Stand-Alone CuFeSe2 (Eskebornite) Nanosheets for Photothermal Cancer Therapy.

ABSTRACT: Two-dimensional CuFeSe2 nanosheets have been successfully obtained via solution-phase synthesis using a sacrificial template method. The high purity was confirmed by X-ray diffraction and the two-dimensional morphology was validated by transmission electron microscopy. The intense absorption in the 400-1400 nm region has been the basis for the CuFeSe2 nanosheets' photothermal capabilities testing. The colloidal CuFeSe2 (CFS) nanosheets capped with S2- short ligands (CFS-S) exhibit excellent biocompatibility in cell culture studies and strong photothermal effects upon 808 nm laser irradiation. The nanosheets were further loaded with the cancer drug doxorubicin and exposed to laser irradiation, which accelerated the release of doxorubicin, achieving synergy in the therapeutic effect.

Functional black phosphorus nanosheets for mitochondria-targeting photothermal/photodynamic synergistic cancer therapy.

Project description:Organelle-targeting nanosystems are envisioned as promising tools for phototherapy, which can generate heat or reactive oxygen species (ROS) induced cytotoxicity in the targeted location but leave the surrounding biological tissues undamaged. In this work, an imaging-guided mitochondria-targeting photothermal/photodynamic nanosystem has been developed on the basis of functionalized black phosphorus (BP) nanosheets (NSs). In the nanosystem, BP NSs are coated with polydopamine (PDA) and then covalently linked with both chlorin e6 (Ce6) and triphenyl phosphonium (TPP) through carbodiimide reaction between the amino group and the carboxyl group, forming BP@PDA-Ce6&TPP NSs. Due to the strong absorbance of BP@PDA in the near-infrared region and the highly efficient ROS generation of Ce6, the as-prepared nanosystem with mitochondria-targeting capacity (TPP moiety) shows remarkably enhanced efficiency in cancer cell killing. Combined photothermal and photodynamic therapy is implemented and monitored by in vivo fluorescence imaging, achieving excellent performance in inhibiting tumor growth. This study presents a novel nanotheranostic agent for mitochondria-targeting phototherapy, which may open new horizons for biomedicine.

Biomimetic MDSCs membrane coated black phosphorus nanosheets system for photothermal therapy/photodynamic therapy synergized chemotherapy of cancer.

Project description:Photothermal therapy is favored by cancer researchers due to its advantages such as controllable initiation, direct killing and immune promotion. However, the low enrichment efficiency of photosensitizer in tumor site and the limited effect of single use limits the further development of photothermal therapy. Herein, a photo-responsive multifunctional nanosystem was designed for cancer therapy, in which myeloid-derived suppressor cell (MDSC) membrane vesicle encapsulated decitabine-loaded black phosphorous (BP) nanosheets (BP@ Decitabine @MDSCs, named BDM). The BDM demonstrated excellent biosafety and biochemical characteristics, providing a suitable microenvironment for cancer cell killing. First, the BDM achieves the ability to be highly enriched at tumor sites by inheriting the ability of MDSCs to actively target tumor microenvironment. And then, BP nanosheets achieves hyperthermia and induces mitochondrial damage by its photothermal and photodynamic properties, which enhancing anti-tumor immunity mediated by immunogenic cell death (ICD). Meanwhile, intra-tumoral release of decitabine induced G2/M cell cycle arrest, further promoting tumor cell apoptosis. In vivo, the BMD showed significant inhibition of tumor growth with down-regulation of PCNA expression and increased expression of high mobility group B1 (HMGB1), calreticulin (CRT) and caspase 3. Flow cytometry revealed significantly decreased infiltration of MDSCs and M2-macrophages along with an increased proportion of CD4+, CD8+ T cells as well as CD103+ DCs, suggesting a potentiated anti-tumor immune response. In summary, BDM realizes photothermal therapy/photodynamic therapy synergized chemotherapy for cancer.

Titanium carbide nanosheets with defect structure for photothermal-enhanced sonodynamic therapy.

