between pupa to adult female and pupa to adult male, respectively. All life stages had a nearly equal split between up and down regulated microRNAs. Ten of the unique 24 miRNA were detected exclusively in one life stage. The egg life stage expressed five microRNA (hil-miR-m, hil-miR-p, hil-miR-r, hil-miR-s, and hil-miR-u) not seen in any other life stages. The female adult and pupa life stages expressed one miRNA each hil-miR-h and hil-miR-ac respectively. Both male and female adult life stages expressed hil-miR-a, hil-miR-b, and hil-miR-y. There were no unique microRNAs found only in the larva stage. Twenty-two microRNAs with 56 experimentally validated target genes in the closely related Drosophila melanogaster were identified. Thus, the microRNA found display the unique evolution of BSF, along with the life stages and potential genes to target for robust mass rearing. Understanding of the microRNA expression in BSF will further their use in the crucial search for alternative and sustainable protein sources.

Project description:Black soldier fly (Hermetia illucens) larvae can convert biowaste and by-products into body mass high in protein (~40% dry matter, DM) and lipid (~30% DM). However, the type of rearing substrate also affects the larval body composition and thus its nutritional value. Hitherto, it remains unclear how and to what extent the larval body composition can be altered by the substrate. This study was therefore performed to examine the possibilities of modifying larval body composition using different rearing substrates. To investigate this, 5-days old larvae were reared for seven days on different locally available waste and by-products: brewer's spent grain, mitigation mussels (Mytilus edulis), rapeseed cake, and shrimp waste meal (Pandalus borealis). Larval composition and performance were compared to larvae reared on a commercial chicken feed as well as a mixed feed (mixture of chicken feed and by-products, with a similar macronutrient composition to chicken feed). Larval body weight was recorded daily to determine growth over time whereas larvae and substrates were sampled at the start and end of the trial and analysed for their nutritional composition. The type of rearing substrate affected both larval body composition and growth performance. There was a clear relation between the nutritional composition of the substrate and larvae for certain fatty acids. Larvae reared on marine-based waste substrates contained a higher share of omega-3 fatty acids than larvae reared on the other substrates, indicating an accumulation of omega-3 fatty acids from the substrate. There was a strong positive linear correlation between the ash content in the substrate and larvae whereas larval lipid, protein, amino acid, and chitin content seemed more affected by larval development. Overall, this study showed that the rearing substrate affects larval composition and development, and that larval composition of certain nutrients can be tailored depending on further food and feed applications.
