The Effects of pH and Excipients on Exenatide Stability in Solution.

ABSTRACT: Exenatide, a glucagon-like peptide-1 receptor agonist, is the active pharmaceutical ingredient in Byetta® and Bydureon®, two type 2 diabetes drug products that have generics and multiple follow-up formulations currently in development. Even though exenatide is known to be chemically and physically unstable at pH 7.5, there lacks a systematic evaluation of the impact of pH and excipients on the peptide solution stability. In this study, we established analytical methods to measure the chemical and physical degradation of the peptide in solution. Exenatide remained relatively stable at pH 4.5 when incubated at 37 °C. At pH 5.5-6.5, degradation was driven by oxidation, while driven by deamidation at pH 7.5-8.5. Significant aggregation of exenatide at pH 7.5 and 8.5 was detected by size exclusion chromatography and dynamic light scattering. Each pH value greater than 4.5 exhibited unique profiles corresponding to a loss of α-helical content and an increase in unordered structures. The addition of sugars, including mannitol, sorbitol and sucrose, conferred small protective effects against peptide aggregation when incubating at pH 7.5 and 37 °C, as measured by size-exclusion chromatography and dynamic light scattering. The results of this study will be useful for investigators developing generic exenatide products, peptide analogs and novel exenatide drug delivery systems.

Project description:Over the past decade polyelectrolyte multilayer (PEM)-based membranes have gained a lot of interest in the field of nanofiltration (NF) as an alternative to conventional polyamide-based thin film composite membranes. With great variety in fabrication conditions, these membranes can achieve superior properties such as high chemical resistance and excellent filtration performance. Some of the most common polyelectrolytes used to prepare NF membranes are weak, meaning that their charge density depends on pH within the normal window of operation relevant for potential applications (pH 0-14). This might cause a dependency of membrane properties on the pH of filtered solutions, as indicated by other applications of PEMs. In this work, the susceptibility of membrane structure (swelling and surface charge) and performance (permeability, molecular weight cutoff, and salt retention) toward the pH of the filtration solution was studied for four fundamentally different PEM systems: poly(diallyldimethylammonium chloride) (PDADMAC)/poly(sodium-4-styrenesulfonate) (PSS) (strong/strong), poly(allylamine hydrochloric acid) (PAH)/poly(acrylic acid) (PAA) (weak/weak), and PAH/PSS (weak/strong) and PAH/PSS+PAH/PAA (asymmetric). Slight variations in structure and performance of the PDADMAC/PSS-based membranes were observed. On the contrary, structure and performance of PAH/PAA-based membranes are very susceptible to feed solution pH. A continuous change in charge density with variation in pH significantly affects salt retention. An increased swelling at pH 9 translates to variation in permeability and molecular weight cutoff of the membrane. The susceptibility of PAH/PSS-based membranes to pH is less pronounced compared to the PAH/PAA-based membranes since only one of the polyelectrolytes involved is weak. No structural changes were observed, indicating additional specific interactions between the polyelectrolytes other than electrostatic forces that stabilize film structure. A combination of the PAH/PSS and PAH/PAA system (8 + 2 bilayers) also displays a clear dependency of both membrane structure and performance on solution pH, where PAH/PSS is dominating due to a higher bilayer number.

Project description:Human lysosomal enzymes acid-beta-glucosidase (GCase) and acid-alpha-galactosidase (alpha-Gal A) hydrolyze the sphingolipids glucosyl- and globotriaosylceramide, respectively, and mutations in these enzymes lead to the lipid metabolism disorders Gaucher and Fabry disease, respectively. We have investigated the structure and stability of GCase and alpha-Gal A in a neutral-pH environment reflective of the endoplasmic reticulum and an acidic-pH environment reflective of the lysosome. These details are important for the development of pharmacological chaperone therapy for Gaucher and Fabry disease, in which small molecules bind mutant enzymes in the ER to enable the mutant enzyme to meet quality control requirements for lysosomal trafficking. We report crystal structures of apo GCase at pH 4.5, at pH 5.5, and in complex with the pharmacological chaperone isofagomine (IFG) at pH 7.5. We also present thermostability analysis of GCase at pH 7.4 and 5.2 using differential scanning calorimetry. We compare our results with analogous experiments using alpha-Gal A and the chaperone 1-deoxygalactonijirimycin (DGJ), including the first structure of alpha-Gal A with DGJ. Both GCase and alpha-Gal A are more stable at lysosomal pH with and without their respective iminosugars bound, and notably, the stability of the GCase-IFG complex is pH sensitive. We show that the conformations of the active site loops in GCase are sensitive to ligand binding but not pH, whereas analogous galactose- or DGJ-dependent conformational changes in alpha-Gal A are not seen. Thermodynamic parameters obtained from alpha-Gal A unfolding indicate two-state, van't Hoff unfolding in the absence of the iminosugar at neutral and lysosomal pH, and non-two-state unfolding in the presence of DGJ. Taken together, these results provide insight into how GCase and alpha-Gal A are thermodynamically stabilized by iminosugars and suggest strategies for the development of new pharmacological chaperones for lysosomal storage disorders.

Project description:Thiamine (vitamin B1) is an essential micronutrient in the human diet, found both naturally and as a fortification ingredient in many foods and supplements. However, it is susceptible to degradation due to heat, light, alkaline pH, and sulfites, among effects from other food matrix components, and its degradation has both nutritional and sensory implications as in foods. Thiamine storage stability in solution was monitored over time to determine the effect of solution pH and thiamine concentration on reaction kinetics of degradation without the use of buffers, which are known to affect thiamine stability independent of pH. The study directly compared thiamine stability in solutions prepared with different pHs (3 or 6), concentrations (1 or 20 mg/mL), and counterion in solution (NO3-, Cl-, or both), including both commercially available salt forms of thiamine (thiamine mononitrate and thiamine chloride hydrochloride). Solutions were stored at 25, 40, 60, and 80 °C for up to one year, and degradation was quantified by high-performance liquid chromatography (HPLC) over time, which was then used to calculate degradation kinetics. Thiamine was significantly more stable in pH 3 than in pH 6 solutions. In pH 6 solutions, stability was dependent on initial thiamine concentration, with the 20 mg/mL thiamine salt solutions having an increased reaction rate constant (kobs) compared to the 1 mg/mL solutions. In pH 3 solutions, kobs was not dependent on initial concentration, attributed to differences in degradation pathway dependent on pH. Activation energies of degradation (Ea) were higher in pH 3 solutions (21-27 kcal/mol) than in pH 6 solutions (18-21 kcal/mol), indicating a difference in stability and degradation pathway due to pH. The fundamental reaction kinetics of thiamine reported in this study provide a basis for understanding thiamine stability and therefore improving thiamine delivery in many foods containing both natural and fortified thiamine.