Project description:Sonodynamic therapy (SDT) has attracted widespread interest in biomedicine, owing to its novel and noninvasive therapeutic method triggered by ultrasound (US). Herein, the Ti3C2 MXene nanosheets (Ti3C2 NSs) are developed as good sonosensitizers via a two-step method of chemical exfoliation and high-temperature treatment. With the high-temperature treatment, the oxygen defect of Ti3C2 MXene nanosheets (H-Ti3C2 NSs) is greatly increased. Therefore, the electron (e-) and hole (h+) generated by US can be separated faster due to the improved degree of oxidation, and then the recombination of e--h+ can be prevented with the abundant oxygen defect under US irradiation, which induced the sonodynamic efficiency greatly to improve around 3.7-fold compared with Ti3C2 NSs without high-temperature treatment. After PEGylation, the H-Ti3C2-PEG NSs show good stability and biocompatibility. In vitro studies exhibit that the inherent property of mild photothermal effect can promote the endocytosis of H-Ti3C2-PEG NSs, which can improve the SDT efficacy. In vivo studies further display that the increased blood supply by the mild photothermal effect can significantly relieve hypoxia in the tumor microenvironment, showing photothermal therapy (PTT) enhanced SDT. Most importantly, the H-Ti3C2-PEG NSs can be biodegraded and excreted out of the body, showing no significant long-term toxicity. Our work develops the defective H-Ti3C2 NSs as high-efficiency and safe sonosensitizers for photothermal-enhanced SDT of cancer, extending the biomedical application of MXene-based nanoplatforms.

Selenide Heterostructure Nanosheets with Efficient Near-Infrared Photothermal Conversion for Therapy.

Project description:Photothermal therapy has been regarded as one of promising ways for tumor treatment. However, nanoagents with highly efficient thermal conversion and good bio-compatibility are still needed to be developed in biomedicine. In this work, we prepared two-dimensional heterostructures with bismuth selenide and tungsten selenide nanosheets as photothermal nanoagents. Near-infrared photothermal conversion of selenide heterostructure nanosheets can reach up to 40.75% under 808 nm excitation. It is known that selenium is a critical element to human health. More importantly, our experiments with mice show that the heterostructure nanosheets have low toxicity and high biocompatibility both in vitro and in vivo. The nanoagents based on heterostructures can effectively realize photothermal tumor ablation. It is suggested that the developed selenide nanosheets have great potential application in cancer therapy.

Dynamic Resistance Training as Stand-Alone Antihypertensive Lifestyle Therapy: A Meta-Analysis.

Project description:Aerobic exercise (AE) is recommended as first-line antihypertensive lifestyle therapy based on strong evidence showing that it lowers blood pressure (BP) 5 to 7 mm Hg among adults with hypertension. Because of weaker evidence showing that dynamic resistance training (RT) reduces BP 2 to 3 mm Hg among adults with hypertension, it is recommended as adjuvant lifestyle therapy to AE training. Yet, existing evidence suggests that dynamic RT can lower BP as much or more than AE. We meta-analyzed 64 controlled studies (71 interventions) to determine the efficacy of dynamic RT as stand-alone antihypertensive therapy. Participants (N=2344) were white (57%), middle-aged (47.2±19.0 years), and overweight (26.8±3.4 kg/m(2)) adults with prehypertension (126.7±10.3/76.8±8.7 mm Hg); 15% were on antihypertensive medication. Overall, moderate-intensity dynamic RT was performed 2.8±0.6 days/week for 14.4±7.9 weeks and elicited small-to-moderate reductions in systolic BP (SBP; d+=-0.31; 95% CIs, -0.43, -0.19; -3.0 mm Hg) and diastolic BP (DBP; d+=-0.30; 95% CIs, -0.38, -0.18; -2.1 mm Hg) compared to controls (Ps<0.001). Greater BP reductions occurred among samples with higher resting SBP/DBP: ≈6/5 mm Hg for hypertension, ≈3/3 mm Hg for prehypertension, and ≈0/1 mm Hg for normal BP (Ps<0.023). Furthermore, nonwhite samples with hypertension experienced BP reductions that were approximately twice the magnitude of those previously reported following AE training (-14.3 mm Hg [95% CIs, -19.0, -9.4]/-10.3 mm Hg [95% CIs, -14.5, -6.2]). Our results indicate that for nonwhite adult samples with hypertension, dynamic RT may elicit BP reductions that are comparable to or greater than those reportedly achieved with AE training. Dynamic RT should be further investigated as a viable stand-alone therapeutic exercise option for adult populations with high BP.

Multicomponent Transition Metal Dichalcogenide Nanosheets for Imaging-Guided Photothermal and Chemodynamic Therapy.

