Influence of Climate on Soil and Wine Bacterial Diversity on a Vineyard in a Non-traditional Wine Region in Argentina.

ABSTRACT: Argentina is the fifth world-wide wine producer, with an area of emerging importance in the Southwest of Buenos Aires Province, where climatic conditions are rather challenging. We studied the variations in soil and wine bacterial diversity through three consecutive vintages, and how climatic conditions affected said diversity. During the years of our study there were two harsh climatic events, a prolonged drought that extended over two vegetative periods, and an unseasonable spring frost in 2017. We found that the bacterial diversity reacted to these climatic events, given that there was a shift in the taxa exclusive to soil and wine, and shared by both, through time. Our results show a core of microorganisms in soil as well as in wine, belonging to different phyla that are conserved across the vintage years. A trend to an enrichment in Actinobacteria was detected in soil samples, whereas a high relative abundance of the Acetobacteraceae family and a scarcity of Lactic Acid Bacteria (LAB) were detected in the wine samples. We believe our results contribute to a better understanding of the impact of climatic conditions on the soil and wine microbiota, and can provide vintners with valuable knowledge for improving their wine production.

Project description:Soil microbial communities have an integral association with plants and play an important role in shaping plant nutrition, health, crop productivity and product quality. The influence of bacteria and fungi on wine fermentation is well known. However, little is known about the role of soil microbes, other than microbial pathogens, on grape composition or their role in vintage or site (terroir) impacts on grape composition. In this study, we used an amplicon sequencing approach to investigate the potential relationships between soil microbes and inherent spatial variation in grape metabolite composition - specifically, the concentration of the 'impact aroma compound' rotundone in Shiraz grapes (Vitis vinifera L.) grown in a 6.1 ha vineyard in the Grampians region of Victoria, Australia. Previous work had demonstrated temporal stability in patterns of within-vineyard spatial variation in rotundone concentration, enabling identification of defined 'zones' of inherently 'low' or 'high' concentration of this grape metabolite. 16S rRNA and ITS region-amplicon sequencing analysis of microbial communities in the surface soils collected from these zones indicated marked differences between zones in the genetic diversity and composition of the soil bacterial and fungal microbiome. Soils in the High rotundone zone exhibited higher diversity of bacteria, but lower diversity of fungi, compared to the soils in the Low rotundone zone. In addition, the network analysis of the microbial community in the High rotundone zone soils appeared well structured, especially with respect to the bacterial community, compared to that in the Low rotundone zone soils. The key differences in the microbial community structure between the rotundone zones are obvious for taxa/groups of both bacteria and fungi, particularly for bacteria belonging to Acidobacteria-GP4 and GP7, Rhizobiales, Gaiellaceae, Alphaproteobacteria and the Nectriaceae and Tremellaceae families of fungi. Although mulching in some parts of the vineyard caused changes in bacterial and fungal composition and overall microbial catabolic diversity and activity, its effects did not mask the rotundone zone-based variation. This finding of a systematic rotundone zone-based variation in soil microbiomes suggests an opportunity to bring together understanding of microbial ecology, plant biochemistry, and viticultural management for improved management of grape metabolism, composition and wine flavor.

Project description:Soil microbes regulate the carbon cycle and affect the formation and stabilization of soil aggregates. However, the interactions between the soil microbial community and soil organic carbon (SOC) fractions, organic carbon (OC) content in aggregates, and soil aggregate stability after afforestation are remain poorly understood. In our study, we investigated SOC fractions in bulk soil, aggregate-associated OC content, soil aggregate stability, and soil bacterial community with high-throughput 16S rRNA sequencing at sites representing natural secondary forest (NF) and managed forest (MF), with cropland (CL) as reference in a degraded karst region of Southwest China. Our results showed that afforestation remarkably increased the SOC fraction and OC content in aggregates, the mean weight diameter (MWD), and the mean geometric diameter (GMD). The most dominant bacterial phyla detected were Acidobacteriota, Actinobacteriota, Proteobacteria, and Chloroflexi across all soils. Afforestation remarkably altered the relative abundances of most of the dominant soil bacteria at the phylum, class, and order levels. Interestingly, such changes in the abundance of soil bacteria taxa had significantly effects on SOC fraction, aggregate-associated OC content, MWD, and MGD. The abundance of dominant bacterial taxa such as Methylomirabilota, Latescibacterota, Methylomirabilia, MB-A2-108, norank_Latescibacterota; Dehalococcoidia, Rokubacteriales, Gaiellales, Microtrichales, norank_c__MB-A2-108, norank_c__norank_p__Latescibacterota, Rhizobiales, and S085 not only remarkably increased but also had significant positive effects on SOC fractions and aggregate-associated OC content after afforestation. Moreover, MWD and MGD were positively correlated with the relative abundance of Methylomirabilota, Methylomirabilia, Rokubacteriales, Latescibacterota, and Rhizobiales. Results indicated the importance of certain soil bacteria for regulating SOC storage and soil aggregate stability. We concluded that afforestation on cropland could alter the abundance of soil bacteria, and these changes modulate the stability of soil aggregates and SOC fractions.