Project description:Transition metal dichalcogenides (TMDs) have received considerable attention due to their strong absorption in the near-infrared (NIR) region, strong spin-orbit coupling, and excellent photothermal conversion efficiency (PCE). Herein, CoFeMn dichalcogenide nanosheets (CFMS NSs) are prepared via facile vulcanization of a lamellar CoFeMn-layered double hydroxide (LDH) precursor followed by polyvinyl pyrrolidone modification (to give CFMS-PVP NSs), and found to show excellent photoacoustic (PA) imaging and synergistic photothermal/chemodynamic therapy (PTT/CDT) performance. The as-prepared CFMS-PVP NSs inherit the ultrathin morphology of the CoFeMn-LDH precursor and exhibit an outstanding photothermal performance with a η of 89.0%, the highest PCE reported to date for 2D TMD materials. Moreover, 50% of maximum catalytic activity (Michaelis-Menten constant, K m) is attained by CFMS-PVP NSs with 0.26 × 10-3 m H2O2 at 318 K, markedly lower than the endogenous concentration of H2O2 inside tumor cells. In addition, complete apoptosis of HepG2 cancer cells and complete tumor elimination in vivo are observed after treatment with CFMS-PVP NSs at a low dose, substantiating the NSs' remarkable PTT/CDT efficacy. This work provides a new and facile approach for the synthesis of high-quality multicomponent TMD nanosheets with precise process control, the potential for mass production, and outstanding performance, providing great promise in cancer theranostics.

In situ Growth of ZIF-8 Nanoparticles on Graphene Oxide Nanosheets: A Multifunctional Nanoplatform for Combined Ion‐interference and Photothermal Therapy

Project description:In summary, highly stability and dispersed BSArGO@ZIF8 NSs were prepared by a simple electrostatic interaction method. According to the delivery effect of BSArGO@ZIF8 NSs, we found that BSArGO@ZIF8 NSs possess higher lethality to cancer cells. Furthermore, under NIR irradiation, BSArGO@ZIF8 NSs-mediated PTT could further kill cancer cells, and the increased temperature caused by photothermal conversion accelerated the Fenton reaction rate, enhancing the efficiency of BSArGO@ZIF8 NSs cell lethality. RNA-seq analysis confirmed that. BSArGO@ZIF8 NSs could promote cancer death through activate bim-mediated mitochondrial apoptotic events, induce disruption of microtubule function, loss of integrity of the nuclear membrane, DNA flagmentation, and change pro-apoptptic genes expression.

Fabrication of Polydopamine-Coated High-Entropy MXene Nanosheets for Targeted Photothermal Anticancer Therapy.

Project description:Transition metal carbides, nitrides, and carbonitrides (MXenes) have emerged as a promising class of 2D materials that can be used for various applications. Recently, a new form of high-entropy MXenes has been reported, which contains an increased number of elemental species that can increase the configurational entropy and reduce the Gibbs free energy. The unique structure and composition lead to a range of intriguing and tunable characteristics. Herein, the fabrication of high-entropy MXene TiVNbMoC3Tx (T = surface terminations) with a layer of polydopamine is reported, followed by immobilization of a phthalocyanine-based fluorophore for imaging and the peptide sequence QRHKPREGGGSC for targeting the epidermal growth factor receptor (EGFR) overexpressed in cancer cells. The resulting nanocomposite exhibits high biocompatibility and superior photothermal property. Upon laser irradiation at 808 nm, the light-to-heat conversion efficiency is up to 56.1%, which is significantly higher than that of conventional 2D materials. In vitro studies show that these nanosheets could be internalized selectively into EGFR-positive cancer cells and effectively eliminate these cells mainly through photothermal-induced apoptosis. Using 4T1 tumor-bearing mice as an animal model, the nanosheets could accumulate at the tumor and effectively eradicate the tumor upon laser irradiation without causing noticeable adverse effects to the mice.

Hybrid Polypyrrole and Polydopamine Nanosheets for Precise Raman/Photoacoustic Imaging and Photothermal Therapy.

Project description:The development of near-infrared light responsive conductive polymers provides a useful theranostic platform for malignant tumors by maximizing spatial resolution with deep tissue penetration for diagnosis and photothermal therapy. Herein, the self-assembly of ultrathin 2D polypyrrole nanosheets utilizing dopamine as a capping agent and a monolayer of octadecylamine as a template is demonstrated. The 2D polypyrrole-polydopamine nanostructure has tunable size distribution which shows strong absorption in the first and second near-infrared windows, enabling photoacoustic imaging and photothermal therapy. The hybrid double-layer is demonstrated to increase Raman intensity for 3D Raman imaging (up to two orders of magnitude enhancement and spatial resolution up to 1 µm). The acidic environment drives reversible doping of polypyrrole, which can be detected by Raman spectroscopy. The combined properties of the nanosheets can substantially enhance performance in dual-mode Raman and photoacoustic guided photothermal therapy, as shown by the 69% light to heat conversion efficiency and higher cytotoxicity against cancer spheroids. These pH-responsive features highlight the potential of 2D conductive polymers for applications in accurate, highly efficient theranostics.
